Sarcomere length non-uniformities dictate force production along the descending limb of the force-length relation

<p>The force-length relation is one of the most defining features of the sliding filament theory of muscle contraction, and yet a topic of debate in the literature. The force-length relation predicts that the force produced by muscle fibers is directly proportional to the degree of overlap between myosin and actin filaments, but several studies have shown forces that are larger than predicted, especially at long sarcomere lengths. The reason for such discrepancies may be due to technical difficulties – studies have been conducted with muscle fibers, a preparation containing thousands of sarcomeres that makes accurate length measurements challanging. The aim of this study was to directly evaluate force production and sarcomere dynamics in isolated myofibrils and single sarcomeres to enhance our understanding about mechanisms governing deviations from the theoretically predicted force-length relation. Contractions at varying sarcomere lengths (SLs) along the plateau (SL=2.25m-2.39m) and the descending limb (SL&gt;2.39m) of the force-length relation were induced by solution switching, and different modes of force measurements were used. Our results show that single sarcomeres and extrapolated forces resemble the theoretical force-length relation closely, while myofibrils with sarcomere length dispersions show an extension of the plateau region and forces elevated above predicted levels. We also found an increase in sarcomere length non-uniformity and slowed rate of force production at long lengths in myofibrils with sarcomeres in series but not in single sarcomere preparations. We conclude that the deviation of the descending limb of the force-length relation obtained from plateau forces is correlated with the degree of sarcomere length non-uniformity and slowed force development.</p>

Data from: Non-uniform evolutionary response of gecko eye size to changes in diel activity patterns

Geckos feature a large range of eye sizes, but what drives this phenotypic diversity is currently unknown. Earlier studies point towards diel activity patterns (DAP) and locomotory mode, but phylogenetic comparative studies in support of the proposed adaptive mode of eye evolution are lacking. Here we test the hypothesis of DAP as the driver of eye size evolution with a dataset on 99 species of gecko. Results from phylogenetic generalized least square analysis (PGLS) and multivariate model-fitting reveal smaller eyes in diurnal geckos consistent with different phenotypic optima. However, Bayesian analyses of selective regime shifts demonstrate that only 2 of 9 transitions from nocturnal to diurnal activity are coupled with decreases in eye size, and 2 other regime shifts are not associated with DAP transitions. This non-uniform evolutionary response suggests that eye size is not the only functionally relevant variable. Evolutionary adaptations may therefore include different combinations of several traits (e.g., photoreceptors), all with the same functional outcome. Our results further demonstrate that DAP only partially explains eye size diversity in geckos. As open habitats favor the evolution of large eyes while obstructed habitats favor small eyes, the degree of habitat clutter emerges as another potential axis of eye diversification.

Data from: Predicted effects of climate factors on mountain species are not uniform over different spatial scales

The selection of relevant factors and appropriate spatial scale(s) is fundamental when modelling species response to climate change. We evaluated whether the effects of climate factors on species distribution/occurrence are consistently modelled over different spatial scales in birds, and used a two-scale approach to identify species-climate correlations unlikely to represent causal effects. We used passerine birds inhabiting mountain grassland in the Apennines (Italy) as a model. We surveyed four grassland species at 400 sampling points, and built habitat selection models (territory scale) and distribution models (7 algorithms, landscape scale). We compared the effect of climatic predictors on occurrence/distribution highlighted by models over to the two spatial scales, and with the effects supposed a priori based on the climatic niche of each species. Models at the territory level included at least one climatic predictor for three species; the observed effect of climatic predictors was seldom consistent with supposed effects. At the broadest scale, distribution models for all species included climatic predictors, with varying consistence with supposed effects and findings at the finer scale. Despite the importance of climate for species distribution, occurrence could be more directly related to other factors, with important implications for understanding/predicting the impacts of climate/environmental changes. Our approach revealed key variables for grassland birds, and highlighted the scale-dependent perceived importance of climate. At the local scale, climate effects were weak or hard to interpret. We found a general lack of consistence between supposed and observed effects at the territory level, and between landscape and territory models. Our results show the importance of predicting the potential effect of climatic factors prior to the analyses, carefully selecting ecologically meaningful variables and scales, and evaluating the nature and scale of climate-species links. We call for caution when predicting under future climates, especially when mechanistic effects and consistency across scales lack.

Data from: Genetic uniformity characterizes the invasive spread of water hyacinth (Eichhornia crassipes), a clonal aquatic plant

Aquatic plant invasions are often associated with long-distance dispersal of vegetative propagules and prolific clonal reproduction. These reproductive features combined with genetic bottlenecks have the potential to severely limit genetic diversity in invasive populations. To investigate this question we conducted a global scale population genetic survey using Amplified Fragment Length Polymorphism (AFLP) markers of the world's most successful aquatic plant invader – Eichhornia crassipes (water hyacinth). We sampled 1140 ramets from 54 populations from the native (South America) and introduced range (Asia, Africa, Europe, North America, Central America and the Caribbean). Although we detected 49 clones, introduced populations exhibited very low genetic diversity and little differentiation compared with those from the native range, and ~80% percent of introduced populations were composed of a single clone. A widespread clone ('W') detected in two Peruvian populations accounted for 70.9% of the individuals sampled and dominated in 74.5% of the introduced populations. However, samples from Bangladesh and Indonesia were composed of different genotypes, implicating multiple introductions to the introduced range. Nine of 47 introduced populations contained clonal diversity suggesting that sexual recruitment occurs in some invasive sites where environmental conditions favor seedling establishment. The global patterns of genetic diversity in E. crassipes likely result from severe genetic bottlenecks during colonization and prolific clonal propagation. The prevalence of the "W" genotype throughout the invasive range may be explained by stochastic sampling, or possibly because of pre-adaptation of the "W" genotype to tolerate low temperatures.

