CESM 1.2 climate model simulation output for: The Essential Role of Westerly Wind Bursts in ENSO Dynamics and Extreme Events Quantified in Model 'Wind Stress Shaving' Experiments
<p>Westerly wind bursts (WWBs)—brief but strong westerly wind anomalies in the equatorial Pacific—are believed to play an important role in El Niño Southern Oscillation (ENSO) dynamics, but quantifying their effects is challenging. Here, we investigate the cumulative effects of WWBs on ENSO characteristics, including the occurrence of extreme El Niño events, via modified coupled model experiments within Community Earth System Model (CESM1) in which we progressively reduce the impacts of wind stress anomalies associated with model-generated WWBs. In these "wind stress shaving" experiments we limit momentum transfer from the atmosphere to the ocean above a preset threshold, thus "shaving off" wind bursts. To reduce the tropical Pacific mean state drift, both westerly and easterly wind bursts are removed, although the changes are dominated by WWB reduction. As we impose progressively stronger thresholds, both ENSO amplitude and the frequency of extreme El Niño decrease, and ENSO becomes less asymmetric. The warming center of El Niño shifts westward, indicating less frequent and weaker Eastern Pacific (EP) El Niño events. Removing most of wind bursts-related wind stress anomalies reduces ENSO amplitude by 22%. The essential role of WWBs in the development of extreme El Niño events is revealed in the suppressed eastward migration of the western Pacific warm pool and hence a weaker Bjerknes feedback under wind shaving. Overall, our results reaffirm the importance of WWBs in shaping the characteristics of ENSO and its extreme events and imply that WWB changes with global warming could influence future ENSO.</p>
Single-cell burst size estimates
<p>Bacteriophage burst size is the average number of phage virions released from infected bacterial cells, and its magnitude depends on the duration of an intracellular progeny accumulation phase. Burst size is often measured at the population, not single-cell, level, and consequently, statistical moments are not commonly available. In this study, we estimated the bacteriophage lambda (λ) single-cell burst size mean and variance following different length intracellular accumulation periods by employing E. coli lysogens bearing lysis-deficient λ prophages. Single lysogens can be isolated and chemically lysed at desired times following prophage induction to quantify progeny intracellular accumulation within individual cells. Our data show that λ phage burst size initially increased exponentially with increased lysis time (i.e., period between induction and chemical lysis), and then saturated at longer lysis times. We also demonstrated that cell-to-cell variation or "noise" in lysis timing did not contribute significantly to burst size noise. The burst size noise remained constant with increasing mean burst size. The most likely explanation for the experimentally observed constant burst size noise was that cell-to-cell differences in burst size originated from intercellular heterogeneity in cellular capacities to produce phages. The mean burst size measured at different lysis times was positively correlated to cell volume, which may determine the cellular phage production capacity. However, experiments controlling for cell size indicated that there are other factors in addition to cell size that determine this cellular capacity.</p>

Data from: Legacy and emerging per- and polyfluoroalkyl substances suppress the neutrophil respiratory burst
<p>Per- and polyfluoroalkyl substances (PFASs) are used in a multitude of processes and products, including nonstick coatings, food wrappers, and fire-fighting foams. These chemicals are environmentally persistent, ubiquitous, and can be detected in the serum of 98% of Americans. Despite evidence that PFASs alter adaptive immunity, few studies have investigated their effects on innate immunity. We investigated the impact of nine environmentally relevant PFASs [perfluorooctanoic acid (PFOA), perfluorooctane sulfonic acid potassium salt (PFOS-K), perfluorononanoic acid (PFNA), perfluorohexanoic acid (PFHxA), perfluorohexane sulfonic acid (PFHxS), perfluorobutane sulfonic acid (PFBS), ammonium perfluoro(2-methyl-3-oxahexanoate) (GenX), 7H-perfluoro-4-methyl-3,6-dioxaoctane sulfonic acid (Nafion byproduct 2), and perfluoromethoxyacetic acid sodium salt (PFMOAA-Na)] on one component of the innate immune response, the neutrophil respiratory burst. The respiratory burst is a key innate immune process by which microbicidal reactive oxygen species (ROS) are rapidly induced by neutrophils in response to pathogens, and defects in the respiratory burst can increase susceptibility to infection. We utilized larval zebrafish, a human neutrophil-like cell line, and primary human neutrophils to ascertain whether PFAS exposure inhibits ROS production in the respiratory burst. We observed that exposure to PFHxA and GenX suppresses the respiratory burst in zebrafish larvae and a human neutrophil-like cell line. GenX also suppresses the respiratory burst in primary human neutrophils. This report is the first to demonstrate that these PFASs suppress neutrophil function and supports the utility of employing zebrafish larvae and a human cell line as screening tools to identify chemicals that may suppress human immune function. </p>
