Data from: A pioneering experimental investigation of a novel in-situ dynamic characterization of the tensile/compression stress-strain mechanism on human plantar soft tissue

<p><span>We have conducted the first in-situ and in-vivo dynamic mechanical test on human plantar soft tissue. A dynamic mechanical analysis (DMA)-like device has been invented to perform the in-situ and in-vivo stress-strain tests on living plantar in order to characterize the material mechanism of biological soft tissue, whereas it is nearly impossible to prepare a sample from a living body for classical tests. A series of pioneering tests of tensile/compression on the heel of ten volunteers are reported, with the reference of tests on mimic foot model made by silicon rubber, standard silicon rubber brick sample, and finite elementary analysis. In addition to demonstrating the effectiveness of the device and approach, interesting correlations between the results and clinic data were found, suggesting considerable potential for the invention in future research.</span></p>

Data from: Experimental analysis of organ decay and pH gradients within a carcass and the implications for phosphatization of soft tissues

<p>Replacement of soft-tissues by calcium phosphate yields spectacular fossils. Decay experiments have shown that pH is a major control on the precipitation of calcium phosphate and tissue replication: for this to occur pH must fall below the carbonic acid dissociation constant (pH 6.38). However, in the fossil record, phosphatisation is highly selective - some internal organs, such as muscles, stomachs, and intestines, appear to preferentially phosphatise while other organs seldomly phosphatise. The reasons for this are unclear but one hypothesis is that, during decay, organs create distinct chemical microenvironments and only some fall below the critical pH threshold for mineralization to occur. Here, we present a novel investigation using microelectrodes that records fluctuating dynamic spatial and temporal pH gradients inside of organs within a carcass in real time. Our experiments demonstrate that within a decaying carcass, organ-specific microenvironments are not generated. Rather, a pervasive pH environment forms within the body cavity (i.e. the coelom) which persists until integumentary failure. With no evidence to support the development of organ-specific microenvironments during decay other factors must control organ phosphatisation. We propose it is tissue histology that plays an important role in selective phosphatisation. Tissues with high phosphate content (and those rich in collagen) are most likely to phosphatise. Internal organs that have low tissue-bound phosphate, including the integuments of the stomach and intestine, only phosphatise when associated with ingested phosphate-rich organic matter. Identifying the driver behind selective phosphatisation may provide insights into other highly selective modes of soft-tissue preservation i.e. pyritization.</p>

Data from: Skeletal and soft tissue completeness of the acanthodian fossil record

<p>Acanthodians are a poorly understood paraphyletic grade of extinct Palaeozoic fishes. They play an increasingly prominent role in our understanding of vertebrate evolution as part of the chondrichthyan stem-group even though their evolutionary history is scarce. The limited preservation of their mostly cartilaginous skeleton largely results in a bias towards isolated remains such as fin spines and scales. Here, we quantify the quality of the acanthodian fossil record by using a variation of the Skeletal Completeness Metric (SCM), an approach that calculates how complete the skeletons of individuals are compared to their theoretical complete skeleton. A novel Soft Tissue Completeness Metric (STCM) is introduced to estimate the percentage of soft body tissue preserved as an alternate measurement of completeness. Completeness scores for &gt;1600 specimens comprising &gt;300 taxa obtained from museum collection visits and literature surveys were assembled into a database. Acanthodian completeness peaks in the Lochkovian, Givetian, Bashkirian-Moscovian, and the Asselian, with lowest scores in the Llandovery. There is no correlation between acanthodian taxonomic richness and completeness. Acanthodians show a significantly lower completeness distribution than many tetrapod groups, but a similarly low distribution to bats. Skeletons deposited in freshwater are significantly more complete than in marine environments where sea level significantly negatively correlates with observed completeness. Our assessment of completeness reveals only weak spatial biases influencing the acanthodian fossil record while environmental biases are much higher. Variation in completeness will have an impact on the utility of characters for assessing the phylogenetic relationships and inclusion of the various acanthodian groups.</p>

