Data from: Non-industry sponsored preclinical studies on statins yield greater efficacy estimates than industry-sponsored studies: a meta-analysis
Industry-sponsored clinical drug studies are associated with publication of outcomes that favor the sponsor, even when controlling for potential bias in the methods used. However, the influence of sponsorship bias has not been examined in preclinical animal studies. We performed a meta-analysis of preclinical statin studies to determine whether industry sponsorship is associated with either increased effect sizes of efficacy outcomes and / or risks of bias in a cohort of published preclinical statin studies. We searched Medline (January 1966 - April 2012) and identified 63 studies evaluating the effects of statins on atherosclerosis outcomes in animals. Two coders independently extracted study design criteria aimed at reducing bias, results for all relevant outcomes, sponsorship source, and investigator financial ties. The I2 statistic was used to examine heterogeneity. We calculated the standardized mean difference (SMD) for each outcome and pooled data across studies to estimate the pooled average SMD using random effects models. In a priori subgroup analyses, we assessed statin efficacy by outcome measured, sponsorship source, presence or absence of financial conflict information, use of an optimal time window for outcome assessment, accounting for all animals, inclusion criteria, blinding, and randomization. The effect of statins was significantly larger for studies sponsored by non-industry sources (-1.99; 95% CI -2.68, -1.31) versus studies sponsored by industry (-0.73; 95% CI -1.00, -0.47) (p value < 0.001). Statin efficacy did not differ by disclosure of financial conflict information, use of an optimal time window for outcome assessment, accounting for all animals, inclusion criteria, blinding, and randomization. Possible reasons for the differences between non-industry and industry sponsored studies, such as selective reporting of outcomes, require further study.
Data from: Effects of riparian forest harvest on streams: a meta-analysis
1. Riparian forest harvesting impacts streams in many ways, from altering temperature regimes, shifting geomorphic structure, increasing sediment fluxes and affecting fish populations. However, we have noted considerable variation in the results between studies that led us to ask whether the effects of forest harvesting on streams were consistent between studies. We used meta-analysis of 34 replicated studies to address the effects of riparian logging on biological and chemical components of streams in contrast to control sites. 2. We found that the overall effect sizes of several response variables in replicated studies were significantly higher than zero, especially benthic invertebrates, and nitrogen and potassium concentrations. However, there was a very large amount of variation in the effect sizes between studies, and for many measures, the effect sizes from different studies were positive or negative, indicating site-specific responses. 3. We explored whether stream size, stream gradient and regional potential evapotranspiration could explain some of the effect size variation between studies. Relations with these environmental variables were weak, but suggestive that some of the context-specific, individual outcomes might be due to underlying environmental differences between sites. 4. Synthesis and applications. Despite relatively low numbers of replicated studies, we found significant overall effects of riparian forest harvesting although the magnitude and direction of responses within individual studies were site specific. This lack of consistency in the direction of effect sizes suggests we need a more context-dependent approach to the protection of freshwaters from forest management.
Data from: An investigation of the therapeutic effects of statins with metformin on polycystic ovary syndrome: a meta-analysis of randomized controlled trials
Objectives: To investigate the therapeutic effects of statins with metformin on polycystic ovary syndrome (PCOS). Settings: Endocrinology department. Participants: MEDLINE, EMBASE and Cochrane Central Register of Controlled Trials were searched until October 2014. Studies comparing statins and placebo, as well as the combination of statins and metformin and metformin alone, were included in the analysis. Interventions: Data were independently extracted by two researchers; any convergence was resolved by a third reviewer. Primary and secondary outcome measures: The following properties were extracted from the qualified trials to identify the effects of statins: clinical variables, metabolic characteristics, hormone outcomes, sign of inflammation, glucose parameters and insulin outcomes. Results: Data from four trials comparing statin and metformin with metformin alone were analysed. The combination of statins and metformin decreases the levels of C reactive protein (standardised mean difference (SMD) −0.91; 95% CI −1.81 to −0.02; p=0.046), triglyceride (SMD −1.37; 95% CI −2.46 to −0.28; p=0.014), total cholesterol (SMD −1.28; 95% CI −1.59 to −0.97; p=0.000) and low-density lipoprotein (LDL) cholesterol (SMD −0.74; 95% CI −1.03 to −0.44; p=0.000). However, the combined therapy fails to reduce fasting insulin (SMD −0.92; 95% CI −2.07 to 0.24; p=0.120), homeostasis model assessment of insulin resistance (SMD −1.15; 95% CI −3.36 to 1.06; p=0.309) and total testosterone (SMD −1.12; 95% CI −2.29 to 0.05; p=0.061). Analysis of the five trials comparing statin with placebo shows that statin monotherapy reduces LDL-cholesterol, triglyceride and total cholesterol. Conclusions: Combined statin and metformin therapy can improve lipid and inflammation parameters, but cannot effectively improve insulin sensitivity and reduce hyperandrogenism in women with PCOS. A large-scale randomised controlled study must be conducted to ascertain the long-term effects of the therapy.
