Refined tamoxifen administration in mice by encouraging voluntary consumption of palatable formulations.

<p>Drug administration in preclinical rodent models is essential for research and the development of novel therapies. Compassionate administration methods have been developed, but these are mostly incompatible with water-insoluble drugs such as <span>tamoxifen</span> <span>or</span> do not <span>allow for</span> precise timing or dosing of the drugs. <span>For more than two decades, tamoxifen has been administered by oral gavage or injection to </span>CreER<sup>T2</sup>/loxP gene-modified mouse models<span> </span>to <span>spatiotemporally control gene expression,</span><span> with the numbers of such inducible models </span><span>steadily increasing in recent years. </span>Animal-friendly procedures <span>for</span> accurately administering <span>tamoxifen or </span><span>other water-insoluble </span><span>drugs </span>would, therefore, have an important impact on animal <span>welfare.</span> <span>Based on a previously published micropipette feeding protocol, we developed palatable formulations to encourage voluntary consumption of tamoxifen. We </span><span>evaluated</span><span> </span><span>the acceptance of the new formulations by mice during training and treatment and assessed the efficacy of </span><span>tamoxifen-mediated induction of </span>CreER<sup>T2</sup>/loxP-dependent reporter <span>gene</span><span>s</span><span>.</span> <span>Both sweetened </span><span>milk</span><span> and syrup-based formulations encouraged mice to consume tamoxifen voluntarily, but only sweetened milk formulations were statistically </span><span>non-inferior</span><span> to oral </span><span>gavage or intraperitoneal injections in inducing </span><span>CreER<sup>T2</sup>-mediated</span><span> gene expression. </span><span><span>Serum concentrations of tamoxifen metabolites, quantified using an in-house developed cell assay, confirmed the lower efficacy of syrup- as compared to sweetened milk-based formulations.</span></span> We found dosing with a micropipette to be more accurate than oral gavage or injection, with the added advantage that the method requires little training for the experimenter.&nbsp;<span>The new palatable solutions encourage voluntary consumption of tamoxifen without loss of </span><span>efficacy</span><span> compared to oral gavage or injections and </span><span>thus represent</span><span> </span>a refined administration method.</p>

Pulsed administration for physiological estrogen replacement in mice

<p>In this study, mice were ovariectomized&nbsp;and treated with physiological doses of 17&beta;-estradiol-3-benzoate (E2) dissolved in miglyol or PBS. Subcutaneous injections were performed every 4 days to resemble the estrus cycle in mice. Results show that OVX induces an osteoporotic phenotype, fat accumulation and impairment of the locomotor ability, as expected. Pulsed administration of physiological doses of E2 dissolved in miglyol rescues the phenotypes induced by OVX. However, when E2 is dissolved in PBS the effects are less pronounced, possibly due to rapid wash out of the steroid.</p>

Administrative Units

<p>Intra-state water conflicts have increased substantially over the past few decades particularly in developing regions, and partly attributed to water scarcity. However, empirical studies linking water scarcity and violent conflicts are sparse, while existing quantitative studies have used mostly climate variables (precipitation and temperature) to understand this link. Most studies that have used climate variables concluded that they were not strong predictors of water conflicts. The aim of this study was to identify water scarcity hotspots and to understand the links between water scarcity and violent conflicts across the Sahel and Lake Chad Basin (LCB) over the period 2000-2021. To achieve this, we combine outputs from a global hydrological model and demographic data to develop six water scarcity metrics. The developed metrics show varying levels of water scarcity across the study region. The Falkenmark index across all capital cities (Ouagadougou-Burkina Faso, Ndjamena-Chad, Bamako-Mali, and Niamey-Niger), and Maradi-Niger and Jigawa &amp; Kano states in Nigeria was less than 100 m<sup>3</sup>/capital/year, indicating acute water scarcity in those areas. Findings further indicated that green water scarcity (GWS) and the Falkenmark index were closely linked with water conflicts compared to the other metrics. Our findings suggest that water conflicts cannot be explained by hydroclimatic factors alone without incorporating other socioeconomic variables like demographic information. Results from this study may be used by stakeholders to tackle endemic water scarcity and to predict and mitigate water conflicts in the region.</p>

Data from: Postnatal administration of S-adenosylmethionine restores developmental AHR activation-induced deficits in CD8+ T cell function during influenza A virus infection

