``HiPen'': a new dataset for validating (S)QM/MM free energy simulations

<p>Calculating free energy differences between levels of theory (i.e., <span class="math-tex">\(\Delta A^{low \to high}\)</span>) is integral to performing indirect (S)QM/MM free energy simulations. However, connecting levels of theory via free energy simulations has proved difficult due to (1) bond/angle degrees of freedom, (2) dihedral degrees of freedom, and (3) solvent arrangement differences between levels of theory, largely due to partial charge differences between levels of theory. In order to improve calculation of (S)QM/MM free energy simulations, the free energy simulation community should begin to compare methods based on convergence success relative to overall computational time and resource requirements. We have begun to compile such a dataset by calculating <span class="math-tex">\(\Delta A^{MM \to SCC-DFTB}\)</span> in gas phase for 22 drug-like molecules, as seen in our recent publication, Kearns, et al. <strong>2018</strong>, <em>Molecules</em>, Submitted, and we hope that future practitioners will do the same. With this work we hope to provide a standard for comparison for future FES methodologies; additionally, in the near future we hope to continue to add to this dataset including results in more complicated environments such as in solution and in enzyme. All data can be found in our publication and in the accompanying Supporting Information; raw data (such as simulation trajectories and raw data files) can be made available upon request. The purpose of this dataset publication is to make available all starting coordinates, topologies, parameter sets, and input files necessary to replicating the results published in our work.</p>

Energy-related CO2 Emission Accounts and Datasets for 40 Emerging Economies in 2010 - 2019

<p>The Carbon Emission Accounts and Datasets for emerging economies (CEADs, <a href="https://ceads.net">https://ceads.net</a>)&nbsp;aims to provide transparent, verifiable, open-access data on the CO<sub>2</sub> emissions of 40 emerging economies (accounting for 17.5% of the emissions and 12.9% of GDP of the world) for the period 2010-2019. Given the variety of statistical capacity among these countries, we standardized sectors using relevant data on energy and economic statistics and compiled emissions inventories at the regional level using sub-national statistics. For energy-related data, we included emissions from burning biomass, given its significance as an emissions source in these economies. The emissions dataset covers 47 economic sectors and 8 major energy categories in the 40 emerging&nbsp;economies, and in&nbsp;28 we provide&nbsp;a subnational inventory. The emissions inventories&nbsp;are&nbsp;compiled&nbsp;for 47 economic sectors and 8 major categories.</p>

Elevated energy costs of biomass production in mitochondrial-respiration deficient Saccharomyces cerevisiae

Data bundle uploaded by Pranas Grigaitis, p.grigaitis [at] vu.nl.</p> <p><em>Abstract</em></p> <p>Microbial growth requires energy for maintaining the existing cells and producing components for the new ones. Microbes therefore invest a considerable amount of their resources into proteins needed for energy harvesting. Growth in different environments is associated with different energy demands for growth of yeast <em>Saccharomyces cerevisiae</em>, although the cross-condition differences remain poorly characterized. Furthermore, a direct comparison of the energy costs for the biosynthesis of the new biomass across conditions is not feasible experimentally; computational models, on the contrary, allow comparing the optimal metabolic strategies and quantify the respective costs of energy and nutrients. Thus in this study, we used a resource allocation model of <em>S. cerevisiae</em> to compare the optimal metabolic strategies between different conditions. We found that <em>S. cerevisiae</em> with respiratory-impaired mitochondria required additional energetic investments for growth, while growth on amino acid-rich media was not affected. Amino acid supplementation in anaerobic conditions also was predicted to rescue the growth reduction in mitochondrial respiratory shuttle-deficient mutants of <em>S. cerevisiae</em>. Collectively, these results point to elevated costs of resolving the redox imbalance caused by <em>de novo</em> biosynthesis of amino acids in mitochondria. To sum up, our study provides an example of how resource allocation modeling can be used to address and suggest explanations to open questions in microbial physiology.</p>

Variable Geometry Turbines Industrialization for a Circular Economy of Distributed Wind Energy

<p>In this work&nbsp;we present a novel VAWT with passive variable geometry (PVG), which combines variable radius and variable pitch of the free blades. The turbine was optimized through extensive numerical simulations, with a final design featuring short start-up time, high steady-state efficiency, low construction complexity, and low material costs. A crucial reduction of nearly 50% in the blades&#39; mass was obtained through the use of compression moulded composites. A full-scale physical prototype was tested in a dedicated wind tunnel facility, where the typical intermittent air flow of the UBL was replicated. The turbine was connected to standard power electronics found in the solar photovoltaic market. The measured electrical power output closely matches the numerical simulations, suggesting that our PVG design can achieve a 64% increase in the capacity factor compared to a fixed-geometry turbine. Thanks to this engineering breakthrough we believe that, for the first time, wind installations in the UBL can be competitive on the energy market. We conclude our contribution by discussing&nbsp;industrialization and sustainability aspects of our PVG-VAWT technology, and by projecting a deployment scenario towards the uptake of a distributed urban wind market.</p>

In order to find the best solution for the increased flow temperatures compared to a new building for the renovation, various heat pump configurations are evaluated according to energy and economic criteria. A refrigerant with low greenhouse gas potential (GWP) is used as the working medium, e.g. R1234ze (E)) less than 10 used. A favorable refrigeration circuit configuration for high flow temperatures is a twin-circuit system, which essentially consists of two heat pumps with different condensing temperatures and the same evaporation temperatures. An alternative to this is a single-stage configuration with a significantly larger condenser for improved supercooling. Both systems should enable efficient operation when renovating buildings.</p>