Data from: Is there a temperate bias in our understanding of how climate change will alter plant-herbivore interactions? A meta-analysis of experimental studies
Climate change can drive major shifts in community composition and interactions between resident species. However, the magnitude of these changes depends on the type of interactions and the biome in which they take place. We review the existing conceptual framework for how climate change will influence tropical plant-herbivore interactions and formalize a similar framework for the temperate zone. We then conduct the first biome-specific tests of how plant-herbivore interactions change in response to climate-driven changes in temperature, precipitation, ambient CO2, and ozone. We used quantitative meta-analysis to compare predicted and observed changes in experimental studies. Empirical studies were heavily biased toward temperate systems, so testing predicted changes in tropical plant-herbivore interactions was virtually impossible. Furthermore, most studies investigated the effects of CO2 with limited plant and herbivore species. Irrespective of location, most studies manipulated only one climate change factor despite the fact that different factors can act in synergy to alter responses of plants and herbivores. Finally, studies of belowground plant-herbivore interactions were also rare; those conducted suggest that climate change could have major effects on belowground subsystems. Our results suggest that there is a disconnection between the growing literature proposing how climate change will influence plant-herbivore interactions and the studies testing these predictions. General conclusions will also be hampered without better integration of above- and belowground systems, assessing the effects of multiple climate change factors simultaneously, and using greater diversity of species in experiments.
Data from: Starvation effects on nitrogen and carbon stable isotopes of animals: an insight from meta-analysis of fasting experiments
Nitrogen and carbon stable isotopic compositions (δ15N and δ13C) of consumers have been used for physiological and food web studies. Previous studies have shown δ15N and δ13C values are affected by several biological and environmental factors during starvation, but the generality of the effect of starvation on δ15N and δ13C values has not yet been tested. Here, we performed a meta-analysis to evaluate the effects of starvation on δ15N and δ13C values of consumers, and the underlying factors that may explain the observed variation. The δ15N and δ13C values were calculated as the differences between the final δ15N and δ13C values of consumers (post-starvation) and the pre-starvation values on each experiment. Our meta-analysis showed a large variation in the δ15N and δ13C values of consumers (δ15N range: –0.82 to 4.30‰; mean: 0.47‰ and δ13C range: –1.92 to 2.62‰; mean: 0.01‰). The δ15N values of most consumers increased along the length of the starvation period and were influenced by nitrogen excretion and thermoregulation types, probably because differences in nitrogen metabolism and thermoregulation affect nitrogen processing and excretion rates. None of our predictor variables accounted for the variation in δ13C values, which showed both increases and decreases due to fasting. Our findings suggest that starvation results in changes in consumer δ15N values which are mainly explained by the length of the fasting period and by nitrogen and energy metabolism, but the underlying mechanisms of the starvation effects on δ13C values seem to be more complex than previously thought.

