Raw data for PIP2 interaction with TRPC3, explored through computation and electrophysiology
<p>The transient receptor potential canonical type 3 (TRPC3) channel plays a pivotal role in regulating neuronal excitability within the brain via its constitutive activity. The channel is intricately regulated by lipids and has previously been demonstrated to be positively modulated by PIP2. Using molecular dynamics simulations and patch clamp techniques, we reveal that PIP2 predominantly interacts with TRPC3 at the L3 lipid binding site, located at the intersection of pre-S1 and S1 helices. We propose a novel signal transduction pathway from the L3 through the re-entrant loop to a salt bridge between the TRP helix and S4-S5 linker. Notably, we find that both stimulated and constitutive TRPC3 activity require PIP2. These structural insights into the function of TRPC3 are invaluable for understanding the role of the TRPC subfamily in health and disease in native tissue.</p>

<p>Hourly renewable generation profiles compiled for the Western United States as part of the <a href="https://gridlab.org/gridpathratoolkit/">GridPath Resource Adequacy Toolkit</a>.</p> <p>Profiles are stated as a capacity factor (a fraction of nameplate capacity). There are 3 different levels of processing or aggregation provided, all at hourly resolution: Individual plant (wind) or generator (solar) output, capacity-weighted averages of wind and solar output aggregated to the level of balancing authority territories (or transmission zones for larger balancing authorities), and that same aggregated output but with some problematic individual generator profiles modified such that they match the overall production curve of the balancing authority they are within. This data also contains some daily weather data from several sites across the western US and tables describing the way in which individual wind and solar projects were aggregated up to the level of balancing authority or transmission zone.</p>
Compiled database, code and raw data for the article "A Comprehensive Database of Leaf Temperature, Water, and CO2 Fluxes in Young Oil Palm Plants Across Diverse Climate Scenarios for the Evaluation of Functional-Structural Models"
<p>This dataset results from an experiment on young oil palm plants (<em>Elaeis guineensis</em>) in the Ecotron facility from CNRS in Montpellier. Four plants were put in a microcosm one by one with varying climatic conditions to investigate the effect of climate on leaf temperature, CO2, and H2O fluxes at the plant scale. The conditions were defined based on typical daily conditions from a location where it is grown (Libo, Indonesia), <em>i.e.</em>, a day with no rainfall and near-average air temperature and humidity. This base condition was then modified by adding more CO2 (400, 600 and 800ppm), less radiation (typical cloudy sky), and more or less temperature and vapour pressure deficit (± 30%).</p> <p>Find more details from the <code>README.md</code> file in the repository or from the associated <a href="https://github.com/PalmStudio/Biophysics_database_palm" target="_blank" rel="noopener">Github repository</a>.</p>

<p><strong>Abstract</strong><br> The use of wood-based materials in building and construction is constantly increasing as environmental aspects and sustainability gain importance. For structural applications, however, there are many examples where hybrid material systems are needed to fulfil the specific mechanical requirements of the individual application. In particular, metal reinforcements are a common solution to enhance the mechanical properties of a wooden structural element. Metal-reinforced wood components further help to reduce cross-sectional sizes of load-bearing structures, improve the attachment of masonry or other materials, enhance the seismic safety and tremor dissipation capacity, as well as the durability of the structural elements in highly humid environments and under high permanent mechanical load. A critical factor to achieve these benefits, however, is the mechanical joint between the different material classes, namely the wood and metal parts. Currently, this joint is formed using epoxy or polyurethane (PU) adhesives, the former yielding highest mechanical strengths, whereas the latter presents a compromise between mechanical and economical constraints. Regarding sustainability and economic viability, the utilization of different adhesive systems would be preferable, whereas mechanical stabilities yielded for metal-wood joints do not permit for the use of other common adhesive systems in such structural applications.<br> This study extends previous research on the use of non-thermal air plasma pretreatments for the formation of wood-metal joints. The plasma treatments of Norway spruce (Picea abies (L.) Karst.) wood and anodized (E6/EV1) aluminum AlMgSi0.5 (6060) F22 were optimized, using water contact angle measurements to determine the effect and homogeneity of plasma treatments. The adhesive bond strengths of plasma-pretreated and untreated specimens were tested with commercial 2-component epoxy, PU, melamine-urea formaldehyde (MUF), polyvinyl acetate (PVAc), and construction adhesive glue systems. The influence of plasma treatments on the mechanical performance of the compounds was evaluated for one selected glue system via bending strength tests. The impact of the hybrid interface between metal and wood was isolated for the tests by using five-layer laminates from three wood lamellae enclosing two aluminum plates, thereby excluding the influence of congeneric wood-wood bonds. The effect of the plasma treatments is discussed based on the chemical and physical modifications of the substrates and the respective interaction mechanisms with the glue systems. </p>
ACR PET phantom raw data and templates for advanced analysis
<p>A zipped folder containing raw PET data of the ACR phantom, which was acquired first for 30 minutes without any activity outside the axial field of view (FOV), followed by another 30 minutes of acquisition with activity outside the FOV.</p> <p>Each acquisition comes with the UTE mu-map in DICOM format, included in both raw data folders, <raw> and <raw_ofov>.</p> <p>Since the MR-based mu-maps are not of sufficient accuracy, the synthetic mu-map has been included (and also the generated hardware mu-map).</p> <p>The design for the templates for generating the synthetic mu-map, NAC PET image, and sampling VOIs are included in folder <design>.</p> <p> </p>

