Data for: Effects of treated wastewater on the ecotoxicity of small streams – unravelling the contribution of chemicals causing effects

<p>Wastewater treatment plant effluents are important point sources of micropollutants. To assess how the discharge of treated wastewater affects the ecotoxicity of small to medium-sized streams we collected water samples up- and downstream of 24 wastewater treatment plants across the Swiss Plateau and the Jura regions of Switzerland. We investigated estrogenicity, inhibition of algal photosynthetic activity (photosystem II, PSII) and growth, and acetylcholinesterase (AChE) inhibition. At four sites, we measured feeding activity of amphipods (<em>Gammarus fossarum</em>) <em>in situ</em> as well as water flea (<em>Ceriodaphnia dubia</em>) reproduction in water samples. Ecotoxicological endpoints were compared with results from analyses of general water quality parameters as well as a target screening of a wide range of organic micropollutants with a focus on pesticides and pharmaceuticals using liquid chromatography high-resolution tandem mass spectrometry.</p> <p>Measured ecotoxicological effects in stream water varied substantially among sites: 17&beta;-estradiol equivalent concentrations (EEQ<sub>bio</sub>, indicating the degree of estrogenicity) were relatively low and ranged from 0.04 to 0.85 ng/L, never exceeding a proposed effect-based trigger (EBT) value of 0.88 ng/L. Diuron equivalent (DEQ<sub>bio</sub>) concentrations (indicating the degree of photosystem II inhibition in algae) ranged from 2.4 to 1576 ng/L and exceeded the EBT value (70&nbsp;ng/L) in one third of the rivers studied, sometimes even upstream of the WWTP. Parathion equivalent (PtEQ<sub>bio</sub>) concentrations (indicating the degree of AChE inhibition) reached relatively high values (37 to 1278 ng/L) mostly exceeding the corresponding EBT (196 ng/L PtEQ<sub>bio</sub>). Decreased feeding activity by amphipods or waterflea reproduction compared to the upstream site was observed at one of four investigated sites only. Results of the combined algae assay (PSII inhibition) correlated best with results of chemical analysis for PSII inhibiting herbicides. Estrogenicity was partly and AChE inhibition strongly underestimated based on measured steroidal estrogens respectively organophosphate and carbamate insecticides. An impact of dissolved organic carbon on results of the AChE inhibition assay was obvious. For this assay more work is required to further explore the missing correlation of bioassay data with &nbsp;chemical analytical data. Overall, the discharge of WWTP effluent led to increased estrogenicity, PSII and AChE inhibition downstream, irrespective of upstream land use.</p>

Replication files for: Integrative modeling of the spread of serious infectious diseases and corresponding wastewater dynamics

<p>This repository contains the inputs used and outputs produced by the urban water management modelling software ++SYSTEMS for the paper Integrative Modeling of the Spread of Serious Infectious Diseases and Corresponding Wastewater Dynamics. It includes the following files:</p> <ol> <li>simulation_output.zip:</li> <ol> <li>In the subfolder infection_model, .csv and .txt files containing the agent-based model outputs for 250 simulations with homogeneous infection initialisation (2024_09_17) or localised infection initialisation (2024_10_15). These files were used as inputs for ++SYSTEMS.</li> <li>In the subfolder wastewater_model, .txt files containing the flow rates by pipe and the viral concentrations by sampling location for each combination of ABM simulation and rain/decay scenario. These files were the outputs of ++SYSTEMS.</li> </ol> <li>S1_INSIDe_Demonstrator_AreaList.txt: .txt file defining the area types and number of inhabitants for each surface area within the synthetic neighbourhood used in the paper. This file was also used as an input for ++SYSTEMS.<span>&nbsp;</span></li> <li>S2_systems_model_files.zip: .csv files defining the characteristics of the sewage system for the synthetic neighbourhood as well as the rain scenarios used in the paper. Simulated wastewater sequencing data for benchmarking SARS-CoV-2 variant abundance estimation

<p>To evaluate the accuracy of variant abundance&nbsp;predictions from wastewater sequencing, we built a collection of benchmarking datasets that resemble real wastewater samples. For each variant (B.1.1.7, B.1.351, B.1.427, B.1.429, P.1) we created a series of 33 benchmarks by simulating sequencing reads from a variant genome, as well as a collection of background (non-variant of concern/interest) sequences, such that the variant abundance ranges from 0.05% to 100%. Analogously, we created a second series of benchmarks, simulating reads only from the Spike gene of each SARS-CoV-2 genome. We refer to the first set of benchmarks as &quot;whole genome&quot; (WG)&nbsp;and to the second set of benchmarks as &quot;S-only&quot;. We repeated these simulations at different sequencing depths: 100x and 1000x coverage for the whole genome benchmarks, and 100x, 1000x, and 10,000x coverage for the S-only benchmarks.</p>

Toileting behaviours of the UK public: insights for reducing gender bias wastewater-based epidemiology sampling strategies

<p>Cross-sectional survey results from a toileting behaviour survey conducted&nbsp;between the 27th to the 28th of June 2022. Participants (<em>n</em>&nbsp;= 2109) were aged 18 years or older and were living in the UK.&nbsp;The survey consisted of 17 closed-ended questions, with 7 of the questions addressing specific demographic topics and 10 questions addressed toileting behaviour. The questionnaire was designed by a team consisting of environmental microbiologists, public health specialists, wastewater-based epidemiologists, and social scientists, based on the study objectives and incorporating information from previous studies on the same topic.&nbsp; First, self-report questions were asked on typical frequency of urination and defecation, followed by self-reports of frequency of urination and defecation at a variety of locations including at home, at work, educational buildings, transport hubs, and in public toilets. The comfort in urination/defecation at these locations for defecation and urination was also measured. Other questions about toileting behaviour and health monitoring were measured by statements with a 5-point Likert scale (e.g., strongly disagree to strongly agree).&nbsp;</p>
