Fitness consequences of biochemical adaptation in Drosophila melanogaster populations under simultaneous selection for faster pre-adult development and extended life-span

<p>In holometabolous insect like <i>Drosophila melanogaster</i>, critical size is an important time point during larval life, for irreversible commitment to metamorphosis. Here, we studied the impact of restricted growth duration in terms of selection for faster pre-adult development in <i>Drosophila melanogaster</i> populations which resulted in the evolution of reduced critical size on adult life history traits. Selection for faster pre-adult development resulted in biochemical adaptation in larval physiology with no compromise in major biomolecules at critical size time point.  The flies from the selected populations seem to not only commit to metamorphosis on the attainment of critical size but also seem to channelize resources to reproduction as indicated by similar life-time fecundity of CS and NS flies from selected populations, while the Control CS flies significantly lower life-time fecundity compared to Control NS flies. The flies from selected populations seem to achieve longevity comparable to control flies despite being significantly smaller in size- thus resource constrained due to faster pre-adult development.</p>

Genome alignments for 'Machine-driven parameter-space exploration of biochemical reactions'

<p>The development of complex, multi-step <em>omics</em> methods in molecular biology is a laborious, costly, iterative and often intuition-bound process where an optimum is sought in a parameter space through step-by-step optimisations. The the difficulty of miniaturising assays and the cost of the experiments limit the dynamic range and the number of parameters that can be explored. However, because of non-linearities of the response of biochemical systems to their reagent concentrations, a broad dynamic range is necessary. Here we demonstrate the use of a high-performance nanoliter handling platform (Labcyte Echo 525) and computer generation of liquid transfer programs to explore in quadruplicates more than 600 combination of 4 parameters of a biochemical reaction, which lead us to uncover non-linear responses, parameter interactions and novel mechanical insights. With the increased availability of &laquo; <em>cloud biology</em> &raquo; computer-driven laboratory platforms, our results participate in changing methods development for biotechnology towards reproducible, computer-aided exhaustive characterisation of biochemical systems.</p> <p>This dataset contains the sequence alignments and other processing files produced by running the raw data (10.5281/zenodo.1680999) through a processing pipeline using the MOIRAI workflow manager. Arbuscular mycorrhizal fungi improve the growth and drought tolerance of Cinnamomum migao by enhancing physio-biochemical responses

<p>Drought is the main limiting factor for plant growth in karst areas with a fragile ecological environment. <em>Cinnamomum</em> <em>migao</em> H. W. Li is an endemic medicinal woody plant present in the karst areas of southwestern China, and it is endangered due to poor drought tolerance. Arbuscular mycorrhizal fungi (AMF) are known to enhance the drought tolerance of plants. However, few studies have examined the contribution of AMF in improving the drought tolerance of <em>Cinnamomum</em> <em>migao</em> seedlings. Therefore, we conducted a series of experiments to determine whether a single inoculation and coinoculation of AMF (<em>Claroideoglomus</em> <em>lamellosum</em> and <em>Claroideoglomus</em> <em>etunicatum</em>) enhanced the drought tolerance of <em>Cinnamomum</em> <em>migao</em>. Further, we compared the effects of single inoculation and coinoculation with different inoculum sizes (20, 40, 60, and 100 g; four replicates per treatment) on mycorrhizal colonization rate, plant growth, photosynthetic parameters, antioxidant enzyme activity, and malondialdehyde (MDA) and osmoregulatory substance contents. The results showed that compared with nonmycorrhizal plants, AMF colonization significantly improved plant growing status; net photosynthetic rate; superoxide dismutase, catalase, and peroxidase activities; and soluble sugar, soluble protein, and proline contents. Further, AMF colonization increased relative water content and reduced MDA content in cells. These combined cumulative effects of AMF symbiosis ultimately enhanced the drought tolerance of seedlings and were closely related to the inoculum size. With an increase in inoculum size, the growth rate and drought tolerance of plants first increased and then decreased. The damage caused by drought stress could be reduced by inoculating 40–60 g of AMF, and the effect of co-inoculation was significantly better than that of single inoculation at 60 g of AMF, while the effect was opposite at 40 g of AMF. Additionally, the interaction between AMF and inoculum sizes had a significant effect on drought tolerance. In conclusion, the inoculation of the AMF <em>Claroideoglomus lamellosum</em> and <em>Claroideoglomus</em> <em>etunicatum</em> improved photosynthesis, activated antioxidant enzymes, regulated cell osmotic state, and enhanced the drought tolerance of <em>Cinnamomum</em> <em>migao</em>, enabling its growth in fragile ecological environments.</p>

Dataset related to article: Differential effect of climate of origin and cultivation climate on structural and biochemical plant traits

