Data from: Detecting genes for variation in parasite burden and immunological traits in a wild population: testing the candidate gene approach

Identifying the genes underlying phenotypic variation in natural populations can provide novel insight into the evolutionary process. Here we test the candidate gene approach to identifying loci involved in variation in gastrointestinal parasite burden, in a wild population of Soay sheep. A comprehensive literature review, Gene Ontology databases, and comparative genomics resources were used to generate a list of candidate genes. In a pilot study these candidates, along with 50 random genes, were then sequenced in pools of Soay sheep with low and high gastrointestinal nematode burden. Further candidates were identified from SNPs that were highly differentiated between high and low resistance sheep breeds. A panel of 192 candidate and control SNPs were then typed in 960 individual Soay sheep to examine whether they individually explained variation in parasite burden, as measured as faecal egg count (FEC), as well as two immune measures. The cumulative effect of the candidate and control SNPs were estimated by fitting genetic relationship matrices (GRMs) as random effects in animal models of the three traits. No more significant SNPs were identified in the pilot sequencing experiment and association study than expected by chance. Furthermore, no significant difference was found between the proportions of candidate or control SNPs that were found to be significantly associated with parasite burden/immune measures. No significant effect of the candidate or control gene GRMs was found. There is thus little support for the candidate gene approach to the identification of loci explaining variation in parasitological and immunological traits in this population.

Genetic and immunological evaluation of children with inborn errors of immunity and hypoxemic COVID-19 pneumonia

<p><strong><em>Background:</em></strong> SARS-CoV-2 has infected more than 450 million people around the world. In about 10% of cases, the infection leads to hypoxemic pneumonia, which is much rarer in children, while multisystem inflammation is seen almost only in children.</p> <p><strong><em>Methods:</em></strong> We evluated 31 young patients (0.5-19 years) who had both pre-existing inborn errors of immunity (IEI) and were later diagnosed with COVID-19 complications. Whole-exome sequencing was performed, SARS-CoV-2 specific antibodies and autoantibody against type I interferons were measured and plasma inflammatory factors were profiled. We also reviewed COVID-19 disease severity and outcome in IEI patients in the literature.</p> <p><strong><em>Results:</em></strong> &nbsp;A potential genetic cause of the IEI was identified in 28 patients (90.3%). The IgM and IgG-specific antibody response against the spike protein of the virus was positive in 14 (66.6%) of the IEI patients tested. Autoantibodies neutralizing type I interferons were identified in the plasma of two patients (6.8% of tested samples). Five patients fulfilled the diagnostic criteria of multisystem inflammatory syndrome in children (MIS-C). Eleven patients (35.4%) died due to COVID-19 complications showing a 3000 fold higher mortality rate compared to normal COVID-19 death rate in children (0.01%). For the additional 696 IEI patients reported till date, severe presentation of COVID-19 was observed in 23.3% of cases. Furthermore, a 5-fold higher mortality rate was observed, when compared with global infection-fatality reports.</p> <p><strong><em>Conclusions:</em></strong> Elucidating the genetic basis of IEI patients with severe/critical COVID-19 may help to better understand the human innate and adaptive immune response against SARS-CoV-2, which may assist to develop better preventive and therapeutic strategies.</p>

Nannochloropsis spp. as Feed Additive for the Pacific White Shrimp: Effect on Midgut Microbiology, Thermal Shock Resistance and Immunology

<p>This work aimed to evaluate Nannochloropsis spp. as feed additive in the diet of Pacific<br> white shrimp for their effect on midgut microbiology, thermal shock resistance and immunological<br> parameters. Initially, the digestibility of the microalgae meal was assessed, and the apparent<br> digestibility coefficient (ADC) was determined. The ADC was, in general, high in lipids (78.88%)<br> and eicosapentaenoic fatty acid (73.86%). Then, Nannochloropsis spp. were included in diets at four<br> levels (0, 0.5, 1 and 2% inclusion). The shrimp were reared in 500 L clear water tanks containing 20<br> shrimp per tank with an initial weight of 6.05 &plusmn; 0.06 g and fed four times a day. Shrimp fed with<br> supplemented diets containing Nannochloropsis spp. (0.5 and 2%) presented higher resistance to<br> thermal shock when compared to the non-supplemented group (control). Shrimp fed with 1 and 2%<br> of algae inclusion had a higher production of reactive oxygen species (ROS) when compared to<br> other treatments. No statistical difference was observed in the immunological parameters and<br> microbiology of the intestinal tract. Thus, the inclusion of Nannochloropsis spp. in shrimp diets at 0.5<br> and 2% levels increases resistance to thermal shock and ROS production in shrimp.</p>

Immunological indices and parasites in blackbirds

<p>1. Despite the intensive use of immune indices in immunoecology, whether to interpret the results of immune indices in terms of actual immune competence (i.e., ability to control and clear parasite infections as indicated by high values of immune indices associated with low parasite loads) or current immune activation (pathogenic infection being associated with high parasite load and high values of immune indices) is still an open question. Most studies to date have produced contrasting results focused on the effect of a single parasite species despite the fact that hosts usually harbour a community of parasites that influence one another's impact on host immune response.</p> <p>2. We simultaneously assessed blood parasites, intestinal parasites and ectoparasite loads in male blackbirds and compared these measures to several immune indices to investigate how parasites explain the variation around the mean of these immune indices.</p> <p>3. Parasite loads covaried within hosts. Immune indices better reflected the interacting effects of these parasites than the independent effect of each parasite. Immune indices may therefore be better indicators of ongoing pathogenic infections than immune competence. Furthermore, intestinal parasites explained a significant part of the variance in most immune indices through their interactions with other parasites, suggesting that they have a strong influence in modulating immune function.</p> <p>4. Taking the parasite community into account in immunoecology studies will certainly help increase our understanding of immune indices.</p>
