Dual GIP and GLP-1 receptor agonist tirzepatide improves beta-cell function and insulin sensitivity in type 2 diabetes
<p><span><b>Context:</b> Novel dual GIP and GLP-1 receptor agonist (RA) tirzepatide demonstrated substantially greater glucose control and weight loss (WL) compared with selective GLP-1RA dulaglutide.</span></p> <p><span><b>Objective:</b> Explore mechanisms of glucose control by tirzepatide. </span></p> <p><span><b>Design:</b> <i>Post-hoc</i> analyses of fasting biomarkers and multiple linear regression analysis.</span></p> <p><span><b>Setting:</b> 47 sites in 4 countries.</span></p> <p><span><b>Patients or Other Participants:</b> 316 subjects with Type 2 diabetes.</span></p> <p><span><b>Interventions:</b> Tirzepatide (1, 5, 10, 15 mg), dulaglutide (1.5 mg), placebo. </span></p> <p><span><b>Main Outcome Measures:</b> Analyze biomarkers of beta-cell function and insulin resistance (IR) and evaluate WL contributions to IR improvements at 26 weeks. </span></p> <p><span><b>Results:</b> HOMA2-B significantly increased with dulaglutide and tirzepatide 5, 10, and 15 mg compared with placebo (p<u><</u>0.02). Proinsulin/insulin and proinsulin/C-peptide ratios significantly decreased with tirzepatide 10 and 15 mg compared with placebo and dulaglutide (p<u><</u>0.007). Tirzepatide 10 and 15 mg significantly decreased fasting insulin (p<u><</u>0.033) and tirzepatide 10 mg significantly decreased HOMA2-IR (p=0.004) compared with placebo and dulaglutide. Markers of improved insulin sensitivity (IS) adiponectin, IGFBP-1, and IGFBP-2 significantly increased by one or more doses of tirzepatide (p<0.05). To determine whether improvements in IR were directly attributable to WL, multiple linear regression analysis with potential confounding variables age, sex, metformin, triglycerides, and HbA1c was conducted. WL significantly (p<u><</u>0.028) explained only 13% and 21% of improvement in HOMA2-IR with tirzepatide 10 and 15 mg, respectively. </span></p> <p><span><b>Conclusions:</b> Tirzepatide improved markers of IS and beta-cell function to a greater extent than dulaglutide. IS effects of tirzepatide were only partly attributable to WL, suggesting dual receptor agonism confers distinct mechanisms of glycemic control.</span></p>
Endocannabinoid signalling in stem cells and cerebral organoids drives differentiation to deep layer projection neurons via CB1 receptors
<p>The endocannabinoid (eCB) system, <i>via</i> cannabinoid CB<sub>1</sub> receptor, regulates neurodevelopment by controlling neural progenitor proliferation and neurogenesis. CB<sub>1</sub> receptor signalling <i>in vivo</i> drives corticofugal deep layer projection neuron development through the regulation of <span>BCL11B </span>and <span>Satb2</span> transcription factors. Here, we investigated the role of eCB signalling in mouse pluripotent embryonic stem cell-derived neuronal differentiation. Characterization of the eCB system revealed increased expression of eCB-metabolizing enzymes, eCB ligands and CB<sub>1</sub> receptors along neuronal differentiation. CB<sub>1</sub> receptor knockdown inhibited neuronal differentiation of deep layer neurons and increased upper layer neuron generation, and this phenotype was rescued by CB<sub>1</sub> re-expression. Pharmacological regulation with CB<sub>1</sub> receptor agonists or elevation of eCB tone with a monoacylglycerol lipase inhibitor promoted neuronal differentiation of deep layer neurons at the expense of upper layer neurons. Patch-clamp analyses revealed that enhancing cannabinoid signalling facilitated neuronal differentiation and functionality. Noteworthy, incubation with CB<sub>1</sub> receptor agonists during human iPSC-derived cerebral organoid formation also promoted the expansion of BCL11B<sup>+</sup> neurons. These findings unveil a cell-autonomous role of eCB signalling that, <i>via</i> CB<sub>1</sub> receptor, promotes mouse and human deep layer cortical neuron development.</p>
Data from: A public database of memory and naive B-cell receptor sequences
The vast diversity of B-cell receptors (BCR) and secreted antibodies enables the recognition of, and response to, a wide range of epitopes, but this diversity has also limited our understanding of humoral immunity. We present a public database of more than 37 million unique BCR sequences from three healthy adult donors that is many fold deeper than any existing resource, together with a set of online tools designed to facilitate the visualization and analysis of the annotated data. We estimate the clonal diversity of the naive and memory B-cell repertoires of healthy individuals, and provide a set of examples that illustrate the utility of the database, including several views of the basic properties of immunoglobulin heavy chain sequences, such as rearrangement length, subunit usage, and somatic hypermutation positions and dynamics.

