Data from: The wheat curl mite Aceria tosichella (Acari: Eriophyoidea) is a complex of cryptic lineages with divergent host ranges: evidence from molecular and plant bioassay data

Aceria tosichella (the wheat curl mite, WCM) is a global pest of wheat and other cereals, causing losses by direct damage, as well as the transmission of plant viruses. The mite is considered to have an unusually wide host range for an eriophyoid species. The present study tested the commonly held assumption that WCM is a single, highly polyphagous species by assessing the host range of genetically distinct lineages of WCM occurring in Poland on different host plants. Genotyping was performed by analyzing nucleotide sequence data from fragments of the mitochondrial cytochrome c oxidase subunit I (COI) and the nuclear D2 region of 28S rDNA. Mean between-lineage distance estimated using COI data was found to be one order of magnitude greater than the within-clade lineage and, in some cases, comparable to distances between WCM lineages and a congeneric outgroup species. Host acceptance was tested by quantifying population growth for different WCM mitochondrial (mt)DNA lineages when transferred from source host plants to test plants. These experiments revealed significant differences in host colonization ability between mtDNA lineages, ranging from highly polyphagous to more host-specific. The present study reveals that WCM is composed of several discrete genetic lineages with divergent host-acceptance and specificity traits. Genetic variation for host acceptance within A. tosichella s.l. may act as a reproductive barrier between these lineages, most of which had narrow host ranges. Two lineages appear to have high pest potential on cereals, whereas several others appear to specialize on wild grass species. We conclude that WCM is not a homogeneous species comprising polyphagous panmictic populations rather it is a complex of genetically distinct lineages with variable host ranges and therefore variable pest potential.

Data from: Natural selection on gall size: variable contributions of individual host plants to population-wide patterns

Studies that provide estimates of the form and magnitude of selection on herbivore traits at the level of individual plants in natural populations represent a vital step in understanding the interaction of selection and gene flow among host-affiliated insect populations when individual plants equate to differing selective regimes. We analyzed phenotypic selection on the trait gall size for a host-specific gall former at both the individual host plant and population level (across host plants) in each of two years. Linear and nonlinear selection and the fitness function relating gall size to the probability of survivorship in the absence of natural enemies were estimated for each level and year. Selection imposed by the host plant was observed in 19 of the 22 subpopulations monitored. At the population level, linear and nonlinear selection were evident each year. However, population-level estimates masked the significant heterogeneity in the form and direction of selection evident among plants each year. Heterogeneity among gall-former subpopulations is emphasized by our findings that selection varied from directional to stabilizing among plants and the majority of selection gradients estimated for individual plants did not fall within the 95% CIs of the population-level estimates.

Data from: Using a comprehensive DNA barcode library to detect novel egg and larval host plant associations in a Cephaloleia Rolled-leaf Beetle (Coleoptera: Chrysomelidae)

To fully understand the ecology and evolution of plant-herbivore interactions, information regarding the life history of both immature and adult insect stages is essential. However, most knowledge of plant-herbivore associations is derived from observations of adults. One reason for this bias is that species identification of immature stages is usually challenging. DNA barcodes can be used to identify immature stages to the species-level. This technique compares short sequences of the appropriate DNA barcode loci (e.g., mitochondrial COI gene for insects) of an unidentified specimen to a known DNA barcode library. The accuracy of DNA-based identifications depends on the comprehensiveness of the DNA barcode library. We generated a comprehensive DNA barcode library for a community of Rolled-leaf Beetles (Coleoptera: Chrysomelidae) in a premontane tropical forest in Costa Rica. The DNA barcode COI accurately identified all beetle species included in this study. Using this DNA barcode library, we identified eggs and larvae of Cephaloleia histrionica Baly with 100% confidence. This new record of C. histrionica is unique in that this species completes its life cycle on a bromeliad, whereas most Cephaloleia species are associated with plants from the order Zingiberales. The life cycle, diet breadth, immature stages, and sexual dimorphism are described for C. histrionica.