<p><span>Exploring patterns and causes of intraspecific trait variation is crucial for a better understanding of the effects of climate change on plant populations and ecosystems. However, our current understanding of the intraspecific trait variation is mainly based on structural (morphological) traits, and we have limited knowledge on patterns and causes of variation in biochemical traits (e.g., leaf pigments), which are also crucial for plant adaptation. As a result, we also do not know how similar the climatic effects on structural versus biochemical traits are. </span></p> <p><span>Using plant traits from 110 genotypes representing 11 <em>Festuca</em> <em>rubra</em> populations grown in 4 different climates, we studied trait covariation among structural traits (linked to fitness, resource use, gas exchange, and reproduction) and biochemical traits (linked to photosynthesis, photoprotection, and oxidative stress). We also disentangled the relative role of the climate of origin and the climate of cultivation in the structural versus biochemical traits and tested for adaptive plasticity in the traits. </span></p> <p><span>We found that 1) biochemical traits did not covary with structural traits and represent independent '<em>photoharvesting</em> – <em>photoprotection</em>' strategy dimension of functional variation; 2) interactive effects of climate of origin and cultivation were more pronounced for biochemical than structural traits</span>. 3) T<span>rait plasticity was affected by the climate of origin (</span>precipitation and temperature as well as their interaction<span>); 4) </span><em>F. rubra</em> showed both adaptive and mal-adaptive plasticity, and adaptiveness depended upon trait type, cultivation climate, and climate of origin.</p> <p><span>Overall, our results suggest that structural and biochemical plant traits respond differentially to climate and thus the response of one group of traits cannot be predicted from the other. Responses are also strongly determined by interactions between the climate of origin and cultivation. Thus, more studies on variation in biochemical traits, their correspondence to other traits, and their variation with climate are needed. </span></p>

Characterization of the biochemical and behavioral effects of cannabidiol: implications for migraine

<p>This dataset comprises the findings obtained in the study aimed at investigating the anti-migraine effects of cannabidiol (CBD) in the migraine rat models based by nitroglycerin (NTG) administration. We assayed CBD distribution (15 and 30mg/kg, i.p.) in plasma and in cranial areas related to migraine pain in male Sprague Dawley rats treated chronically (5 days). Successively, we tested CBD activity on the behavioral and biochemical effects induced in the acute and the chronic migraine animal models by NTG administration. In the acute migraine model, rats received CBD (15 mg or 30 mg/kg, i.p) 3 h after NTG (10 mg/kg i.p.) or vehicle injection. In the chronic migraine model, rats were treated with CBD and NTG every other day over nine days with the following doses: CBD 30 mg/kg i.p., NTG 10 mg/kg i.p. We evaluated behavioral parameters with the open field and the orofacial formalin tests. We explored the fatty acid amide hydrolase (FAAH) gene expression, cytokines mRNA and protein levels in selected brain areas and CGRP serum level. CBD levels in the meninges, trigeminal ganglia, cervical spinal cord, medulla pons, and plasma were higher 1 h after the last treatment than after 24 h, suggesting that CBD penetrates but does not accumulate in these tissues. In the acute model, CBD significantly reduced NTG-induced trigeminal hyperalgesia and CGRP and cytokine mRNA levels in peripheral and central sites. In the chronic model, CBD caused only a significant decrease in NTG-induced IL-6 protein levels in the medulla-pons, and trigeminal ganglion. It also reduced CGRP serum levels. In both experimental conditions, there was no modulation of anxiety, motor/exploratory behavior, or grooming.</p> <p>The in vivo and ex vivo assessments were:</p> <p>1)&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; mRNA expression levels: CGRP, SP, interleukin-6 (IL-6), tumor necrosis factor-alpha (TNF-alpha) and FAAH in cervical spinal cord, medulla-pons and trigeminal ganglia. mRNA levels were measured by rt-PCR. All samples were assayed in triplicate and gene expression levels were calculated according to 2&minus;∆∆Ct = 2&minus; (∆Ct gene &minus; ∆Ct housekeeping gene) formula by using Ct (cycle threshold) values.</p> <p>2)&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; Central and peripheral CBD levels were quantified in rat brain tissue and plasma samples using a previously proven online solid phase extraction (SPE) high-performance liquid chromatography (HPLC) method coupled with tandem mass spectrometry (MS/MS).</p> <p>3)&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp;&nbsp; Cytokines and CGRP protein levels were measured using the ELISA kits. Contents and isotope carbon compositions (δ13C) of main biochemical fractions in organs at different growth stages in Phyllostachys edulis

<p><em><span>Phyllostachys edulis</span></em><span> is a spectacularly fast-growing species that completes its height growth within </span><span>two</span><span> months after the shoot emerges </span><span>without producing leaves</span><span> (fast-growing period, FGP). This phase was considered heterotrophic, the carbon necessary for the growth being transferred from the mature culms via the rhizomes, although previous studies observed key enzymes and anatomical features related to C<sub>4</sub>-carbon fixation in developing culms. </span><span>We tested whether C<sub>4</sub>-photosynthesis or dark-CO2 fixation through anaplerotic reactions significantly contributes to the FGP, resulting in differences in the natural abundance of </span><span>δ<sup>13</sup>C</span><span> in bulk organic matter and organic compounds. Further, pulse-<sup>13</sup>CO<sub>2</sub>-labelling was performed on developing culms, either from the surface or from the internal hollow, to ascertain whether significant CO<sub>2</sub> fixation occurs in developing culms.</span><span> δ<sup>13</sup>C of</span><span> young </span><span>shoots and developing culms were higher (-26.3–-26.9‰) </span><span>compared to all organs of mature bamboos (-28.4</span><span>–</span><span>-30.1</span><span>‰). </span>Developing culms contained chlorophylls, most observed in the skin tissues. After <span>pulse-<sup>13</sup>CO<sub>2</sub>-labelling, t</span>he polar fraction extracted from the skin tissues was slightly enriched in <sup><span>13</span></sup><span>C, </span>and only a weak <sup><span>13</span></sup><span>C </span>enrichment was observed in inner tissues<span>. Main carbon source sustaining the FGP was not assimilated by the developing culm, while a </span><span>limited anaplerotic fixation of respired CO<sub>2</sub></span><span> cannot be excluded and is </span>more likely than <span>C<sub>4</sub>-</span>photosynthetic carbon fixation<span>.</span></p>