<p>Abstract Developmental exposures can influence life-long health; yet, counteracting negative consequences is challenging due to poor understanding of cellular mechanisms. The aryl hydrocarbon receptor (AHR) binds many small molecules, including numerous pollutants. Developmental exposure to the signature environmental AHR ligand 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) significantly dampens adaptive immune responses to influenza A virus (IAV) in adult offspring. CD8+ cytotoxic T lymphocytes (CTL) are crucial for successful infection resolution, which depends on the number generated and the complexity of their functionality. Prior studies showed developmental AHR activation significantly reduced the number of virus-specific CD8+ T cells, but impact on their functions is less clear. Other studies showed developmental exposure was associated with differences in DNA methylation in CD8+ T cells. Yet, empirical evidence that differences in DNA methylation are causally related to altered CD8+ T cell function is lacking. The two objectives were to ascertain whether developmental AHR activation affects CTL function, and whether differences in methylation contribute to reduced CD8+ T cell responses to infection. Developmental AHR triggering significantly reduced CTL polyfunctionality, and modified the transcriptional program of CD8+ T cells. S-adenosylmethionine (SAM), which increases DNA methylation, but not Zebularine, which diminishes DNA methylation, restored polyfunctionality and boosted the number of virus-specific CD8+ T cells. These findings suggest that diminished methylation, initiated by developmental exposure to an AHR-binding chemical, contributes to durable changes in antiviral CD8+ CTL functions later in life. Thus, deleterious consequence of development exposure to environmental chemicals are not permanently fixed, opening the door for interventional strategies to improve health.</p>

Microbiota of piglets with three different feed additives via maternal or neonatal administration route

<p>Feed additives aiming to improve gastrointestinal health are frequently supplied to piglets after weaning, but might be more effective when administered before weaning. In this period, feed additives can either be administered directly to neonates, or indirectly via sows feed. It is yet unknown what the effect of the administration route is on gut functionality and health. Therefore, we compared the effect of different dietary interventions on gut functionality after maternal administration (lactation feed) to the neonatal administration route (oral gavage). These feed interventions included medium chain fatty acids (MCFA), beta-glucans (BG), and galacto-oligosaccharides (GOS). We measured intestinal gene expression and microbiota composition after birth (d1) and after weaning (d31). Our results show that the type of intervention and the administration route influence gut functionality (microbiome and gene expression profiles). MCFA administration led to a more differentially orchestrated response when comparing the neonatal and maternal administration route then the other two additives, indicating the route of administration of the feed interventions is determinative for the outcome.. This implies that for each nutritional intervention in early life of a pig the optimal route of administration needs to be determined.</p>

Calorie restriction and pravastatin administration during pregnancy in obese rhesus macaques modulates maternal and infant metabolism and infant brain and behavioral development

<p>Maternal obesity has been associated with a higher risk of pregnancy-related complications in mothers and offspring; however, effective interventions have not yet been developed. We tested two common interventions, calorie restriction and pravastatin administration, during pregnancy in a rhesus macaque model with the hypothesis that these interventions would normalize metabolic dysregulation in pregnant mothers leading to an improvement in infant metabolic and cognitive/social development. A total of 19 obese mothers were assigned to either one of the two intervention groups (n=5 for calorie restriction; n=7 for pravastatin) or an obese control group (n=7) with no intervention, and maternal gestational samples and postnatal infant samples were compared with lean control mothers (n=6). Gestational calorie restriction normalized one-carbon metabolism dysregulation in obese mothers but altered energy metabolism in their offspring. Although administration of pravastatin during pregnancy tended to normalize blood cholesterol in the mothers, it potentially impacted the gut microbiome and kidney function of their offspring. In the offspring, both calorie restriction and pravastatin administration during pregnancy tended to normalize the activity of AMPK in the brain at 6 months, and while results of the Visual Paired-Comparison test, which measures infant recognition memory, were not significantly impacted by either of the interventions, gestational pravastatin administration, but not calorie restriction, tended to normalize anxiety assessed by the Human Intruder test. Although the two interventions tested in a non-human primate model led to some improvements in metabolism and/or infant brain development, negative impacts were also found in both mothers and infants. Our study emphasizes the importance of assessing gestational interventions for maternal obesity on both maternal and offspring long-term outcomes.</p>
