Assessing the use of bison for savanna restoration at Cedar Creek Ecosystem Science Reserve: Consumption
Oak savanna is the most threatened ecosystem in Minnesota and fire, alone, is not restoring and preserving it. Our savanna restoration research started more than a half century ago in what had once been native savanna at Cedar Creek. It has shown that burning about 4 to 7 times per decade eliminates shrubs and non-savanna tree species and restores prairie grassland species. However, our 50 years of research is also showing that these frequent and intense fires are preventing oaks from regenerating. Bison are now known to be a keystone species for restoring and preserving grasslands, but their roles in savanna ecosystems remain unknown. In grasslands, bison preferentially graze the dominant warm season grasses that would otherwise outcompete wildflowers, thereby promoting plant coexistence and enhancing plant diversity. Here we propose to test whether bison grazing might promote the growth and survivorship of oak seedlings in burned savannas by reducing grass fuel for fires and by knocking back dominant grass competitors. We will maintain the existing fire frequencies and the design of the long-term burning experiment, while adding bison grazing as an additional factor in part of several burn units on the southeast side of the property. Bison will graze during the summer and early fall seasons. Grazing exclosures will be established, and oak seedlings will be planted, to test effects of bison grazing on early oak growth and survivorship. The outcomes we plan to achieve are to: (1) discover better restoration and preservation practices for savanna ecosystems; (2) determine how these practices impact savanna biodiversity; and (3) educate Minnesotans about the ecological heritage of their state, including the roles that bison, fire and biodiversity play in the functioning of savannas and other Minnesota ecosystems. Data from: Residual eDNA detection sensitivity assessed by quantitative real-time PCR in a river ecosystem
Several studies have demonstrated that environmental DNA (eDNA) can be used to detect the presence of aquatic species, days to weeks after the target species has been removed. However, most studies used eDNA analysis in lentic systems (ponds or lakes), or in controlled laboratory experiments. While eDNA degrades rapidly in all aquatic systems, it also undergoes dilution effects and physical destruction in flowing systems, complicating detection in rivers. However, some eDNA (i.e. residual eDNA) can be retained in aquatic systems, even those subject to high flow regimes. Our goal was to determine residual eDNA detection sensitivity using quantitative real-time polymerase chain reaction (qRT-PCR), in a flowing, uncontrolled river after the eDNA source was removed from the system; we repeated the experiment over two years. Residual eDNA had the strongest signal strength at the original source site and was detectable there up to 11.5 hours after eDNA source removal. Residual eDNA signal strength decreased as sampling distance downstream from the eDNA source site increased, and was no longer detectable at the source site 48 hours after the eDNA source water was exhausted in both experiments. This experiment shows that residual eDNA sampled in surface water can be mapped quantitatively using qRT-PCR, which allows a more accurate spatial identification of the target species location in lotic systems, and relative residual eDNA signal strength may allow the determination of the timing of the presence of target species.

Supporting Data for: Assessing the potential of amino acid δ13C and δ15N analysis in terrestrial and freshwater ecosystems
<p>Understanding the structure and dynamics of food webs requires accurate estimates of energy flow among organisms. Bulk tissue carbon (<em>δ</em><sup>13</sup>C) and nitrogen (<em>δ</em><sup>15</sup>N) isotope analysis is often used to this end, however, the limitations of this technique can outweigh the benefits. The isotope analysis of individual amino acids is being increasingly employed to trace energy flow and estimate consumer trophic level. Central to this compound-specific approach are the concepts of essential amino acid (AA<sub>ESS</sub>) <em>δ</em><sup>13</sup>C fingerprinting and amino acid (AA) <em>δ</em><sup>15</sup>N beta-values, both of which have been understudied and are poorly constrained in terrestrial and freshwater producers.</p> <p>We present AA<sub>ESS</sub> <em>δ</em><sup>13</sup>C data for 112 terrestrial and freshwater producers collected from two aridland habitats in the northern Chihuahuan Desert (New Mexico, USA) and AA <em>δ</em><sup>15</sup>N data for a subset (n=28) of these samples. We characterized AA<sub>ESS</sub> <em>δ</em><sup>13</sup>C fingerprints by performing linear discriminant analysis on the <em>δ</em><sup>13</sup>C values of isoleucine, leucine, lysine, phenylalanine, threonine, and valine for four producer groups – C<sub>3</sub> plants, C<sub>4</sub> plants, CAM plants, and filamentous green algae. We explored potential biochemical mechanisms underlying these AA<sub>ESS</sub> <em>δ</em><sup>13</sup>C fingerprints by calculating differences between the <em>δ</em><sup>13</sup>C values of AA<sub>ESS</sub> products and their AA precursors. This allowed us to estimate and compare isotopic discrimination for specific AA<sub>ESS</sub> synthesis pathways across producer groups.</p> <p>We found near perfect separation of AA<sub>ESS</sub> <em>δ</em><sup>13</sup>C fingerprints among producer groups; all groups reclassified with >95% success within our multivariate framework. We also found varied isotopic discrimination for specific AA<sub>ESS</sub> synthesis pathways among producer groups. Contrary to previous studies, we found no differences in beta-values between terrestrial C<sub>3</sub> and C<sub>4</sub> plants for any trophic-source AA pairing. Furthermore, we found that Lys <em>δ</em><sup>15</sup>N values were less variable and more closely related to bulk tissue <em>δ</em><sup>15</sup>N values than Phe <em>δ</em><sup>15</sup>N values in terrestrial and freshwater producers.</p> <p><span><span><span><span><span>We conclude that AA<sub>ESS</sub> <em>δ</em></span></span></span></span></span><sup>13</sup><span><span><span><span><span>C fingerprints are a higher-resolution tracer for freshwater food webs where instream algae have overlapping bulk tissue <em>δ</em></span></span></span></span></span><sup>13</sup><span><span><span><span><span>C values with terrestrial C<sub>3</sub> plants.