Conformational changes in insulin receptor illuminated by live-cell linear dichroism imaging and a FLIP biosensor
<p>Response of GP2(1)-eGFP-CD59, a FLIP biosensor co transfected with insulin receptor against anatgonists and agonists of IR was measured and qunatified using polarization-resolved fluorescence microscopy. Linear dichoirism values were measured by analyinzing cells using already published imageJ macros (Bondar, Rybakova et al. 2021). This data set contains both raw images obtained from polarization micrscopy and analyzed data.</p>

AIRRSHIP: Example synthetic B cell receptor repertoire data
<p>Example repertoire data generated by AIRRSHIP (https://github.com/Cowanlab/airrship). Four repertoires are available (two with SHM, two without), each of which contains 100,000 sequences produced using the default AIRRSHIP parameters. Sequence data is contained in the FASTA files, TSV files give details of each step in the generation process, summary file shows the command given to AIRRSHIP and the locus file contains the alleles used in the repertoire. CD4 T cell receptor hierarchies are stable and independent of HIV-mediated dysregulation of immune homeostasis
<p>LT-ART and A5248 folders contain preprocessed CyTOF files (FCS format) from a 31-marker mass cytometry panel to examine all major PBMC lineages and specifically CD4 and CD8 T cell memory dynamics in people with HIV (PWH) who are durably ART suppressed for an average of 6.7 years (LT-ART, n = 10) and PWH in the first 500 days following ART initiation (A5248, n = 10). The panel also includes markers of activation (HLA-DR, CD38, CCR5), activation/exhaustion (PD-1), proliferation (Ki67), survival (Bcl-2) and long-lived memory (CD127).</p> <p>Preprocessed annotated data objects (A5248_subsample.h5ad, LT-ART_subsample.h5ad) for unsupervised analysis can be accessed using the 'read_h5ad' function in Scanpy.</p> <p>CD4_MSI and CD8_MSI folders contain the MSI data for the Gamma fixed-effects regression models. </p> <p>All source code for reproduction of the results can be found in the GitHub repository: <a href="https://github.com/glab-hiv/immune-recovery">https://github.com/glab-hiv/immune-recovery</a></p>
IL-3 receptor signaling suppresses chronic intestinal inflammation by controlling mechanobiology and tissue egress of regulatory T cells
<p>RNA from unchallenged mouse <em>Il3r</em><sup>-/-</sup> and <em>Il3r</em><sup>+/+</sup> thymocytes was isolated with the NucleoSpin RNA Kit for RNA isolation (Machery Nagel). RNA-Analysis and sequencing was performed by the Core unit next generation sequencing of the University Hospital Erlangen. The quality of isolated RNA was determined using an Agilent 2100 bioanalyzer with RNA 6000 Nano kit and related software (Agilent). Sequencing libraries were generated from RNA samples using the TruSeq Stranded mRNA Kit (Illumina). Libraries were sequenced on a HiSeq 2500 platform (Illumina). The reads were aligned with Star v2.5.4b against the GRCm38 reference genome. Reads were counted with featureCounts of the Rsubread package v2.8.1.</p>