Concentration of selected proteins in plasma of patient with soft tissue infections or sepsis

<p>The dataset includes two tabular sets of concentration measurements of selected proteins in plasma samples from patients with soft tissue infections or sepsis. Patients with soft tissue infections are classified into three groups: Patients with necrotizing soft tissue infections (NSTI), suspected NSTI cases but where no necrotic tissue was found during surgical exploration (Non-NSTI) and cellulitis. The sepsis patient cohort has heterogeneous etiologies and location of infection. Additionally, we included a group of patients that had surgery related to non-infectious conditions as a healthy cohort of patients (Surgical control).</p> <p>The measurements were performed by Luminex® multiplex immunoassay or ELISA. Each of the two sets were obtained from independent rounds of measurements and differ in the panel of analytes measured and samples included. The first set of measurements (Set01) covers 39 analytes measured in two sets of customized multiplex plates (32-plex and 5-plex), and two in independent ELISA assays. These analytes were measured in 251 NSTI samples, 20 Non-NSTI, 19 cellulitis and 20 surgical controls. The second set of measurements (Set02) consist of a subset of 10 analytes included in the first set and were measured in two multiplex plates (4-plex and 6-plex). The second round of measurements were carried out in 60 additional NSTI patients and 24 sepsis patients.</p> <p>Lastly, imputation of censored data was carried out only in the first set of measurements and the resulting data is also available in the current data set (Set01_imputed).</p>

Data from: A Silurian ophiuroid with soft tissue preservation

<p>Palaeozoic brittle stars are not equipped with the fused arm ossicles (vertebrae) that facilitate the remarkable mode of walking that characterizes living forms. Here we describe a stem ophiuroid from the Herefordshire Lagerstätte (Silurian, Wenlock Series) which is remarkable in preserving the body cavity uncompacted and long tube feet which lack suckers. We tentatively assign the specimen to Protaster. The morphology of the arms and attitude of the specimen suggest that locomotion was achieved by arm propulsion combined with podial walking. The ophiuroid was also capable of different types of feeding strategies. This protasterid ophiuroid increases the diversity of echinoderm higher taxa with preserved soft parts represented in the Herefordshire Lagerstätte.</p>

Preservational modes of some ichthyosaur soft tissues (Reptilia, Ichthyopterygia) from the Jurassic Posidonia Shale of Germany

<p><em><span>Konservat-Lagerstätten</span></em><span>, such as the Toarcian (Early Jurassic) Posidonia Shale of southwestern Germany, are renowned for their spectacular fossils. Ichthyosaur skeletons recovered from this formation are frequently associated with soft-tissues; however, the preserved material ranges from three-dimensional, predominantly phosphatized structures to dark films of mainly organic matter. We examined soft-tissue residues obtained from two ichthyosaur specimens using an integrated ultrastructural and geochemical approach. Our analyses revealed that the superficially-looking 'films' in fact comprise sections of densely aggregated melanosome (pigment) organelles sandwiched between phosphatized layers containing fibrous microstructures. We interpret this distinct layering as representing condensed and incompletely degraded integument from both sides of the animal. When compared against previously documented ichthyosaur fossils, it becomes readily apparent that a range</span> <span>of preservational modes exists between presumed 'phosphatic' and 'carbonized' soft-tissue remains. Some specimens show high structural fidelity (e.g., distinct integumentary layering), while others, including the fossils examined in this study, retain few original anatomical details. This diversity of soft-tissue preservational modes among Posidonia Shale ichthyosaurs offers a unique opportunity to examine different biostratinomic, taphonomic, and diagenetic variables that potentially could affect the process of fossilization. Soft-tissue preservation in the Posidonia Shale likely was regulated by a multitude of factors, including decay efficacy and speed of phosphatic mineral nucleation; these in turn were governed by a seafloor with sustained microbial mat activity fuelled by high organic matter input and seasonally fluctuating oxygen levels. </span></p>