Data from: Phenotype uniformity in combined-stress environments has a different genetic architecture than in single-stress treatments

For crop production it is desirable for the mapping between genotype and phenotype to be consistent, such that an optimized genotype produces uniform sets of individual plants. Uniformity is strongly selected in breeding programs, usually automatically, as harvest equipment eliminates severely non-uniform individuals. Uniformity is genetically controlled, is known to be increased by interplant competition, and is predicted to increase upon abiotic stress. We mapped maize loci controlling genotype by environment interaction in plant height uniformity. These loci are different than the loci controlling mean plant height. Uniformity decreases upon combining two abiotic stresses, with alleles conferring greater uniformity in a single stress showing little improvement in a combined stress treatment. The maize B73 and Mo17 inbreds do not provide segregating alleles for improvement in plant height uniformity, suggesting that the genetic network specifying plant height has a past history of selection for robustness.

Data from ``Developments in Stochastic Coupled Cluster Theory: The initiator approximation and application to the Uniform Electron Gas''

<p>We describe further details of the Stochastic Coupled Cluster method and a diagnostic of such calculations, the shoulder height, akin to the plateau found in Full Configuration Interaction Quantum Monte Carlo. We describe an initiator modification to Stochastic Coupled Cluster Theory and show that initiator calculations can be extrapolated to the unbiased limit. &nbsp;We apply this method to the 3D 14-electron uniform electron gas and present complete basis set limit values of the CCSD and previously unattainable CCSDT correlation energies for up to $r_s=2$, showing a requirement to include triple excitations to accurately calculate energies at high densities.</p>

High synchrony of tree growth is followed by a uniform pattern of de-synchronization

<p><strong>Aim:&nbsp;</strong>Growth asynchrony in trees increases deviation in the average of forest growth and, consequently, brings uncertainty in modeling forest productivity and other functions. Despite recognition of growth variability among trees, the process of asynchronous growth and its ecological implications are poorly understood. Here we used tree-ring data to show that, although forest growth is influenced by climatic factors, inter-annual growth changes of individual trees often do not respond in a synchronous manner.</p><p><strong>Location:&nbsp;</strong>China.</p><p><strong>Time period:&nbsp;</strong>The past five and a half centuries.</p><p><strong>Major taxa studied:&nbsp;</strong>Forest tree growth.</p><p><strong>Methods:&nbsp;</strong>We used tree-ring methods and tree-ring data obtained from increment core samples in 1046 juniper trees at 32 sites and 538 pine trees at 20 sites to explore the implications of growth asynchrony in maintaining forest stability in the face of environmental fluctuations.</p><p><strong>Results:&nbsp;</strong>After highly synchronous growth events, the number of trees keeping continuous growth synchrony declined progressively and, notably, in a relatively uniform pattern.</p><p><strong>Main conclusions:&nbsp;</strong>We argue that diverse rather than synchronized growth spreads the capacity of trees to respond to disturbances and stresses at the individual level, consequently enhances forest resilience to risks of climate uncertainty. This resilience-acquisition strategy is an inherent and an adaptive property of forest tree growth that is similar to maintaining a diversified financial investment portfolio. This new perspective on widespread asynchronous tree growth sheds insight into assessment of forest health risks under future climate change.</p>

Bimaristan Nur Al Din, Pointcloud uniform

Lidar data, from open heritage. Uniform and clean point cloud. Over eight centuries old, the Bimaristan Nur al-Din, is the earliest surviving site for the development of Islamic medicine. Throughout the building's lifetime, the site has functioned as a medical school as well as a place of healing in the heart of the old city of Damascus. Built in 1154, the building is characterized by its red brick muqarnas, elaborate entrance block, and finely carved wooden doors. The hospital was in use until the 20th century when it was converted into a Museum of Arabic Medicine and Science. The preservation of the building is a reminder of the major contributions of Islamic medicine to the world today. HOMULA-RIR: A Room Impulse Response Dataset for Teleconferencing and Spatial Audio Applications Acquired Through Higher-Order Microphones and Uniform Linear Microphone Arrays

<p>In this paper, we present HOMULA-RIR, a dataset of room impulse responses (RIRs) acquired using both higher-order microphones (HOMs) and a uniform linear array (ULA), in order to model a remote attendance teleconferencing scenario. Specifically, measurements were performed in a seminar room, where a 64-microphone ULA was used as a multichannel audio acquisition system in the proximity of the speakers, while HOMs were used to model 25 attendees actually present in the seminar room. The HOMs cover a wide area of the room, making the dataset suitable also for applications of virtual acoustics. Through the measurement of the reverberation time and clarity index, and sample applications such as source localization and separation we demonstrate the effectiveness of the HOMULA-RIR dataset.</p>
