Growing Arabidopsis pollen tubes expressing genetically encoded reporters for calcium and pH

<p>Spatiotemporal dynamics of intracellular pH (H+) and calcium (Ca2+) in growing Arabidopsis pollen tubes. Fluorescence time-lapses&nbsp;of&nbsp;pollen tubes expressing ratiometric sensors under the pollen-specific LAT52 promoter,&nbsp;namely CaMeleon (Ca2+) [Damineli et al., 2017; DOI: 10.1093/jxb/erx032] and pHluorin (pH) [Hoffmann et al., 2020; DOI: 10.1038/s41467-020-16253-1], were acquired every 4&thinsp;seconds with an inverted microscope equipped with a front-illuminated camera (2560 &times; 2160, pixel size 6.45&thinsp;&mu;m), a fluorescence illuminator, and a &times;63 1.2NA water immersion objective lens. The CaMeleon filters were set as follows: excitation 426-450 nm (CFP) and 505-515 nm (YFP), emission 458-487&thinsp;nm (CFP) and 520-550 nm (YFP). As for pHluorin, the filter settings were: excitation 318-390&thinsp;nm (DAPI) and 428-475&thinsp;nm (FITC), emission 435-448&thinsp;nm (DAPI) and 523-536&thinsp;nm (FITC) [from Damineli et al., 2017;&nbsp;Hoffmann et al., 2020].</p>

Decreased FAM13B expression increases atrial fibrillation susceptibility by regulating sodium current and calcium handling

<p><strong>Objectives</strong>: To determine the causal genetic variant and gene and the mechanism for the atrial fibrillation (AF) genome wide association study (GWAS) locus on chromosome 5q31.</p> <p><strong>Background</strong>: <em>FAM13B</em> expression is strongly associated with the lead AF GWAS variant at 5q31.  However, the regulatory variant controlling <em>FAM13B</em> expression and the mechanism by which <em>FAM13B</em> impacts AF susceptibility are not known.</p> <p><strong>Methods</strong>: Bioinformatics, reporter gene transfections, gel shifts, and gene editing were used to identify the variant regulating <em>FAM13B</em> expression. RNAseq after <em>FAM13B</em> knockdown in stem cell-derived cardiomyocytes (iCMs) identified downstream processes. Patch clamp and calcium transient assays were used to assess downstream mechanisms. <em>Fam13b</em> knockout (KO) mice were studied for heart structural and functional changes, and pacing-induced arrhythmia.</p> <p><strong>Results</strong>: rs17171731 was identified as the regulatory variant controlling <em>FAM13B</em> expression, with decreased enhancer activity of the risk allele.  Knockdown of <em>FAM13B</em> in iCMs altered expression of &gt;1000 genes including <em>SCN2B</em> and led to pro-arrhythmogenic changes in the late sodium current and Ca<sup>2+</sup> cycling.  FAM13B is a member of the Rho GTPase-activating protein (RhoGAP) gene family, but failed to demonstrate RhoGAP activity.  GFP-tagged <em>FAM13B</em> expressed in iCMs localized at the Z-disc and plasma membrane. Fam13b knockout mice had increased basal p-wave duration and QT interval, and were more susceptible to pacing-induced arrhythmias vs. controls.</p> <p><strong>Conclusions</strong>: This study went from an AF GWAS locus to identify the causal variant and gene, mechanisms for this association, and confirmed arrhythmia susceptibility in <em>Fam13b</em> KO mice. FAM13B and downstream effectors are potential targets for patient-specific therapeutics.</p>

Forest Inventory of a Calcium Amended Northern Hardwood Forest: Watershed 1, 2006, Hubbard Brook Experimental Forest

In order to evaluate the role of Ca supply in regulating the structure and function of base-poor forest and aquatic ecosystems, the Ca content of soil was increased through the application of wollastonite (CaSiO3) in October 1999. Forest inventory surveys were initiated in 1996 and repeated at 5 year intervals. This data set includes 2006 inventory measurements.The data consists of a total inventory of all trees >=10 cm diameter-at-breast-height (dbh) on the whole of the watershed (11.8 ha), as measured in each of the 200 25 m x 25 m plots. Trees >=2 to <=10 cm dbh were subsampled using a 3 meter wide strip along one edge of each 25 m x 25 m plot. With the addition of tree tags in 2006 on all trees >=10 cm dbh, tracking of individual trees is now possible. These data were gathered as part of the Hubbard Brook Ecosystem Study (HBES). The HBES is a collaborative effort at the Hubbard Brook Experimental Forest, which is operated and maintained by the USDA Forest Service, Northern Research Station.

Differential impacts of calcium and aluminum treatments on foliar and sapwood nutrition and metabolism of sugar maple trees growing at the west edge of WS6 of the Hubbard Brook Experimental Forest from 1997-2009.

Plot levels Ca and Al additions study at WS6 BACKGROUD: To better evaluate the interaction of Ca depletion and Al mobilization on a northern hardwood forest, the Nutrient Perturbation (NuPert) study was initiated in 1995, west of the biogeochemical reference watershed (W6) at the HBEF (43.95411°N, 71.74779°W). The study area is on a south-facing slope, with an elevational range of 700–760 m, and most soils are classified as either Aquic Haplorthods or Aquic Haplumbredts (Halman et al. 22015 and references therein). Twelve sugar maple (Acer saccharum Marshall) dominated plots (45 m × 45 m) were randomly assigned to one of three treatments (Ca addition, Al addition, or control (no addition)), yielding four replicates of each treatment in the study. In addition to sugar maple, American beech (Fagus grandifolia Ehrh.) and yellow birch (Betula alleghaniensis Britt.) are co-occurring tree species in these plots, whereas hobblebush (Viburnum lantanoides Michx.) and striped maple (Acer pensylvanicum L.) dominate the understory. Treatments began in 1995 with annual CaCl2 (2.5 g·m−2) and AlCl3 (0.9 g·m−2) applications occurring each fall or spring during leafless periods. The use of CaCl2 was halted in 1999 in favor of a one-time application (38 g·m−2) of wollastonite (CaSiO3, a slow-release form of Ca). Thereafter, AlCl3 additions occurred in alternate years in fall or spring. Foliage was collected from sunlit branches in the upper to mid canopy dominant sugar maple trees. Five trees per plot of each group were selected for sampling in August 2008. There was an ice storm in 1998 at this site which might have interfered with this study findings. In 2009, tiny sapwood plugs were also collected. All the analyses were carried out using the procedures described under section A (Ca-supplementation study). Data from: Nonlinearities between inhibition and T-type calcium channel activity bidirectionally regulate thalamic oscillations

<p>Absence seizures result from 3-5 Hz generalized thalamocortical oscillations that depend on highly regulated inhibitory neurotransmission in the thalamus. Efficient reuptake of the inhibitory neurotransmitter GABA is essential, and reuptake failure worsens seizures. Here, we show that blocking GABA transporters (GATs) in acute brain slices containing key parts of the thalamocortical seizure network modulates epileptiform activity. As expected, we found that blocking either GAT1 or GAT3 prolonged oscillations. However, blocking both GATs unexpectedly suppressed oscillations. Integrating experimental observations into single-neuron and network-level computational models shows how a non-linear dependence of T-type calcium channel opening on GABA<sub>B</sub> receptor activity regulates network oscillations. Receptor activity that is either too brief or too protracted fails to sufficiently open T-type channels necessary for sustaining oscillations. Only within a narrow range does prolonging GABA<sub>B</sub> receptor activity promote channel opening and intensify oscillations. These results have implications for therapeutics that modulate GABA transporters.</p>
