Polymer nanoparticles pass the plant interface

As agriculture strives to feed an ever-increasing number of people, it must also adapt to increasing exposure to minute plastic particles. To learn about the accumulation of nanoplastics by plants, we prepared well-defined block copolymer nanoparticles by aqueous dispersion polymerisation. A fluorophore was incorporated via hydrazone formation and uptake into roots and protoplasts of Arabidopsis thaliana was investigated using confocal microscopy. Here we show that uptake is inversely proportional to nanoparticle size. Positively charged particles accumulate around root surfaces and are not taken up by roots or protoplasts, whereas negatively charged nanoparticles accumulate slowly and become prominent over time in the xylem of intact roots. Neutral nanoparticles penetrate rapidly into intact cells at the surfaces of plant roots and into protoplasts, but xylem loading is lower than for negative nanoparticles. These behaviours differ from those of animal cells and our results show that despite the protection of rigid cell walls, plants are accessible to nanoplastics in soil and water.

Dataset for the paper "The Relationship Between Different Python Argument-Passing Mechanisms and Fixes: An Empirical Study"

This is the replication package of the article

The Relationship Between Different Python Argument-Passing Mechanisms and Fixes: An Empirical Study

Abstract:

Modern programming languages (such as Python, but not only) have introduced a variety of constructs and syntactical elements to make software development more efficient and concise. Examples include lambda functions, comprehension collections, of mechanisms to facilitate the passing of arguments to a function. While many of such constructs may, in principle, be beneficial for developers, recent studies have shown that certain programming constructs introduce atoms of confusion, and may therefore affect program understanding. Moreover, recent work has shown that changes to some constructs, namely functional constructs of Python, induce more fixes than other changes. This paper studies the effect of different Python argument-passing mechanisms to investigate their relationship with code proneness to be fixed. Specifically, we study the fix-proneness for what concerns the function definition, as well as its invocation. This is done by analyzing the evolutionary history of 200 Python projects, for a total of about 3M functions and 12M call sites. While there are varying effects for what concerns parameter declaration mechanisms, we found evidence that keyword-based argument passing is less defect prone than positional argument passing, and this is not affected by size-related confounding factors.

PASS - A SCORING SYSTEM TO EVALUATE PERSISTENT ACUTE KIDNEY INJURY IN CRITICALLY ILL ADULT ICU PATIENTS

Acute Kidney Injury (AKI) and sepsis is highly prevalent in Intensive Care Units (ICU) and has direct effect on mortality and morbidity. We aimed to evaluate if course and recovery of sepsis-AKI can be predicted using variables collected at the time of admission, and to develop a formula - Persistent AKI Scoring System (PASS) to predict recovery.

63 consecutive patients admitted for sepsis-AKI in our ICUs were evaluated over a period of 8 months. Baseline demographic and clinical/laboratory parameters ,including serum creatinine (SCr), Base Excess (BE), Plethysmographic Variability Index (PVI), Caval Index, R wave Variability (RVI), Mean Arterial Pressure (MAP) and Renal Resistivity Index (RI) using renal doppler and need for inotropes were assessed on admission. Patients were managed with standard protocols with fluid resuscitation and other specific therapy. After 6 of hours of fluid resuscitation, hemodynamic status and volume status of the patient was assessed. Patients were classified as Volume Responders or Non-responders depending on hemodynamic stabilization using parameters like MAP. Re- assessment of all the variables was done at 24 hours and 72 hours after admission. The primary outcome was persistent AKI after 72hrs. The secondary outcome was initiation of dialysis and death within 15 days of admission.

34 subjects recovered from AKI. 32 patients showed volume response with respect to hemodynamic parameters and 31 patients were non-responders. Response to fluid, MAP at admission, MAP after 6 hours, BE at admission, inotrope requirement, and PVI at admission did not correlate with recovery of AKI. RI, RVI and S Cr at admission significantly correlated with recovery from AKI These 3 parameters were evaluated further to develop the model for predicting recovery from AKI.• Score> 7.8 predicted recovery from AKI with sensitivity of 79.4%, specificity of 72.4%. PPV 81.8% and NPV 76.7%. PASS Score>7.8 had AUROC of 0.85.

