Data from: Non-industry sponsored preclinical studies on statins yield greater efficacy estimates than industry-sponsored studies: a meta-analysis

Industry-sponsored clinical drug studies are associated with publication of outcomes that favor the sponsor, even when controlling for potential bias in the methods used. However, the influence of sponsorship bias has not been examined in preclinical animal studies. We performed a meta-analysis of preclinical statin studies to determine whether industry sponsorship is associated with either increased effect sizes of efficacy outcomes and / or risks of bias in a cohort of published preclinical statin studies. We searched Medline (January 1966 - April 2012) and identified 63 studies evaluating the effects of statins on atherosclerosis outcomes in animals. Two coders independently extracted study design criteria aimed at reducing bias, results for all relevant outcomes, sponsorship source, and investigator financial ties. The I2 statistic was used to examine heterogeneity. We calculated the standardized mean difference (SMD) for each outcome and pooled data across studies to estimate the pooled average SMD using random effects models. In a priori subgroup analyses, we assessed statin efficacy by outcome measured, sponsorship source, presence or absence of financial conflict information, use of an optimal time window for outcome assessment, accounting for all animals, inclusion criteria, blinding, and randomization. The effect of statins was significantly larger for studies sponsored by non-industry sources (-1.99; 95% CI -2.68, -1.31) versus studies sponsored by industry (-0.73; 95% CI -1.00, -0.47) (p value < 0.001). Statin efficacy did not differ by disclosure of financial conflict information, use of an optimal time window for outcome assessment, accounting for all animals, inclusion criteria, blinding, and randomization. Possible reasons for the differences between non-industry and industry sponsored studies, such as selective reporting of outcomes, require further study.

Development of preclinical diagnostic tools of hepatobiliary tumors associated with Opisthorchis felineus infection in population of endemic regions

<p>According to the official medical statistics, the incidence of hepatobiliary tumors (including cholangiocarcinoma) in the Tomsk region was 9.31 cases per 100,000 population in 2011-2013, which was twice the average in Russia [Fedorova OS, Kovshirina Yu.V., et al., 2017]. Cholangiocarcinoma is a group of intra- and / or extrahepatic bile ducts cancer with high mortality due to its aggressiveness, lack of specific symptoms and / or prolonged asymptomatic course and resistance to therapy [Bridgewater J, Galle PR, Khan SA et al., 2014]. One of the significant risk factors for liver cancer is the high prevalence of parasitic infection caused by liver fluke O. felineus in Western Siberia, whose carcinogenic potential has been demonstrated in a number of experimental studies in animal models of the disease, as well as in research on autopsy material, surgical data [Gouveia MJ, Pakharukova MY, et al., 2017; Brazhnikov N.A. et al., 2002; Maksimova GA, Pakharukova MY, 2017; Bychkov V.G., 2003; Kovshirina YV, Fedorova OS, Kovshirina AE et al, in press, 2018]. In this regard, it is necessary to develop diagnostic tools allowed timely detection of severe hepatobiliary diseases (including cancer) associated with opisthorchiasis in order to jugulate it and improve the prognosis for patients in endemic regions. In this way, the aim of this project is creation a diagnostic tools for early preclinical diagnostics of the hepatobiliary tumors (cholangiocarcinoma) in patients with chronic O. felineus infection in endemic regions. For these aims a comprehensive fundamental research will be performed, including evaluation of biochemical measures (lactate, alanine aminotransferase, aspartate aminotransferase, gamma-glutamyl transferase, alkaline phosphatase, glucose, direct and indirect bilirubin, high and low density lipoproteins, triacylglycers, as well as low-density lipotropes. Immunological parameters (TGFb) and tumor markers (CA 19-9, СYFRA 21-1) in serum samples obtained from patients with O. felineus infection having periductal fibrosis and without it, patients with cholangiocarcinoma and individuals without opisthorchiasis and bile ducts cancer. Statistical analysis was carried out using the programming language R version 3.6.1. The study results indicate that ultrasound of the liver and biliary system with the assessment of opistorchosis-associated periductal fibrosis together with the determination of biochemical indicators (alkaline phosphatase, direct bilirubin, AST, glucose), as well as oncomarker CA 19-9, as predictors of fibrosis, can be used as an algorithm for early preclinical diagnosis of malignant tumors of the hepatobiliary system (cholangiocarcinoma) against the background of chronic infection O. felineus, in the population of endemic regions.</p>

Synthesis and preclinical evaluation of a novel fluorine-18 labeled small-molecule PET radiotracer for imaging of CXCR3 receptor in mouse models of atherosclerosis

<p>Background CXCR3 is a chemokine receptor and is expressed in innate and adaptive immune cells. It promotes<br> the recruitment of T-lymphocytes and other immune cells to the inflammatory site in response to the binding of cognate<br> chemokines. Upregulation of CXCR3 and its chemokines has been found during atherosclerotic lesion formation.<br> Therefore, detection of CXCR3 by positron emission tomography (PET) radiotracer can be a useful tool for detecting the development of atherosclerosis in a noninvasive manner. Herein, we report the synthesis, radiosynthesis, and characterization of a novel fluorine-18 (F-18, 18F) labeled small-molecule radiotracer for the imaging of the CXCR3 receptor in mouse models of atherosclerosis.<br> Results The reference standard <strong>1</strong> and its precursor <strong>9</strong> were synthesized over 5 steps from starting materials in good to moderate yields. The measured Ki values of CXCR3A and CXCR3B were 0.81 &plusmn; 0.02 nM and 0.31 &plusmn; 0.02 nM, respectively. [18F]<strong>1</strong> was prepared by a two-step radiosynthesis with a decay-corrected radiochemical yield of 13 &plusmn; 2%, radiochemical purity &gt; 99%, and specific activity of 44.4 &plusmn; 3.7 GBq/&mu;mol at the end of synthesis (n = 6). The baseline studies showed that [18F]<strong>1</strong> displayed high uptake in the atherosclerotic aorta and brown adipose tissue in Apolipoprotein E (ApoE) knockout (KO) mice fed with a high-fat diet over 12 weeks. The uptake of [18F]<strong>1</strong> in these regions was reduced significantly in self-blocking studies, demonstrating CXCR3 binding specificity. Contrary to this, no significant differences in uptake of [18F]<strong>1</strong> in the abdominal aorta of C57BL/6 control mice fed with a normal diet were observed in both baseline and blocking studies, indicating increased CXCR3 expression in atherosclerotic lesions. Immunohistochemistry studies demonstrated that [18F]<strong>1</strong>-positive regions were correlated with CXCR3 expression, but some<br> atherosclerotic plaques with significant size were not detected by [18F]<strong>1</strong>, and their CXCR3 expressions were minimal.<br> Conclusion [18F]<strong>1</strong> was synthesized with good radiochemical yield and high radiochemical purity. In PET imaging<br> studies, [18F]<strong>1</strong> displayed CXCR3-specific uptake in the atherosclerotic aorta in ApoE KO mice. [18F]<strong>1</strong> visualized CXCR3<br> expression in different regions in mice aligned with the tissue histology studies. Taken together, [18F]<strong>1</strong> is a potential<br> PET radiotracer for imaging CXCR3 in atherosclerosis.</p>
