Supplementary Tables (Mapping the Associations of the Plasma Lipidome With Insulin Resistance and Response to an Oral Glucose Tolerance Test)

Context: Insulin resistance (IR) remained a global health challenge. Lipidomics offers a unique opportunity to identify biomarkers; a key step towards understanding mechanisms of IR associated with abnormal lipid metabolism.

Objective: To determine whether plasma lipid species are associated with indices of insulin resistance and to evaluate the plasma lipidome response to an OGTT.

Design and Setting: Community based cross-sectional

Participants and sample: Plasma samples (collected at 0 and 120 min during an OGTT) from non-obese, young adults aged between 18-34 years (n = 246) were analysed using liquid chromatography tandem mass spectrometry.

Main outcome measures: Using linear models (controlling for age, sex, BMI, total cholesterol, HDL-cholesterol and triglycerides), the associations between indices of insulin resistance and individual lipid species or changes in lipid levels during an OGTT were tested.

Results: Some (213) and (199) lipid species associate with HOMA-IR and insulin AUC respectively. Alkylphosphatidylcholine (10), alkenylphosphatidylcholine (23) and alkylphosphatidylethanolamine (6) species were associated with insulin AUC in men only. Species of phosphatidylcholine (7) and sphingomyelin (5) were associated in women only. In response to an oral glucose tolerance test, a perturbation in the plasma lipidome, particularly in acylcarnitine species was observed; and the changes in many lipid species were associated with insulin AUC.

Conclusions: The plasma lipidome and changes in lipid levels during an OGTT were associated with indices of insulin resistance. These findings underlie the involvement of molecular lipid species in the pathogenesis of insulin resistance and possibly crosstalk between insulin resistance and sex-specific regulation of lipid metabolism.

Data from: Comparative transcriptome and lipidome analyses reveal molecular chilling responses in chilling-tolerant sorghums
Chilling temperatures (0 to 15°C) are a major constraint for temperate cultivation of tropical-origin crops, including the cereal crop sorghum (Sorghum bicolor [L.] Moench). Northern Chinese sorghums have adapted to early-season chilling, but molecular mechanisms of chilling tolerance are unknown. We used RNA sequencing of seedlings to compare the chilling-responsive transcriptomes of a chilling-tolerant Chinese accession with a chilling-sensitive US reference line, and mass spectrometry to compare chilling-responsive lipidomes of four chilling-tolerant Chinese accessions with two US reference lines. Comparative transcriptomics revealed chilling-induced up-regulation of cold-response regulator C-repeat binding factor (CBF) transcription factor and genes involved in reactive oxygen detoxification, jasmonic acid (JA) biosynthesis, and lipid remodeling phospholipase Dα1 (PLDα1) gene in the chilling-tolerant Chinese line. Lipidomics revealed conserved chilling-induced increases in lipid unsaturation, as well as lipid remodeling of photosynthetic membranes that is specific to chilling-tolerant Chinese accessions. Our results point to CBF-mediated transcriptional regulation, galactolipid and phospholipid remodeling, and JA as potential molecular mechanisms underlying chilling adaptation in Chinese sorghums. These molecular systems underlying chilling response could be targeted in molecular breeding for chilling tolerance.

A deep dive into fat: Investigating blubber lipidomics fingerprint of killer whales and humpback whales in northern Norway

In cetaceans, blubber is the primary and largest lipid body reservoir. Our current understanding about lipid stores and uses in cetaceans is still limited and most studies only focused on a single narrow snapshot of the lipidome. We documented an extended lipidomics fingerprint in two cetacean species present in northern Norway during wintertime. We were able to detect 817 molecular lipid species in blubber of killer whales (Orcinus orca) and humpback whales (Megaptera novaeangliae). The profiles were largely dominated by triradylglycerols in both species and to a lesser extent, by other constituents including glycerophosphocholines, phosphosphingolipids, glycerophosphoethanolamines and diradylglycerols. Through a unique combination of traditional statistical approaches, together with a novel bioinformatics tool (LION/web), we showed contrasting fingerprints composition between species. The higher content of triradylglycerols in humpback whales is necessary to fuel their upcoming half a year fasting and energy-demanding migration between feeding and breeding grounds. In adipocytes, we assume that the intense feeding rate of humpback whales prior to migration translates into an important accumulation of triacylglycerols content in lipid droplets. Upstream, the endoplasmic reticulum is operating at full capacity to supply acute lipid storage, consistent with the reported enrichment of glycerophosphocholines in humpback whales, major components of the endoplasmic reticulum. There was also an enrichment of membrane components which translates into higher sphingolipids content in the lipidome of killer whales, potentially as a structural adaptation for their higher hydrodynamic performance. Finally, the presence of both lipid-enriched and lipid-depleted individuals within the killer whale population in Norway suggests dietary-specialization, consistent with significant differences of δ15N and δ13C isotopic ratios in skin between the two groups, with higher values and a wider niche for the lipid-enriched individuals. Results suggest the lipid-depleted killer whales were herring-specialists, while the lipid-enriched individuals might feed on both herrings and seals.

A liposome assay showing the absence of scrambling in TTYH proteins and a lipidomics analysis of TTYH2

The Tweety homologues (TTYHs) are members of a conserved family of eukaryotic membrane proteins that are abundant in the brain. The three human paralogs were assigned to function as anion channels that are either activated by Ca2+ or cell swelling. To uncover their unknown architecture and its relationship to function, we have determined the structures of human TTYH1–3 by cryo-electron microscopy. All structures display equivalent features of a dimeric membrane protein that contains five transmembrane segments and an extended extracellular domain. As none of the proteins shows features reminiscent of an anion channel, we revisited functional experiments and did not find any indication of ion conduction. Instead, we find density in an extended hydrophobic pocket contained in the extracellular domain that emerges from the lipid bilayer, which suggests a role of TTYH proteins in the interaction with lipid-like compounds residing in the membrane.