<p><b>Objective: </b>To conduct a comprehensive analysis of circulating metabolites and incident stroke in large prospective population-based settings.</p> <p><b>Methods: </b>We investigated the association of metabolites with risk of stroke in seven prospective cohort studies including 1,791 incident stroke events among 38,797 participants in whom circulating metabolites were measured by Nuclear Magnetic Resonance (<sup>1</sup>H-NMR) technology. The relationship between metabolites and stroke was assessed using Cox proportional hazards regression models. The analyses were performed considering all incident stroke events and ischemic and hemorrhagic events separately.</p> <p><b>Results: </b>The analyses revealed ten significant metabolite associations. Amino acid histidine (hazard ratio (HR) per standard deviation (SD) = 0.90, 95% confidence interval (CI): 0.85, 0.94; <i>P</i> = 4.45×10<sup>-5</sup>), glycolysis-related metabolite pyruvate (HR per SD = 1.09, 95% CI: 1.04, 1.14; <i>P</i> = 7.45×10<sup>-4</sup>), acute phase reaction marker glycoprotein acetyls (HR per SD = 1.09, 95% CI: 1.03, 1.15; <i>P</i> = 1.27×10<sup>-3</sup>), cholesterol in high-density lipoprotein (HDL) 2 and several other lipoprotein particles were associated with risk of stroke. When focusing on incident ischemic stroke, a significant association was observed with phenylalanine (HR per SD = 1.12, 95% CI: 1.05, 1.19; <i>P = </i>4.13×10<sup>-4</sup>) and total and free cholesterol in large HDL particles.</p> <p><b>Conclusions: </b>We found association of amino acids, glycolysis-related metabolites, acute phase reaction markers, and several lipoprotein subfractions with the risk of stroke. These findings support the potential of metabolomics to provide new insights into the metabolic changes preceding stroke.</p>

Coagulation abnormalities in dengue fever infection: a systematic review and meta-analysis
<p><b>Background:</b> Coagulation mechanisms are reported to be affected in dengue illness and evidenced by prolonged activated partial thromboplastin time (APTT) and prothrombin time (PT). The main aim of this systematic review and meta-analysis is to determine the magnitude of coagulation abnormalities among patients with dengue fever infection.</p> <p><b>Method:</b> This systematic review and meta-analysis were conducted per the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guideline. The Joana Brigg's Institute (JBI) critical appraisal checklist was used for quality appraisal. STATA version 11 software was used for meta-analysis. The magnitude of coagulation abnormalities among dengue fever patients was determined by using a random-effects model. Subgroup and sensitivity analysis were performed to investigate the possible source of heterogeneity. Egger weighted regression tests were used to check the presence of publication bias among the included articles.</p> <p><b>Result: </b>Forty-two studies with a total of 12,221 dengue fever patients were eligible for meta-analysis in this study. Of which 22, 15, and 26 studies were used to determine the magnitude of prolonged APTT, PT, and thrombocytopenia, respectively. The magnitude of prolonged APTT and PT among patients with dengue fever infection were 42.91% (95% CI: 30.95, 54.87) I<sup>2</sup> = 99.1% and 16.48% (95% CI: 10.95, 22.01) I<sup>2</sup>=97.0%, respectively. Besides, the magnitude of thrombocytopenia among dengue fever patients was 70.29% (95% CI: 62.69, 77.89) I<sup>2</sup>=99.3%. The magnitude of prolonged APTT in children and adults was 51.21% (95% CI: 24.54, 77.89) and 44.89% (95% CI: 28.32, 61.45), respectively. Similarly, the overall magnitude of prolonged PT in children and adults were 13.40% (95% CI: 6.09, 20.71) and 18.73% (95% CI: 7.49, 29.96), respectively.</p> <p><b>Conclusion: </b>The result of this study showed that there is a high magnitude of prolonged APTT and PT in dengue fever patients. Therefore, screening and early correction of coagulation abnormalities may be helpful to reduce further complications in those patients.</p>
The incidence and prevalence of drug resistant epilepsy: a systematic review and meta-analysis
<p><strong>Objective:</strong> In order to evaluate the incidence and prevalence of drug-resistant epilepsy (DRE) as well as its predictors and correlates, we conducted a systematic review and meta-analysis of observational studies.</p> <p><strong>Methods: </strong>Our protocol was registered with PROSPERO and the PRISMA and MOOSE reporting standards were followed. We searched MEDLINE, Embase, and Web of Science. We used a double arcsine transformation and random-effects models to carry out our meta-analyses. We performed random-effects meta-regressions using study-level data.</p> <p><strong>Results:</strong> Our search strategy identified 10,794 abstracts. Of these, 103 articles met our eligibility criteria. There was high inter-study heterogeneity and risk of bias. The cumulative incidence of DRE was 25.0 % (95% CI: 16.8, 34.3) in child studies but 14.6% (95% CI: 8.8, 21.6) in adult/mixed ages studies. The prevalence of DRE was 13.7% (95% CI: 9.2, 19.0) in population/community-based populations but 36.3% (95% CI: 30.4, 42.4) in clinic-based cohorts. Meta-regression confirmed that the prevalence of DRE was higher in clinic-based populations and in focal epilepsy. Multiple predictors and correlates of DRE were identified. The most reported of these were having a neurological deficit, an abnormal EEG, and symptomatic epilepsy. The most reported genetic predictors of DRE were polymorphisms of the ABCB1 gene.</p> <p><strong>Conclusions:</strong> Our observations provide a basis for estimating the incidence and prevalence of DRE, which vary between populations. We identified numerous putative DRE predictors and correlates. These findings are important to plan epilepsy services, including epilepsy surgery, a crucial treatment option for people with disabling seizures and DRE.</p>
