Development of Ensemble Steric and Electrostatic Chirality (ESEC) descriptors for modelling chromatographic enantioseparations

For example for the Excel file with the title "Borate_chiral_explicit_biased_charged_uncharged.xlsx":

- In the first tab, titled "Exp_water_acn_biased_charge", you can find the biased chiral descriptors for the molecules in their pH 9 state, simulated in explicit solvent.

- In the second tab, titled "Exp_water_acn_biased_uncharged", you can find the biased chiral descriptors for the molecules in their uncharged state, simulated in explicit solvent.

- The third tab, titled "Chiral log alfaRS charge uncharge", includes the responses (log αRS and αRS), along with the retention times and retention factors (k) for each molecule. In addition, this tab contains the descriptors from both the first and the second tab.

- The fourth and the fifth tabs present the experimental and predicted responses (log αRS in the fourth tab and αRS in the fifth tab) for the models that were built.

The structure of the other files follows a similar pattern: the first tabs provide the descriptor values, followed by a tab containing the modelling information (response(s) and various descriptor sets) and finally, a tab is included with the predicted response values.

A predictive flight-altitude model for avoiding future conflicts between an emblematic raptor and wind energy development in the Swiss Alps

Deployment of wind energy is proposed as a mechanism to reduce greenhouse gas emissions. Yet, wind energy and large birds, notably soaring raptors, both depend on suitable wind conditions. Conflicts in airspace use may thus arise between wind energy development and wildlife protection due to the risks of collisions of birds with the blades of wind turbines. Using locations of GPS-tagged bearded vultures, a rare scavenging raptor reintroduced into the Alps, we built a spatially-explicit model to predict potential areas of conflict with future wind turbines deployments in the Swiss Alps. We modelled the probability of bearded vultures flying within or below the rotor-swept zone of wind turbines as a function of wind and environmental conditions, including food supply (presence of wild ungulates). Flight activity at potential risk of collision was generally high, concentrating on south-exposed mountainsides, especially in areas where ibex carcasses have a high occurrence probability, with critical areas covering vast expanses throughout the Swiss Alps. Our model provides a spatially-explicit decision tool that will guide authorities and energy companies for planning the deployment of wind farms in a proactive manner to reduce risk to emblematic Alpine wildlife.

Data from: How well do embryo development rate models derived from laboratory data predict embryo development in sea turtle nests?

Development rate of ectothermic animals varies with temperature. Here we use data derived from laboratory constant temperature incubation experiments to formulate development rate models that can be used to model embryonic development rate in sea turtle nests. We then use a novel method for detecting the time of hatching to measure the in situ incubation period of sea turtle clutches to test the accuracy of our models in predicting the incubation period from nest temperature traces. We found that all our models overestimated the incubation period. We hypothesize three possible explanations which are not mutually exclusive for the mismatch between our modeling and empirically measured in situ incubation period: (1) a difference in the way the incubation period is calculated in laboratory data and in our field nests, (2) inaccuracies in the assumptions made by our models at high incubation temperatures where there is no empirical laboratory data, and (3) a tendency for development rate in laboratory experiments to be progressively slower as temperature decreases compared with in situ incubation.

Development of a Multi-Level Dynamic Model to Measure the Resilience Level of Transportation Infrastructure Networks

The recent increase in disasters is making the largest critical infrastructure system namely the transportation infrastructure system susceptible to unexpected damage. Discontinuation of services provided by transportation infrastructures will create significant societal, economic, and collateral damages. Therefore, this study aims to identify dimensions to measure the resilience of the transportation infrastructures. This study also aims to develop a model to measure the resilience of the transportation infrastructures resilience. To fulfill the aims of this study, a questionnaire was developed which was supported by a comprehensive literature review. 92 valid responses were received and analyzed qualitatively and quantitatively. Statistically significant variables were used to develop a resilience measurement tool. The developed tool will provide relative resilience measures for multiple projects which will help in identifying the most vulnerable segment of the transportation infrastructure network. Exploratory factor analysis (EFA) was performed to identify the constructs and structural equation modeling (SEM) was used to develop the model. Without previous experience in reconstruction works, handling integrated assets becomes very critical. Also, such inexperience makes it difficult to handle emergency resources properly. However, such issues regarding integrated assets can be resolved by investing in locating integrated assets away from the roadways, so if a break in a railroad crossing or utility line occurs or emergency repairs are needed, the impact on the roadway operations can be minimized. To avoid issues related to access to previous disaster data for the roadway this study suggess investing in preparing an interactive online platform for recording and reviewing data related to disasters as well as previous resilience enhancing activities for the roadway with easy access credentials. The findings of this study will support practitioners and decision-makers in investing in the appropriate resilience enhancement activity project for funding and investment.

Development of human pancreatic cancer avatars as a model for dynamic immune landscape profiling and personalised therapy

Pancreatic ductal adenocarcinoma (PDAC) is the most common form of pancreatic cancer, a disease with dismal overall survival. Advances in treatment are hindered by a lack of preclinical models. Here we show how a personalised organotypic 'avatar' created from resected tissue, allows spatial and temporal reporting on a complete in situ tumour microenvironment, and mirrors clinical responses. Our perfusion culture method extends tumour slice viability, maintaining stable tumour content, metabolism, stromal composition, and immune cell populations for 12 days. Using multiplexed immunofluorescence and spatial transcriptomics, we identify immune neighbourhoods and potential for immunotherapy. We employed avatars to assess the impact of a pre-clinically validated metabolic therapy and show recovery of stromal and immune phenotypes and tumour re-differentiation. To determine clinical relevance, we monitored avatar response to gemcitabine treatment and identified a patient avatar-predicable response from clinical follow-up. Thus, avatars provide valuable information for the syngeneic testing of novel therapeutics and a truly personalised therapeutic assessment platform for patients.
