Accelerated landings in stingless bees are triggered by visual threshold cues

<p>Most flying animals rely primarily on visual cues to coordinate and control their trajectory when landing. Studies of visually-guided landing typically involve animals that decrease their speed before touchdown. Here, we investigate the control strategy of the stingless bee <i>Scaptotrigona depilis</i>, which instead accelerates when landing on its narrow hive entrance. By presenting artificial targets that resemble the entrance at different locations on the hive, we show that these accelerated landings are triggered by visual cues. We also found that <i>S. depilis</i> initiated landing and extended their legs when the angular size of the target reached a given threshold. Regardless of target size, the magnitude of acceleration was the same and the bees aimed for the same relative position on the target suggesting that <i>S. depilis</i> use a computationally simple but elegant 'stereotyped' landing strategy that requires few visual cues.</p>

Hemipteran defensive odors trigger predictable color biases in jumping spider predators

<p>Multimodal warning displays often pair one signal modality (odor) with a second modality (color) to avoid predation. Experiments with bird predators suggest these signal components interact synergistically, with aversive odors triggering otherwise hidden aversions to particular prey colors. In a recent study, this phenomenon was found in a jumping spider (<em>Habronattus trimaculatus</em>), with the defensive odor from a coreid bug (<em>Acanthocephala femorata</em>) triggering an aversion to red. Here, we explore how generalizable this phenomenon is by giving <em>H. trimaculatus</em> the choice between red or black prey in the presence or absence of defensive odors secreted from (1) eastern leaf-footed bugs (<em>Leptoglossus phyllopus</em>, Hemiptera), (2) grass stinkbugs (<em>Mormidea pama</em>, Hemiptera), (3) Asian ladybird beetles (<em>Harmonia axyridis</em>, Coleoptera), and (4) eastern lubber grasshoppers (<em>Romalea microptera</em>, Orthoptera). As expected, in the presence of the hemipteran odors, spiders were less likely to attack red prey (compared to no odor). Unexpectedly, the beetle and grasshopper odors did not bias spiders away from red. Our results with the hemipteran odors were unique to red; follow-up experiments indicated that these odors did not affect biases for/against green prey. We discuss our findings in the context of generalized predator foraging behavior and the functions of multimodal warning displays. </p>

Characteristics of the initial stage and return stroke currents of rocket-triggered lightning flashes in southern China

<p>This study investigates the initial stage (IS) and return stroke (RS) currents of 50 triggered lightning flashes (TLFs) that were conducted in southern China. The IS of the negative TLFs has a longer duration, and larger average current, charge transfer, and action integral than those reported elsewhere, with geometric means (GMs) of 347.9 ms, 132.5 A, 45.1 C, and 10.0 × 10<sup>3</sup> A<sup>2</sup> s, respectively. Two positive TLFs containing no RS have much greater average currents, charge transfers, and action integrals in the IS when compared with the negative TLFs. The RS has a greater peak current (17.2 kA; GM, same to below), charge transfer within 1 ms (1.3 C), and action integral within 1 ms (5.8 × 10<sup>3</sup> A<sup>2</sup> s), and shorter 10% to 90% rise time (0.4 μs) than elsewhere. The peak current is prominently correlated with the rate of rise, charge transfer within 1 ms, and action integral within 1 ms. Furthermore, when the total duration of the RS and any following continuing currents is longer than 40 ms, the peak current, charge transfer within 1 ms, and action integral within 1 ms of the RS are seldom greater than 25 kA, 2.6 C, and 15 × 10<sup>3</sup> A<sup>2</sup> s, respectively. It is indicated that TLFs containing RSs tend to have a longer duration but a smaller charge transfer during the IS than those without RS. The peak current of the RS is weakly correlated with its preceding silence period when there was no channel base current.</p>

PINK1 deficiency rewires early immune responses in a mouse model of Parkinson's disease triggered by intestinal infection

<p>Parkinson&rsquo;s disease is characterized by a period of non-motor symptoms, including gastrointestinal dysfunction, preceding motor deficits by decades. This long prodrome is suggestive of peripheral immunity involvement in the initiation of disease. We previously developed a model system in PINK1 KO mice displaying PD-like motor symptoms at late stages following intestinal infections. Herein, we map the initiating immune events at the site of infection in this model. Using single-cell RNAseq, we demonstrate that peripheral myeloid cells are the earliest highly dysregulated immune cell type in PINK1 KO infected mice followed by an aberrant T cell response shortly after. We elucidate an increased propensity for antigen presentation mediated by myeloid-CD8+ T cell interaction. PINK1 KO activated myeloid cells acquire a proinflammatory profile inducing cytotoxic T cell responses. Together, our study provides the first evidence that PINK1 is a key regulator of immune functions in the gut underlying early PD-related disease mechanisms.</p>

Serine-129 phosphorylation of a-synuclein is an activity-dependent trigger for physiologic protein-protein interactions and synaptic function

<p><strong>Phosphorylation of&nbsp;a-synuclein at the Serine-129 site (a-syn Ser129P) is an established pathologic hallmark of synucleinopathies and a therapeutic target. In physiologic states, only a small fraction of&nbsp;a-syn is phosphorylated at this site, and most studies have focused on the pathologic roles of this post-translational modification. We found that unlike wild-type (WT)&nbsp;a-syn which is widely expressed throughout the brain, the overall pattern of&nbsp;a-syn Ser129P is restricted, suggesting intrinsic regulation. Surprisingly, preventing Ser129P blocked activity-dependent synaptic attenuation by&nbsp;a-syn – thought to reflect its normal function. Exploring mechanisms, we found that neuronal activity augments Ser129P, which is a trigger for protein-protein interactions that are necessary for mediating&nbsp;a-syn function at the synapse. AlphaFold2-driven modeling and membrane-binding simulations suggest a scenario where Ser129P induces conformational changes that facilitate interactions with binding partners. Our experiments offer a new conceptual platform for investigating the role of Ser129 in synucleinopathies, with implications for drug-development.&nbsp;</strong></p>

Phytoplanktonic polysaccharide-mediated sedimentation of particulate organic carbon triggers lagged methane emissions

<p>Reservoirs act as carbon sinks when sedimentation of particulate organic carbon (POC) exceeds CO<sub>2</sub> and CH<sub>4 </sub>emissions. Here, we study the poorly explored process where phytoplankton-derived acidic polysaccharides (APs) aggregate into particulate organic matter, promoting carbon export to sediments. This source of particulate organic carbon (POC) in sediments can mineralize to CO<sub>2</sub> and CH<sub>4</sub> over various timescales. Our research, centered on a Mediterranean reservoir, elucidates phenological trends of APs and POC sedimentation and identifies their predominant drivers. Our findings present synchronic sedimentation patterns of POC and APs but identify a two-week delay between POC sedimentation and CH<sub>4 </sub>emissions. Despite its eutrophic status, our data demonstrate that this reservoir acts as a carbon sink by sequestering 4.33 g C m<sup>-2 </sup>y<sup>-1</sup>, which accentuates the importance of temporal integration at multiple scales when analyzing carbon budget within reservoirs.</p>
