Data from: Sex-specific associations between life history traits and a novel reproductive polymorphism in the Pacific field cricket
<p>Associations between heritable polymorphisms and life-history traits, such as development time or reproductive investment, may play an underappreciated role in maintaining polymorphic systems. This is because selection acting on a particular morph could be bolstered or disrupted by correlated changes in life-history or vice versa. In a Hawaiian population of the Pacific field cricket (<em>Teleogryllus oceanicus</em>), a novel mutation (flatwing) on the X-chromosome is responsible for a heritable polymorphism in male wing structure. We used laboratory cricket colonies fixed for male wing morph to investigate whether males and females bearing the flatwing or normal-wing (wild-type) allele differed in their life-history traits. We found that flatwing males developed faster and had heavier testes than normal-wings, whereas flatwing homozygous females developed slower and had lighter reproductive tissues than normal-wing homozygous females. Our results advance our understanding of the evolution of polymorphisms by demonstrating that the genetic change responsible for a reproductive polymorphism can also have consequences for fundamental life-history traits in both males and females.</p>
Data from: Social associations between California sea lions influence the use of a novel foraging ground
Social relationships define an individual's position in its social network, which can influence the acquisition and spread of information and behavioural variants through the population. Thus, when nuisance behaviours spread through wildlife populations, identifying central individuals may provide valuable insights for problem-species management. We studied the effects of network position on California sea lion (Zalophus californianus) discovery and foraging success at a novel foraging ground—the salmonids that aggregate at the Bonneville Dam tail-race, 235 km up the Columbia River. We found that an individual's centrality in their social network influenced discovery of the Bonneville Dam and whether they returned the next year. Foraging success once at the dam was independent of network position. Extensive lethal and non-lethal removal efforts have been implemented at Bonneville Dam and focused on reducing the number of individual sea lions at the dam. Since social relationships forged at the opening of the Columbia River influence both the discovery and return to the Bonneville Dam, efforts to increase salmon recovery may be enhanced by breaking apart social networks at the opening of the river.
Data from: Finding flies in the mushroom soup: host specificity of fungus-associated communities revisited with a novel molecular method
Fruiting bodies of fungi constitute an important resource for thousands of other taxa. The structure of these diverse assemblages has traditionally been studied with labour-intensive methods involving cultivation and morphology-based species identification, to which molecular information might offer convenient complements. To overcome challenges in DNA extraction and PCR associated with the complex chemical properties fruiting bodies, we developed a pipeline applicable for extracting amplifiable total DNA from soft fungal samples of any size. Our protocol purifies DNA in two sequential steps: (1) initial salt-isopropanol extraction of all nucleic acids in the sample is followed by (2) an extra clean-up step using solid-phase reversible immobilization (SPRI) magnetic beads. The protocol proved highly efficient, with practically all of our samples — regardless of biomass or other properties — being successfully PCR amplified using metabarcoding primers and subsequently sequenced. As a proof-of-concept, we apply our methods to address a topical ecological question: is host specificity a major characteristic of fungus-associated communities, i.e., do different fungus species harbour different communities of associated organisms? Based on an analysis of 312 fungal fruiting bodies representing ten species in five genera from three orders, we show that molecular methods are suitable for studying this rich natural microcosm. Comparing to previous knowledge based on rearing and morphology-based identifications, we find a species-rich assemblage characterized by a low degree of host specialization. Our method opens up new horizons for molecular analyses of fungus-associated interaction webs and communities.

Streptococcus pneumoniae is a major human pathogen, and a leading cause of disease and death worldwide. Pneumococcal invasive disease is triggered by initial asymptomatic colonization of the human upper respiratory tract. The pneumococcal serine-rich repeat protein (PsrP) is a lung-specific virulence factor whose functional binding region (BR) binds to keratin-10 (KRT10) and promotes pneumococcal biofilm formation through self-oligomerization. We present the crystal structure of the KRT10-binding domain of PsrP (BR187–385) determined to 2.0 Å resolution. BR187–385 adopts a novel variant of the DEv-IgG fold, typical for microbial surface components recognizing adhesive matrix molecules adhesins, despite very low sequence identity. An extended β-sheet on one side of the compressed, two-sided barrel presents a basic groove that possibly binds to the acidic helical rod domain of KRT10. Our study also demonstrates the importance of the other side of the barrel, formed by extensive well-ordered loops and stabilized by short β-strands, for interaction with KRT10.
