Integration of environmental DNA metabarcoding technique to reinforce fish biodiversity assessments in seagrass ecosystems: A case study of Gazi Bay Seagrass meadows
<p><span>Assessing biodiversity in marine nearshore ecosystems is crucial for effective management, especially in the context of climate change and overexploitation of marine resources. Conventional methods often fall short in providing comprehensive information for managing seagrass ecosystems. However, the emergence of environmental DNA (eDNA) techniques has transformed the field by enabling non-invasive surveys that are cost-effective and provide detailed information with high resolution. In this study, we utilized eDNA to assess fish diversity and compared its effectiveness to conventional techniques such as catch assessment surveys and underwater surveys. </span>We sampled three habitats (A: mangrove-seagrass, B: seagrass only, and C: coral-seagrass) with 4 replicates. Site A recorded 8 fish species, site B had 16 species, and site C, characterized by coral and seagrass habitats, exhibited the highest fish diversity with 45 species (mean H' index = 2.455), underscoring its ecological importance. To ensure accurate taxonomic identification, we utilized an updated MiFish reference database containing a larger number of fish species compared to the initial library. This expanded reference database with 9,569 fish species, facilitated more precise identification and enhanced the reliability of our findings. Notably, the eDNA technique outperformed conventional methods by detecting 23 additional fish species that went undetected using traditional surveys. Moreover, our study documented five fish species previously unknown to occur within the study region, further emphasizing the value of eDNA analysis in uncovering hidden biodiversity. These findings strongly advocate for integrating eDNA techniques into the monitoring and assessment of biodiversity in shallow tropical habitats of the Western Indian Ocean. By leveraging eDNA surveys, we can gain valuable insights into fish diversity, discover hidden species, and make informed decisions for the conservation and management of these ecologically significant areas.</p>

Environmental impact assessment for large carnivores: a methodological review of the wolf (Canis lupus) monitoring in Portugal
<p>The continuous growth of the global human population results in increased use and change of landscapes, with infrastructures like transportation or energy facilities, being a particular risk to large carnivores. Environmental Impact Assessments were established to identify the probable environmental consequences of any new proposed project, find ways to reduce impacts, and provide evidence to inform decision making and mitigation. Portugal has a wolf population of around 300 individuals, designated as an endangered species with full legal protection. They occupy the northern mountainous areas of the country which has also been the focus of new human infrastructures over the last 20 years. Consequently, dozens of wolf monitoring programs have been established to evaluate wolf population status, to identify impacts, and to inform appropriate mitigation or compensation measures. We reviewed Portuguese wolf monitoring programs to answer four key questions: do wolf programs examine adequate biological parameters to meet monitoring objectives? is the study design suitable for measuring impacts? are data collection methods and effort sufficient for the stated inference objectives? and do statistical analyses of the data lead to robust conclusions? Overall, we found a mismatch between the stated aims of wolf monitoring and the results reported, and often neither aligns with the existing national wolf monitoring guidelines. Despite the vast effort expended and the diversity of methods used, data analysis makes almost exclusive use of relative indices or summary statistics, with little consideration of the potential biases that arise through the (imperfect) observational process. This makes comparisons of impacts across space and time difficult and is therefore unlikely to contribute to a general understanding of wolf responses to infrastructure-related disturbance. We recommend the development of standardized monitoring protocols and advocate for the use of statistical methods that account for imperfect detection to guarantee accuracy, reproducibility, and efficacy of the programs.</p>

Socio-economic and environmental impacts of ants: data to support global assessments
<p>Risk assessments are fundamental to invasive species management and are underpinned by comprehensive characterization of invasive species impacts. Our understanding of the impacts of invasive species is growing constantly, and several recently developed frameworks offer the opportunity to systematically categorize environmental and socio-economic impacts of invasive species. Invasive ants are among the most widespread and damaging invaders. We provide a global, comprehensive assessment on the impacts of ants and propose a priority list of risk species. We used the Socio-Economic Impact Classification for Alien Taxa (SEICAT), Environmental Impact Classification for Alien Taxa (EICAT) and Generic Impact Scoring System (GISS) to analyse 642 unique sources for 100 named species. This dataset includes the raw recrods on which our assessments were based, and some summary data. Other data can be found in the publication.</p>
Assessing the conservation priority of freshwater lake sites based on taxonomic, functional and environmental uniqueness
<p><strong>Aim: </strong>We propose a novel approach that considers taxonomic uniqueness, functional uniqueness and environmental uniqueness, and show how it can be used in guiding conservation planning. We illustrate the approach using data for lake biota and environment.</p> <p><strong>Location: </strong>Lake Puruvesi, Finland.</p> <p><strong>Methods: </strong>We sampled macrophytes and macroinvertebrates from the same 18 littoral sites. By adapting the original 'ecological uniqueness' approach, we used distance‒based methods to calculate measures of taxonomic (LCBD‒t), functional (LCBD‒f) and environmental (LCEH) uniqueness for each site. We also considered the numbers and locations of the sites needed to protect up to 70% of total variation in taxonomic, functional or environmental features in the studied part of the lake.</p> <p><strong>Results: </strong>Relationships between taxonomic (LCBD‒t), functional (LCBD‒f) and environmental (LCEH) uniqueness were generally weak, and only the relationship between macrophyte LCBD‒t and LCBD‒f was statistically significant. Overall, however, if the whole biotic dataset was considered, macroinvertebrate LCBD‒f values showed a consistent positive relationship with macrophyte LCBD‒f. Depending on the measure of site uniqueness, between one third to one half of the sites could help protect up to 70% of the ecological uniqueness of the studied part of Lake Puruvesi.</p> <p><strong>Main conclusions: </strong>Although the dataset examined originated from a large lake system, the approach we proposed here can be applied in different ecosystems and at various spatial scales. An important consideration is that a set of sites has been sampled using the same methods, resulting in species and environmental matrices that can be analysed using the methodological approach proposed here. This framework can be easily applied to grid‒based data, sets of islands, or sets of forest fragments. We suggest that the approach based on taxonomic, functional and environmental uniqueness will be a useful tool in guiding nature conservation and ecosystem management, especially if associated with meta‒system ideas or network thinking.</p>
Data from: Experimental assessment of downstream environmental DNA patterns under variable fish biomass and river discharge rates
<p><span>The development of environmental DNA (eDNA) methods towards implementation as a cost-effective, nonlethal tool for fish biomonitoring in lotic environments requires insights on the temporal and spatial distribution of eDNA in river systems. Yet, little is known on how downstream eDNA dispersal is affected by the combination of river discharge and source biomass effects. In this study, we aimed at unravelling the effect of source- and system-specific processes on the stream reach of eDNA. We used a longitudinal cage study in two river sections characterized by a significantly different discharge rate, where two invasive and two native fish species were introduced under two contrasting biomass treatments. Using droplet digital PCR (ddPCR) analyses, we found that eDNA concentrations become strongly reduced 2 km downstream from the source, an effect that is strengthened with increasing river discharge and coinciding dilution effects. Higher discharge rates resulted in equal or even higher detection probabilities at increasing distance from the source. The introduction of high fish stock biomass resulted in an increase of eDNA concentrations, as well as detection probabilities and in parallel reduced the stochasticity of the measurements. A peak in eDNA concentrations at a downstream distance ranging between 300 m and 2 km confirms the complexity of plume-shaped downstream eDNA patterns. Our results showed interspecific variation in eDNA emission and suggest species-specific differences in eDNA persistence. This study underlines the impact of both river discharge rate and source biomass on downstream eDNA detection and dispersal patterns. </span></p>