Raw and analyzed data for manuscript "The synergistic effect of microwave drying and plasma surface treatments on the wettability of green wood"
<p><strong>Abstract:</strong></p> <p>In spite of being a one-step solution to several problems associated with woodworking and being energy efficient, the application of microwave (MW) modification in wood research remained very limited and the promising method has practically no use in wood industries across the globe. Research done so far in this field primarily sheds light on its potential in enhancing wood permeability, treatability and uniform wood drying. While MW treatments are mostly used on wet or green wood, another modification technique, plasma, has potential benefits to synergistically enhance effects of MW treatment, but has not been applied on wet or green wood specimens, so far. This study takes a first step to investigate effects of plasma treatments (PT) on green wood specimens as well as combinations of MW and plasma treatments. As a preliminary study, the methodology focuses on water contact angle measurements, since these are most commonly used as indicators for surface modifications in industrial applications. On the investigated samples of Norway spruce (<em>Picea abies</em> Karst.), an exponential time dependence was found for the contact angle. Initial contact angle after droplet deposition increased due to drying and migration of organic molecules during treatments. In comparison to the literature, the effect of plasma was significantly less pronounced on wet wood specimens. The initial contact angle showed lowest statistical variations after MW treatment, whereas plasma increased inhomogeneities. The final contact angle on treated specimens was lowest for PT-only specimens as well as specimens treated with plasma after MW. In contrast to the initial contact angle, the final contact angle showed lowest variations after PT. Wetting rates were insignificantly improved by plasma, with reduced statistical variations after all treatments.</p>
Raw data: Diversity in root architecture of durum wheat at stem elongation under drought stress
<p>Raw data on above and below ground traits from a greenhouse drought stress experiment with six durum wheat varieties performed at Tuscia University, Viterbo, Italy. Measurements were performed at stem elongation stage; recorded traits: plant shoot length, dry weight, number of leaves and tillers; total root length, root surface area, mean diameter, volume, number of tips, forks, crossings, root dry weight and root angle. Root measurments were performed on the whole root system and the topsoil area (upper 5 cm). </p>

PloS one, 12(4), e0175856.</em></p> <p><strong>Please cite the above article when using the data.</strong></p> <p>The data set with N=13 subjects is different to ordinary ERP datasets in the sense that the train of stimuli to spell one character (68) is divided into repetitions of two interleaved sequences with length 8 and 18, respectively. We added '#' symbols to the spelling matrix which should never be attended by the subject and hence, are non-targets by definition. The first, shorter sequence, now highlights only ordinary characters, while the second sequence also highlights '#' -- visual blank symbols. By construction, sequence 1 has a higher target ratio than sequence 2. These known, but different target and non-target proportions are then used to reconstruct the target and non-target class means. This approach which does not need explicit class labels is termed Learning from Label Proportions (LLP). It can be used to decode brain signals without prior calibration session. More details can be found in the article.</p> <p>In another study, the above data set was used to simulate a new unsupervised mixture approach which combines the mean estimation of the unsupervised expectation-maximization algorithm by Kindermans et al. (2012, PLoS One) with the means obtained with the LLP approach. This leads to an unsupervised solution for which the performance is as good as in the supervised scenario. In the online experiment, this was repeated 3 times and each time the online unsupervised classifier was reset at the start of the sentence.</p>

<p>Original dataset and processing code supporting the preprint (scientific publication) "mScarlet fluorescence lifetime reports lysosomal pH quantitatively."</p> <p>Publication Abstract: The lysosome maintains a highly acidic pH, which is critical for successful lysosomal catabolism. Lysosomal pH (pHlys) is difficult to measure because of the simultaneous need for a sensor with large dynamic range, genetic targetability, low pKa, and a quantitative readout. Here, we demonstrate that the fluorescence lifetime of the mScarlet-LAMP1 fusion protein quantitatively reports lysosomal pH, exhibiting a large dynamic range and a pKa well-tuned for the lysosome. Because fluorescence lifetime is an intrinsic property, pH measurements can be achieved in a single fluorescence channel. mScarlet-LAMP1 lifetime allows for individual lysosome-resolved recordings, a critical advance in describing and understanding pHlys heterogeneity. Using this biosensor, we quantify heterogeneity of pHlys in cultured cells at rest and over time during drug treatment. We anticipate that mScarlet-LAMP1 will enable new insights into the diversity of lysosomal physiology and ionic milieu.</p>
