Dataset - PONE-D-20-09507 - Usefulness of circulating miR-146a and miR-16-5p microRNAs as prognostic biomarkers in community-acquired pneumonia
<p>This dataset shows a prospective observational study performed in a cohort of 153 patients admitted to hospital with CAP.</p> <p>Clinical and analytical variables were collected, and the main outcome variable was 30-day mortality.</p> <p>Small RNA was purified from patients´ plasma samples by column-based protocol, and retrotranscribed to cDNA (Exiqon's miRCURY ™ series kits), adding synthetic RNA controls (spike-in). The quality of the process was evaluated (QC control) and only 117 samples passed the test.</p> <p>FIRST STEP: Eight samples paired by age and gender were selected (4 patients who had suffered a cardiovascular event or death during follow-up and 4 who had not) and a panel of 752 human miRNAs was tested (miRCURY LNA ™ Universal - Ready-to-Use Human Panel , Exiqon), in order to determine a preliminary pattern of differential miRNA expression between patients with different CAP evolution.</p> <p>SECOND STEP: According to the preliminary data obtained, 25 candidate miRNAs were selected: 5 intended to be used as normalizers, 5 selected by statistical criteria (univariate association with mortality) and 15 selected from an exhaustive bibliographic search on miRNAs, sepsis, inflammation and / or cardiovascular disease, prioritizing those that appeared in a greater number of publications and those related to respiratory diseases. RT-PCR was carried out by hybridization with double-stranded flurochrome (ExiLENT SYBR® Green Master Mix) using the C1000 Touch CFX384 thermocycler (Bio-Rad).</p> <p>The relative amount of each miRNA was calculated with ∆Ct = CtmiRNA - CtUniSp2, and it was later normalized using the GeNorm algorithm. The final data was calculated with the formula 2<sup>-∆Ct</sup> and the values were expressed as the fold change (FC) of each miRNA with respect to UniSP2<em>.</em></p>
Clouds and Radiation in a mock-Walker Circulation model output
<p>Data used in the creation of figures for Silvers and Robinson, Clouds and Radiation in a mock-Walker Circulation. 2020, JAMES. </p> <p>These experiments are described in detail in the paper. They use the AM4.0 physics package on a doubly periodic domain with a warm patch of SST that is fixed in time at the center of the domain. Grid-Spacings of 1km, 2km, 25km, and 100km are explored on different domain sizes. Patterns of Knowledge Circulation in Early Modern East-Central Europe: Tracing Jewish Kabbalistic Textual Units in Multiple-Text Manuscripts
<p>This presentation will provide initial insights into the first comprehensive study of the networks of production and circulation of Jewish esoteric texts in early modern East-Central Europe (1500-1750). This study, which is funded by the German Research Foundation and has begun in 2019, aims to collect datasets and quantitatively analyse the transmission and reception of Jewish esoteric traditions recorded in manuscript form. In doing so, the study makes use of network analysis methods and utilises digital database environment (provided by LAB 1100) that facilitates analysis and visualisation of data with complex temporal, geographical and relational attributes.<br> The scholarship on Jewish books, both in manuscripts and in print, and kabbalah has centred chiefly on studying individual figures, narrowing the focus of study to singular individuals and groups. Such an approach, although important, stops short of unearthing broader patterns and dissemination of ideas, which could be available through analysis of a larger selection of sources. Virtually no study has been to date devoted to examining broader networks of esoteric textual knowledge exchange from the perspective of the material evidence of such interactions. Such examination may offer a more comprehensive view on East-Central European dynamics of creating, transmitting, and re-appropriating kabbalistic and esoteric sources.<br> Of special interest is the phenomenon of copying, transcription and compiling of kabbalistic textual units in multiple-text manuscripts, often of complex codicological structure, which abound as the most universal medium of dissemination for esoteric (kabbalistic) texts. In recent years, substantial methodological changes occurred in the field of manuscript studies, which emphasise positioning and contextualizing manuscripts as material objects within their social and cultural milieus, and rediscovers the network of ‘manuscript cultures,’ i.e. ‘the urban micro-cultures,’ which left their imprints on both the external aspects and the contents of the codices. The current presentation will highlight potentials and challenges of network analysis for the study of circulation of textual units and their variants in multiple complex and composite manuscripts, whose transcription gives evidence to conscious decisions of those involved in their dissemination and subsequent transmission in East-Central Europe. As the study will have run for just over one year by the date of the conference, the presentation will refrain from offering final conclusions, but remain open to discussions and suggestions from scholars deploying similar methods to the study of manuscripts and manuscript cultures.</p>

Data from: Heritable variation in circulating glucocorticoids and endocrine flexibility in a free-living songbird
Phenotypic flexibility is a central way that organisms cope with challenging and changing environments. As endocrine signals mediate many phenotypic traits, heritable variation in hormone levels, or their context-dependent flexibility, could present an important target for selection. Several studies have estimated the heritability of circulating glucocorticoid levels under acute stress conditions, but little is known about the potential for either baseline hormone levels or rapid endocrine flexibility to evolve. Here we assessed the potential for selection to operate on the elevation (circulating hormone levels) and flexibility of glucocorticoid reaction norms to acute restraint stress. Multivariate animal models revealed low but significant heritability in baseline (h2=0.13-0.14) and stress-induced glucocorticoids (h2=0.18), and moderate heritability in glucocorticoid flexibility in response to acute stress (h2=0.38) in free-living juvenile tree swallows (Tachycineta bicolor; n=408). Baseline glucocorticoids were not genetically correlated with either stress-induced glucocorticoids or glucocorticoid flexibility. These findings indicate that baseline glucocorticoids and the acute stress response are distinct traits that can be independently shaped by selection. Microevolutionary changes that influence the expression or flexibility of these endocrine mediators of phenotype may be an important way that populations adapt to changing environments and novel threats.

CCT was measured through the digital subtraction angiography (DSA) technique. </p> <p>Statistical Parametric Mapping software (SPM, Wellcome Department of Cognitive Neurology, Institute of Neurology, University College London; http://www.fil.ion.ucl.ac.uk/spm/) and ad-hoc scripts developed in the MATLAB scientific computing environment (http://www.mathworks.com, MathWorks, MA, USA) were applied to valuate the lesion volume and brain volume in MS patients.</p>

Circulation in the Guafo Mouth: the gateway to northern Patagonia
<p>These data include the current velocities collected with an Acoustic Doppler Current Profiler (ADCP) in the Guafo Mouth from April 2021 to November 2023. </p><p>The file '1_ADCP_Corcovado_FR_06.10.2021.mat' is the raw ADCP data collected from April to October 2021. </p><p>The file 'Oct_2021_abr_2022.mat' is the raw ADCP data collected from October 2021 to April 2022. </p><p>The file 'ADCP_NOV2023.mat' is the raw ADCP data collected from April 2023 to November 2023. </p><p>The file 'CTDO_TimeSerie_Guafo_Jan2016_Oct2022_daily.xls' contains the time series of oxygen, salinity and temperature collected at the ADCP mooring from 2016 to 2022.