Effectiveness of acute L-arginine supplementation on physical performance in strength training: a systematic review and meta-analysis
<p><span>The oral administration of L-arginine has been related to improved physical performance due to a likely reduction in muscle fatigue, resulting from the vasodilator effect of nitric oxide on skeletal muscle. However, there is no precise and quantitative analysis of the information in the literature. </span>Objective:<span> The main objective was to assess the effectiveness of L-arginine supplementation on physical performance in strength training with a systematic review and meta-analysis. Methods: Systematic review and meta-analysis, the PRISMA® (Preferred Reporting Items for Systematic Reviews and Meta-Analysis), PICOS statement and methodology guidelines, the quality of evidence by Classification of Assessment, Development and Assessment Recommendations (GRADE), Multiple Systematic Review Tool Assessment (AMSTAR2)</span><span>, and to assess the risk of bias of randomized clinical trials, the RoB 2.0 tool (Risk of Bias - Cochrane risk of bias tool revised for randomized trials)</span><span>. </span><span>The inclusion process established articles with well-designed human experiments that included only L-arginine supplementation testing the effects of L-arginine supplementation on sports performance related to strength training; identical experimental conditions in placebo or control group; and publications in the last ten years (until December 31, 2020). Three studies were included that compared L-arginine supplementation with placebo in anaerobic performance tests. </span>Results:<span> There was no significant heterogeneity (p>0.05) in the analysis of the three selected articles and the effects of L-arginine supplementation in muscular endurance; performance had a mean of 0.26 (95% CI = -0.129; 0.649; p = 0.190), peak torque with a mean of -0.002 (95% CI = -0.531; 0.527; p = 0. 99) of the third series of exercises and, furthermore, when comparing the integrated effect (resistance rate with the peak torque) there was no difference with a mean of 0.168 (95% CI = -0.145; 0.481; p = 0.292). </span></p> <p>Conclusions:<span> Acute L-arginine supplementation provides no ergogenic effect on strength training performance.</span></p>

Method: A total of 59 studies (19 before, 37 during-pandemic,<br> and 3 that included both) using. The Depression, Anxiety and Stress Scale (DASS) were included. A random<br> effects model estimated the means of NEs before and during the pandemic. Results: Studies from 47 countries<br> involving 193,337 participants were included. Globally, NEs increased during the pandemic, and<br> depression had the largest elevation. In Asia, depression and stress were elevated, whereas in Europe,<br> only depression increased, and in America, no differences in NEs between before and during the pandemic<br> were observed. The later time phase of the pandemic was associated with lower stress globally, and lower<br> stress and anxiety in Europe. Being younger was associated with more stress globally, and being older was<br> associated with higher anxiety in Asia. Students had higher anxiety globally and higher NEs in all three<br> aspects in Europe compared to the general population. The COVID-19 infection rate was associated with<br> more stress globally, and stress and anxiety in Europe. During the pandemic, females reported higher levels<br> of depression, anxiety, and stress compared to males, most pronounced in Europe. The impact of rising temperatures on the prevalence of coral diseases and its predictability: a global meta-analysis
<p>Coral reefs are under threat from disease as climate change alters environmental conditions. Rising temperatures exacerbate coral disease, but this relationship is likely complex as other factors also influence coral disease prevalence. To better understand this relationship, we meta-analytically examined 108 studies for changes in global coral disease over time alongside temperature, expressed using average summer sea surface temperature (SST) and cumulative heat stress as weekly sea surface temperature anomalies (WSSTAs). We found that both rising average summer SST and WSSTA were associated with global increases in the mean and variability in coral disease prevalence. Global coral disease prevalence tripled, reaching 9.92% in the 25 years examined, and the effect of ‘year’ became more stable (i.e., prevalence has lower variance over time), contrasting the effects of the two temperature stressors. Regional patterns diverged over time and differed in response to average summer SST. Our model predicted that, under the same trajectory, 76.8% of corals would be diseased globally by 2100, even assuming moderate average summer SST and WSSTA. These results highlight the need for urgent action to mitigate coral disease. Mitigating the impact of rising ocean temperatures on coral disease is a complex challenge requiring global discussion and further study.</p>
