FIGURE 7 in Calcite precipitation forms crystal clusters and muscle mineralization during the decomposition of Cambarellus diminutus (Decapoda: Cambaridae) in freshwater
FIGURE 7. Representative Raman spectra of a mineralized muscle of Cambarellus diminutus (sample C3tank) and observed crystal clusters compared to Raman reference spectra of crystalline calcite and apatite, taken from the RRUFF Raman data base (*R040170, #R060070, Laetsch and Downs, 2006). Raman spectra of the mineralized muscle as well as of the crystal cluster exhibit all main Raman bands typically observed in well crystallized calcite, including the lattice modes, which are absent in amorphous calcium carbonate (Wang et al., 2011).
Hydrolase and Peroxidase Activity of Gold Nanocluster Composites with Lysozyme and Cross-Linked Lysozyme Crystals
<div> <div> <p>Lysozyme (Lyz) and cross-linked Lyz crystals (CLLC) are among the proteins and templates used in the bio-inspired synthesis of subnanometer-sized gold nanoclusters (AuNCs) utilized in various biological applications as biosensors or bioimaging agents and recyclable catalyst, respectively. While previous studies have explored the formation and stabilization of AuNCs in the presence of Lyz, little is known about the enzymatic activity of Lyz after stabilization of AuNCs with the protein (AuNC@Lyz) and in CLLC (AuNC@CLLC). In this study, we conducted a comprehensive analysis of AuNC@Lyz, including hydrodynamic characteristics using multiwavelength fluorescence analytical ultracentrifugation and enzymatic activity assays. AuNC@Lyz exhibited bactericidal performance as efficiently as Lyz at pH 4.0, with enzymatic activity assays indicating more efficient hydrolytic activity of AuNC@Lyz compared to Lyz under the same conditions. Additionally, AuNC@Lyz showed peroxidase-like performance due to the catalytically active surface of AuNCs in the presence of H2O2 and a chromogenic substrate. The multifunctional catalytic capabilities of AuNC@Lyz, including lytic activity and mimic peroxidase function, were highlighted. Additionally, we conducted fluorescent-based hydrolytic activity assays of CLLC, revealing that morphological variations of CLLC influence the catalytic performance of the enzyme crystals depending on the size of solvent channels and Lyz conformation within the CLLC. Furthermore, the bio-inspired synthesis of AuNCs in CLLC resulted in size distributions of AuNCs correlated with pore sizes. As a consequence of the occluded solvent channels and limited breathing of CLLC, AuNC@CLLC displayed a diminishing hydrolytic performance.</p> </div> </div>

Crystal structures determination of CtUGGT-N aka CtUGGT S180C:T742C
<p>Two crystal structures of the double Cys mutant <em>Ct</em>UGGT-N <em>aka</em> <em>Ct</em>UGGT S180C:T742C have been determined by X-ray crystallography. The trigonal one, in space group P3212 was determined at 4.7 Å resolution by molecular replacement using PDB ID 5NV4 as a search model, against the data collected by Mario Hensen and Roberta Ibba on I24@Diamond on 08.08.2018. The orthorhombic one, in space group P212121, was determined to 4.6 Å resolution by molecular replacement using the P3212 <em>Ct</em>UGGT-N model as a search model, against the data collected by Pietro Roversi and Roberta Ibba on I03@Diamond on 05.05.2018. Both structures show the disulphide bridge across the engineered double mutation S180C:T742C, and the TRXL1 and TRXL3 domains clamped shut. In the P3212 form the orientation of TRXL2 with respect to the rest of the protein is the most opened ever observed so far (a rotation of 60 degrees away from the TRXL2 in PDB ID). The orthorhombic form proves that the clamped shut structure (<em>Ct</em>UGGT-N P212121) is compatible with the bent shut structure (<em>Ct</em>UGGT-H, P212121, PDB ID 5NV) <em>i.e.</em> both clamping and bending movements can close at the same time. A quadruple <em>Ct</em>UGGT mutant, containing both D611C:G1050C and S180C:T742C, should be feasible and its human UGGT1 equivalent could provide the best rigidification of the human structure conceivable so far.</p>

In-house high energy remote SAD-phasing using the magic triangle: how to tackle the P1 low symmetry using multiple orientations on the same human IBA57 crystal to increase multiplicity.
<p>The dataset named IBA57-I3C.zip contains pck diffraction images of 9 runs (orientations) of a human IBA57 crystal soaked with 5-amino-2,4,6-triiodoisophthalic acid (I3C) collected on an in-house source. Multiple orientations (runs) of the very same triclinic crystal have been exploited to acquire sufficient real data multiplicity for SAD-phasing. Each folder named run* contains the pck images and the relevant XDS.INP file for processing. The file runs.txt in the top directory gives the phi, kappa, omega and theta details on each of the 9 runs.</p> <p>The dataset named IBA57-native.zip contains pck images of 1 run (360 images around phi only) of a native human IBA57 crystal collected on the same in-house source and the relevant XDS.INP file for processing; this dataset is needed for SIRAS along with the I3C dataset.</p> <p> </p>
