Data from: The classical and alternative circulating renin-angiotensin system in normal dogs and dogs with stage B1 and B2 myxomatous mitral valve disease

The behavior of the comprehensive circulating renin‐angiotensin system (RAS) in dogs with myxomatous mitral valve disease (MMVD) before the onset of congestive heart failure remains largely unexplored.

Hypothesis/Objectives: The classical and alternative RAS activity and aldosterone concentrations will be significantly higher in dogs with American College of Veterinary Internal Medicine (ACVIM) stage B2 MMVD compared to normal dogs and dogs with ACVIM stage B1 MMVD.

Animals: One hundred seventeen client‐owned dogs (normal = 60; B1 = 31; B2 = 26).

Methods: Prospective observational study. Angiotensin peptides (AP) and aldosterone concentrations were measured using liquid chromatography and mass spectrometry. Angiotensin converting enzymes 1 and 2 (ACE, ACE2) and renin activity surrogates were calculated from AP concentrations. Equilibrium dialysis (ED) and immediate protease inhibition (PI) methods of AP quantification were compared in 14 healthy dogs.

Results: Core RAS activity and aldosterone concentrations did not differ among the 3 groups. However, the balance between the alternative and classical RAS differed, with dogs with stage B2 MMVD having significantly higher ACE2 activity surrogate (ACE2surr) when compared to normal dogs (adjusted P = .02; ratio of medians for ACE2surr [B2:normal], 1.89; 95% confidence interval [CI]: 1.4‐2.6). The ED and PI methods of AP quantification were highly correlated (AngI, r = .9, P < .0001; AngII, r = .8, P = .001).

Conclusions and Clinical Importance: Circulating alternative RAS activity, specifically the surrogate measure of ACE2 activity, was increased in dogs with stage B2 MMVD as compared to normal dogs. Equilibrium dialysis results are analogous to immediate protease inhibition in dogs.

Data From: B1 Field inhomogeneity correction for qDESS T2 mapping: application to rapid bilateral knee imaging

Purpose: T2 mapping is a powerful tool for studying osteoarthritis (OA) changes and bilateral imaging may be useful in investigating the role of between-knee asymmetry in OA onset and progression. The quantitative double-echo in steady-state (qDESS) can provide fast simultaneous bilateral knee T2 and high-resolution morphometry for cartilage and meniscus.
The qDESS uses an analytical signal model to compute T2 relaxometry maps, which require knowledge of the flip angle (FA).
In the presence of B1 inhomogeneities, inconsistencies between the nominal and actual FA can affect the accuracy of T2 measurements.
We propose a pixel-wise B1 correction method for qDESS T2 mapping exploiting an auxiliary B1 map to compute the actual FA used in the model.

Methods: The technique was validated in a phantom and in vivo with simultaneous bilateral knee imaging.
T2 measurements of femoral cartilage (FC) of both knees of six healthy participants were repeated longitudinally to investigate the association between T2 variation and B1.

Results: The results showed that applying the B1 correction mitigated T2 variations that were driven by B1 inhomogeneities.
Specifically, T2 left-right symmetry increased following the B1 correction (rc = 0.74 > rc = 0.69). Without the B1 correction, T2 values showed a linear dependence with B1. The linear coefficient decreased using the B1 correction (from 24.3 \(\pm\) 1.6 ms to 4.1 \(\pm\) 1.8) and the correlation was not statistically significant after the application of the Bonferroni correction (p-value > 0.01).

Conclusion: the study showed that B1 correction could mitigate variations driven by the sensitivity of the qDESS T2 mapping method to B1, therefore increasing the sensitivity to detect real biological changes. The proposed method may improve the robustness of bilateral qDESS T2 mapping, allowing for an accurate and more efficient evaluation of OA pathways and pathophysiology through longitudinal and cross-sectional studies.

This repository contains the raw data for reproducing the results reported in the paper. The data are provided in NIFTI format. The repository also contains the MALTAB scripts used to compute B1 maps from the NIFTI data.

Supplementary materials for: Use and detection of a vitamin B1 degradation product yields new views of the marine B1 cycle and plankton metabolite exchange

Vitamin B1 (thiamin) is a vital nutrient for most cells in nature including marine plankton. Early and recent experiments show that B1 degradation products instead of B1 can support the growth of marine bacterioplankton and phytoplankton. However, the use and occurrence of some degradation products remain uninvestigated – namely N-formyl-4-amino-5-aminomethyl-2-methylpyrimidine (FAMP) – which has been a focus of plant oxidative stress research. We investigated the relevance of FAMP in the ocean. Experiments and global ocean meta-omic data indicate that eukaryotic phytoplankton, including picoeukaryotes and harmful algal bloom species, use FAMP while bacterioplankton appear more likely to use deformylated FAMP, 4-amino-5-aminomethyl-2-methylpyrimidine (AmMP). Measurements of FAMP in seawater and biomass revealed that it occurs at picomolar concentrations in the surface ocean, heterotrophic bacterial cultures produce FAMP in the dark – indicating non-photodegradation of B1 by cells, and B1-requiring (auxotrophic) picoeukaryotic phytoplankton produce intracellular FAMP. Our results require an expansion of thinking about vitamin degradation in the sea, but also the marine B1 cycle – where it is now crucial to consider a new B1-related compound pool (FAMP) – as well as generation (dark degradation – likely via oxidation), turnover (plankton uptake), and exchange of the compound within networks of plankton.

H2020 ENODISE: RWTH Numerical Aeroacoustic Database Configuration B1

This RWTH dataset contains numerical data for aerodynamics and acoustics of configuration B1, which was defined in the H2020 ENODISE project (https://www.vki.ac.be/index.php/about-enodise). In this configuration, three 6-bladed XPROP-S propellers are installed side-by-side on the leading edge of a wing with an airfoil shape of NLF-Mod22(B).

The investigated operating point is:

The incoming flow velocity U∞ = 30 m/s, the wing angle of attack AoA = 2 deg, the propeller advance ratio J = 0.8, and the relative blade phase angle between propellers ∆Φ = 0.

Wall-resolved Large eddy simulations (LES) of the configuration are performed based on the multiphysics flow solver m-AIA of RWTH. The far-field noise is predicted by the Ffowcs-Williams and Hawkings (FW-H) method. The data set contains the time-averaged aerodynamic results predicted by the LES simulation and the aeroacoustic results calculated by the FW-H method. It should be noted that the aeroacoustic data is only a preliminary result and it will be updated soon.

More details of the simulation and measurement setups are explained in the attached document ENODISE_B1_RWTH.pptx.
