Soluble T-cadherin promotes pancreatic β-cell proliferation by upregulating Notch signaling

<p class="MsoNormal"><span>Endogenous </span><span>humoral factors </span><span>that</span><span> link systemic and/or local insulin demand to pancreatic β-cells have not been identified. Here</span><span>,</span><span> we demonstrated that T-cadherin, a unique glycosylphosphatidylinositol-anchored cadherin primarily expressed in vascular endothelial cells and cardiac and skeletal muscle cells, but not in pancreatic β-cells, was secreted as soluble forms and was important for β-cell proliferation. <em>Cdh13</em> (T-cadherin) knockout mice exhibited impaired glucose handling due to attenuated β-cell proliferation under high-fat diet conditions. The gene expression analyses indicated the impairment in cell cycle and Notch signaling in the islets of T-cadherin knockout mice under high-fat diet conditions. In streptozotocin-induced diabetes, the replacement of soluble T-cadherin improved β-cell mass and blood glucose </span><span>levels</span><span> in T-cadherin knockout mice. </span><span>R</span><span>ecombinant soluble T-cadherin upregulated Notch signaling in cultured murine islets. We concluded that soluble T-cadherin could work as an endogenous humoral factor whose signaling pathways including Notch signaling regulate β-cell proliferation under diabetic conditions in mice.</span></p>

Clonally related, Notch-differentiated spinal neurons integrate into distinct circuits EPhys raw data

<p>Shared lineage has diverse effects on patterns of neuronal connectivity. In mammalian cortex, excitatory sister neurons assemble into shared microcircuits, whereas throughout the <em>Drosophila</em> nervous system, Notch-differentiated sister neurons diverge into distinct circuits. Notch-differentiated sister neurons have been observed in vertebrate spinal cord and cerebellum, but whether they integrate into shared or distinct circuits remains unknown. Here we evaluate the connectivity between sister V2a/b neurons in the zebrafish spinal cord. Using an <em>in</em> <em>vivo</em> labeling approach, we identified pairs of sister V2a/b neurons born from individual Vsx1+ progenitors and observed that they have similar axonal trajectories and proximal somata. However, paired whole-cell electrophysiology and optogenetics revealed that sister V2a/b neurons receive input from distinct presynaptic sources, do not communicate with each other, and connect to largely distinct targets. These results resemble the divergent connectivity in <em>Drosophila</em> and represent the first evidence of Notch-differentiated circuit integration in a vertebrate system.</p>

Hardaway Side Notched

This stone projectile point is known as a Hardaway Side Notched, and it was found at a multi-component site (31WA2199) in eastern Wake County, NC. This type of artifact was used as a spear or dart point and likely was secondarily used as a knife and/or scraping tool. It dates to the late Paleoindian period (c. 8500-7900 BC). This artifact is made from a metavolcanic stone commonly called rhyolite and measures 5.1 cm long, 3.2 cm wide, and 0.6 cm thick.
