Data for: Actions of Parathyroid hormone ligand analogs in humanized PTH1R knock-in mice
<p>Rodent models are commonly used to evaluate parathyroid hormone (PTH) and PTH-related protein (PTHrP) ligands and analogs for their pharmacologic activities and potential therapeutic utility towards diseases of bone and mineral ion metabolism. Divergence, however, in the amino acid sequences of rodent and human PTH receptors (rat and mouse PTH1Rs are 91% identical to the human PTH1R) can lead to differences in receptor-binding and signaling potencies for such ligands when assessed on rodent versus human PTH1Rs, as shown by cell-based assays in vitro. This introduces an element of uncertainty in the accuracy of rodent models for performing such pre-clinical evaluations. To overcome this potential uncertainty, we used a homologous recombination-based knock-in (KI) approach to generate a mouse (in host strain C57Bl/6N) in which cDNA encoding the human PTH1R replaces a segment (Exon 4) of the murine PTH1R gene such that the human and not the mouse PTH1R protein is expressed. Expression is directed by the endogenous mouse promoter and hence occurs in all biologically relevant cells and tissues and at appropriate levels. The resulting homozygous hPTH1R-KI (humanized) mice were healthy over at least ten generations and showed functional responses to injected PTH analog peptides that are consistent with a fully functional human PTH1R in target bone and kidney cells. The initial evaluation of these mice and their potential utility for predicting behavior of PTH analogs in humans is reported here. </p>
Cystargolide-based amide and ester Pz analogs as proteasome inhibitors and anticancer agents
<p>A series of cystargolide-based beta-lactone analogs containing nitrogen atoms at the Pz portion of the scaffold were prepared and evaluated as proteasome inhibitors and for their cytotoxicity profile towards several cancer cell lines. Inclusion of one, two or even three nitrogen atoms at the Pz portion of the cystargolide scaffold is well-tolerated, producing analogs with low nanomolar proteasome inhibition activity, in many cases superior to carfilzomib. Additionally, analog 8g, containing an ester and pyrazine group at Pz, was shown to possess significant activity towards RPMI 8226 cells (IC50 = 21 nM) and to be less cytotoxic towards the normal tissue model MCF10A cells than carfilzomib.</p>

Compounds with multi-target activity from X-ray structures, corresponding analog series, and associated scaffolds
<p>A total of 702 crystallographic ligands with activity against multiple targets from different protein families (multi-family ligands) were extracted from the Protein Data Bank (PDB). These ligands are made available as aromatic non-stereo SMILES strings together with their target information. A subset of these ligands were also found in the ChEMBL database yielding additional target annotations. Target and family assignments were made following the UniProt classification scheme. In addition, 133 analog-series-based (ASB) scaffolds were derived from series of PDB ligands and structural analogs identified in ChEMBL, which are also made available. For each ASB scaffold, the number of analogs, targets, and target families is provided (union of PDB and ChEMBL annotations).</p>

A high-resolution speleothem record of Marine Isotope Stage 11 as a natural analog to Holocene Asian summer monsoon variations
<p>A full-spectrum characterization of past interglacial climate is a necessary prerequisite for the detection and attribution of climate changes during the current interglacial. Here we present a speleothem record of Asian summer monsoon (ASM) during Marine Isotope Stage (MIS) 11 interglacial (MIS 11c), from Yongxing cave, China. The record’s unprecedented chronologic constraints and decadal-scale temporal resolution allow a precise and direct comparison of ASM between the MIS 11c and the Holocene. Our data suggest that orbital–centennial patterns of ASM were remarkably similar during both interglacial, including their pacing and structure. Notably, a multi-millennial stronger monsoon late in MIS 11c, the ‘Late-MIS 11c shift’, is similar to the Late Holocene strengthening of the ASM, the ‘2-kyr shift’. Thus the multi-centennial ASM weakening at the end of the ‘Late-MIS 11c shift’ could imply that the current century-long ASM waning trend may persist into the future, if only natural forcings are considered.</p>

Impact ionization dust detection with compact, hollow and fluffy dust analogs
<p>Datasets for the Planetary and Space Science article "Impact ionization dust detection with compact, hollow and fluffy dust analogs" (DOI: 10.1016/j.pss.2022.105536). The data contains two folders for dust impacts on the IIT and CAT targets of CDA. The folders contain separate tables with data for each run. Each table shows the registered CDA impacts for the corresponding run with impact time in UTC (determined by CDA) and all additional information about the impact that could be reconstructed from the measurements of CDA and other detectors (impact velocity, dust mass, ...). Data that could not be reconstructed are represented by NAN values in the tables.</p>
GPS data from cellphones for Mechanical Analog
<p>Motivated by the increasing need to develop a quantitative, science-based, predictive understanding of the dynamics and response of cities when subjected to hazards, in this paper we apply concepts from statistical mechanics and microrheology to develop mechanical analogs for cities with predictive capabilities. We envision a city to be a matrix where people (cell-phone users) are driven by the city's economy and other associated incentives while using the collection of its infrastructure networks in a similar way that thermally driven Brownian probe particles are moving within a complex viscoelastic material. Mean-square displacements (ensemble averages) of thousands of cell-phone users are computed from GPS location data to establish the creep compliance and the resulting impulse response function of a city. The derivation of these time-response functions allows the synthesis of simple mechanical analogs that model satisfactorily the city's behavior under normal conditions. Our study concentrates on predicting the response of cities to acute shocks (natural hazards that stress the entire urban area) that are approximated with a rectangular pulse with finite duration, and we show that the solid-like mechanical analogs for cities that we derived predict that cities revert immediately to their pre-event response suggesting that they are inherently resilient. Our findings are in remarkably good agreement with the recorded response of the Dallas metroplex following the February 2021 North American winter storm, which happened at a time for which we have dependable GPS location data.</p>
Data from: Functionally analogous body- and animacy-responsive areas in the dog (Canis familiaris) and human occipito-temporal lobe
<p>Comparing the neural correlates of socio-cognitive skills across species provides insights into the evolution of the social brain and has revealed face- and body-sensitive regions in the primate temporal lobe. Although from a different lineage, dogs share convergent visuo-cognitive skills with humans and a temporal lobe which evolved independently in carnivores. We investigated the neural correlates of face and body perception in dogs (<em>N </em>= 15) and humans (<em>N</em> = 40) using functional MRI. Combining univariate and multivariate analysis approaches, we found functionally analogous occipito-temporal regions involved in the perception of animate entities and bodies in both species and face-sensitive regions in humans. Though unpredicted, we also observed neural representations of faces compared to inanimate objects, and dog compared to human bodies in dog olfactory regions.
