Innovative Strategies for Blood-Brain Barrier (BBB) Permeability Modeling: Harnessing the Power of Machine Learning-based q-RASAR Approach
<p>In the current research, we have unveiled an advanced technique termed the quantitative Read-Across Structure-Activity Relationship (q-RASAR) framework to harnesses the power of machine learning (ML) for significantly enhancing the precision of predictions related to blood-brain barrier (BBB) permeability. It is important to emphasize that the central objective of this study is not to introduce another model for predicting BBB permeability. Instead, our focus is on highlighting the improvement in predicting the BBB permeability of organic compounds by introducing the q-RASAR approach. This innovative methodology strives to enhance the precision of evaluating neuropharmacological implications and streamline the drug development process. In this investigation, we developed an ML-based q-RASAR PLS model using a dataset comprising 1012 compounds of diverse classes of heterocyclic and aromatic hydrocarbons, obtained from the freely accessible B3DB database (accessible at <a href="https://github.com/theochem/B3DB">https://github.com/theochem/B3DB</a>) to predict BBB permeability during the lead discovery phase for central nervous system (CNS) drugs. The model's predictive capability underwent validation using two external sets, encompassing a total of 1,130,315 compounds, including synthetic compounds and natural products (NPs) for data gap filling and other two external sets comprising 116 drug-like/drug compounds from FDA and ChEMBL databases to assess the model's reliability. This study aimed to bridge the data gap by employing a predictive model to estimate the impact of brain-plasma concentration ratios on BBB permeability for both synthetic compounds and natural products (NPs). To further enhance predictability, we have developed various other ML-based q-RASAR models. The insights from the developed model highlight the pivotal roles played by hydrophobicity, electronic effects, degree of ionization and steric factors as essential features facilitating the traversal of the blood-brain barrier. This research not only advances our understanding of the molecular determinants influencing the permeability of central nervous system drugs but also establishes a versatile computational platform for the rapid assessment of diverse compounds, facilitating informed decision-making in the realms of drug development and design.</p>

Data from: Harnessing ant defence at fruits reduces bruchid seed predation in a symbiotic ant-plant mutualism
In horizontally transmitted mutualisms, mutualists disperse separately and reassemble in each generation with partners genetically unrelated to those in the previous generation. Because of this, there should be no selection on either partner to enhance the other's reproductive output directly. In symbiotic ant–plant mutualisms, myrmecophytic plants host defensive ant colonies, and ants defend the plants from herbivores. Plants and ants disperse separately, and, although ant defence can indirectly increase plant reproduction by reducing folivory, it is unclear whether ants can also directly increase plant reproduction by defending seeds. The neotropical tree Cordia alliodora hosts colonies of Azteca pittieri ants. The trees produce domatia where ants nest at stem nodes and also at the node between the peduncle and the rachides of the infloresence. Unlike the stem domatia, these reproductive domatia senesce after the tree fruits each year. In this study, I show that the tree's resident ant colony moves into these ephemeral reproductive domatia, where they tend honeydew-producing scale insects and patrol the nearby developing fruits. The presence of ants significantly reduced pre-dispersal seed predation by Amblycerus bruchid beetles, thereby directly increasing plant reproductive output.
Data from: Harnessing stratigraphic bias at the section scale: conodont diversity in the Homerian (Silurian) of the Midland Platform, England
Fossil abundance and diversity in geological successions are subject to bias arising from shifting depositional and diagenetic environments, resulting in variable rates of fossil accumulation and preservation. In simulations, this bias can be constrained based on sequence-stratigraphic architecture. Nonetheless, a practical quantitative method of incorporating the contribution of sequence-stratigraphic architecture in community palaeoecology and diversity analyses derived from individual successions is missing. As a model of faunal turnover affected by the stratigraphic bias, we use the 'Mulde event', a postulated mid-Silurian interval of elevated conodont turnover, which coincides with global eustatic sea-level changes and which has been based on regionally constrained observations. We test whether conodont turnover is highest at the boundary corresponding to the 'event' and post-'event' interval against the alternative that conodont turnover reflects habitat tracking and peaks at facies shifts. Based on the previously documented, parasequence-level stratigraphic framework of sections in the northern and central part of the Midland Platform, the relative controls of sequence-stratigraphic architecture, time and depositional environment over conodont distribution are evaluated using permutational multivariate analysis of variance. The depositional environment controls the largest part of variability in conodont assemblage composition, whereas the postulated 'Mulde event', or genuine temporal change in conodont diversity, cannot be detected. Depending on the binning of the stratigraphic succession, contrasting diversity and turnover patterns can be produced. The simple approach proposed here, emulating partitioning of β diversity into spatial and temporal components, may help to constrain the stratigraphic bias, even at the scale of an individual section.

<p>This repository contains the data released in the paper "Harnessing the Hubble Space Telescope Archives: A Catalogue of 21,926 Interacting Galaxies"<em> </em>(DOI: <a href="https://ui.adsabs.harvard.edu/abs/2023ApJ...948...40O/abstract">10.3847/1538-4357/acc0ff</a>).</p> <p>We release the catalogue of interacting galaxies found in this work, as well as any objects of interest found during the visual inspection stage of de-contamination. Each catalogue contains the SourceID, Right Ascension, Declination and the "Status" of each object. The status shows whether an object has appeared in the astrophysical literature prior to this work. <strong>Each catalogue is contained in a .csv file</strong>.</p> <p>The exception to this is the interacting galaxy catalogue itself, which also contains the Zoobot prediction of each source to be an interacting galaxy.</p> <p>We also include the images that correspond to each catalogue. <strong>These are contained within the .tar.gz files in the repository.</strong> Each .tar.gz filename corresponds to the catalogue .csv filename. The individual filenames are the SourceIDs of the corresponding objects.</p> <p>Please note: These images have been saved as 150x150 gray images using only the F814W filter of the <em>HST</em>. Different dimension images/multi-band photometry will have to be found by the user. When input into our Zoobot model, these had been resized to 300x300.</p> <p>Please also note: <strong>Each sub-category of object has</strong> <strong>only been classified visually</strong>.</p> <p>A list of the catalogue definitions is below:</p> <ol> <li>agn-catalogue - Catalogue of sources containing an Active Galactic Nuclei or a Quasar.</li> <li>groups-catalogue - Catalogue of sources containing a Galaxy Group.</li> <li>high_z-catalogue - Catalogue of sources containing high redshift galaxies.</li> <li>interacting-catalogue - Catalogue of sources predicted to be interacting galaxies.
