Data from: Regulation of HBEGF by micro-RNA for survival of developing human trophoblast cells
Introduction: The growth factor HBEGF is upregulated post-transcriptionally in the low O2 environment of the human placenta during the first 10 weeks of pregnancy. We have examined the possible roles of HBEGF turnover and micro-RNA (miRNA) in its regulation by O2 in human first trimester trophoblast. Methods: HTR-8/SVneo trophoblast cells were cultured at 2% or 20% O2. The cells were transfected with a dual luciferase reporter construct (psiCHECK-2) containing no insert (control), the HBEGF 3' untranslated region (3'UTR), or sub-regions of the 3'UTR, as well as with siRNA for DGCR8. RNA was extracted from trophoblast cells cultured at 2% O2 for 0–4 h for next-generation sequencing. HBEGF was quantified by ELISA. HBEGF, DGCR8, and β–actin were examined by western blotting. Results: Protein turnover studies, using 10 μg/ml cyclohexamide, 1 μg/ml lactocystin, or 100 μg/ml MG132, demonstrated faster HBEGF degradation at 20% O2 than 2% O2, mediated by the proteasome. However, proteasome inhibition failed to initiate HBEGF accumulation at 20% O2. Reporter assays, comparing to empty vector, demonstrated that the intact HBEGF 3' UTR inhibited expression (0.26), while fragments containing only its flanking regions increased reporter activity (3.15; 3.43). No differential expression of miRNAs was found in trophoblast cells cultured at 2% and 20% O2. Nevertheless, HBEGF upregulation at 2% O2 was blocked when the miRNA-processing protein DGCR8 was silenced, suggesting a role for miRNA. Conclusion: Our findings suggest involvement of flanking regions of the 3'UTR in activating HBEGF protein synthesis in response to 2% O2, possibly through a miRNA-mediated mechanism.

Non-human primate white matter development during the first year of life as assessed by diffusion tensor imaging and quantitative relaxometry
<p>In this project, we use diffusion tensor imaging (DTI) and quantitative relaxometry (QR) metrics to assess longitudinal changes in white matter (WM) throughout the postnatal rhesus macaque brain across the first year of life. DTI and QR metrics are measured at 3, 7, 13, 25, and 53 weeks of age in each of 35 non-human primates (NHPs) in this sample, for a total 175 datapoints (i.e., scans). Leveraging a previously published NHP WM atlas (Adluru et al., 2012; Zakszewski et al., 2014), we assess WM microstructure in 76 WM regions across the brain. DTI metrics include fractional anisotropy (FA), mean diffusivity (MD), radial diffusivity (RD), and axial diffusivity (AD); the QR metric assessed is the longitudinal relaxation rate (qR1). These longitudinal data enable the direct comparison of DTI and QR metrics in very early life on a within-subject level, as well as robust, multimodal modeling of WM growth in infancy, providing insights into early postnatal neurodevelopment that may have important implications neuropsychiatric and neurodevelopmental disorders in childhood.</p>

Transcription profiling of human peripheral blood to development gene expression signatures for practical radiation biodosimetry
To further development of our gene expression approach to biodosimetry we have employed whole genome microarray expression profiling as a discovery platform to identify genes with the potential to distinguish radiation dose across an exposure range relevant for medical decision-making in a radiological emergency. Human peripheral blood from healthy donors was irradiated ex vivo and a 74-gene consensus signature was identified that distinguished between four radiation doses (0.5 2 5 and 8 Gy) and control samples. The same set of genes separated samples by exposure level at both six and 24 hours after treatment with overlap evident only at the highest two doses (5 and 8 Gy). Expression of five genes (CDKN1A FDXR SESN1 BBC3 and PHPT1) from this signature was quantified in the same RNA samples by real-time PCR confirming low variability between donors as well as the predicted radiation response pattern. Experiment Overall Design: Radiation induced gene expression in human blood was measured at 6 and 24 hours after exposure to doses of 0 0.5 2 5 and 8 Gy g-rays. Five independent experiments were performed at each time (6 or 24 hours) using different donors for each experiment