Multiple logistic regression showed that SCr< 2.36 mg%, RVI > 14.45 and RI < 0.8 on admission correlated with recovery.

PASS SCORE Formula was created using the same.

CONCLUSION: PASS score can be a simple bed side method to reliably identify salvageable cases of sepsis-AKI, guiding fluid resuscitation and also aiding early referrals from rural to higher centers for better management.

Psychoacoustic indicators of pass-by road traffic noise

This dataset contains files related to a measurement campaign of pass-by road traffic noise using the Statistical Pass-By (SPB) method according to ISO 11819-1 (2023) on road surfaces in hot mix asphalt.

The compressed folder "Audio files, timestamps and calibration file" is divided into subfolders per measurement day, named according to the measurement date and location. Each subfolder contains continuous recordings as .wav audio files and Excel files with timestamps at 2 seconds before (t0) and 2 s after (tf) the moment a vehicle passage produced the peak noise level in that audio file, besides the air temperature (Tair) and vehicle category (Category). The pass-bys are classified into four categories (Passenger cars, Vans, Dual-axle heavy vehicle - HD and Multiple-axle heavy vehicle - HM).

The file "Calibration file 1Hz 113.7 dB.wav" is a pure-tone 113.7 dB calibration file recorded from the sound level meter and microphone used in the measurements.

The Excel file "Psychoacoustic indicators of pass-by road traffic noise.xlsx" presents nine acoustic and psychoacoustic indicators calculated from the 4s audio excerpts of 2199 pass-by vehicle noise obtained from this measurement campaign. Calculations were conducted using the algorithms implemented on PsySound3. The columns in this Excel file mean the following:

LA,max: average maximum A-weighted sound level, in dB

ΔL: the difference between the peak LA,max and the LA,max at 2 s before the peak was reached, in dB

LA,eq: A-weighted equivalent continuous sound level, in dB

L10: 10% percentile statistical noise level, in dB

L90: 90% percentile statistical noise level, in dB

N50: 50% percentile loudness, in sone

S50: 50% percentile sharpness, in acum

R50: 50% percentile roughness, in asper

FS50: 50% percentile fluctuation strength, in vacil

Tree cone presence and abundance from Niwot Ridge, Rollins Pass, and Rocky Mountain National Park, 2015-2018.
These data were collected to examine the determinants of individual tree cone presence and cone abundance for Engelmann spruce (Picea engelmannii) and subalpine fir (Abies lasiocarpa) in the southern Rocky Mountains, USA. We monitored > 1600 Engelmann spruce and subalpine fir trees for cone presence (an indicator of reproductive maturity) and a subset of those trees for cone abundance (an indicator of seed production) from 2016-2018 in the Colorado Front Range. Cone presence was observed in 1-3 years (2015 or 2016-2018) in 15 plots and cone abundance was observed in 10 of 15 plots in three years (2016-2018). We measured the following for > 24 live trees for each species in each plot: cone presence and/or cone abundance, diameter at breast height, age, height class. For each plot, we measured stand density, basal area, and age of oldest tree to estimate the timing of stand initiation. This database includes three datasets: 1) description of plots (e.g. elevation, aspect, size), 2) cone presence data, and cone abundance data. See Andrus et al. 2020 for more details. Ribosomal stalk proteins RPLP1 and RPLP2 promote biogenesis of flaviviral and cellular multi-pass transmembrane proteins

The ribosomal stalk proteins, RPLP1 and RPLP2 (RPLP1/2), which form the ancient ribosomal stalk, were discovered decades ago but their functions remain mysterious. We had previously shown that RPLP1/2 are exquisitely required for replication of dengue virus (DENV) and other mosquito-borne flaviviruses. Here, we show that RPLP1/2 function to relieve ribosome pausing within the DENV envelope coding sequence, leading to enhanced protein stability. We evaluated viral and cellular translation in RPLP1/2-depleted cells using ribosome profiling and found that ribosomes pause in the sequence coding for the N-terminus of the envelope protein, immediately downstream of sequences encoding two adjacent transmembrane domains (TMDs). We also find that RPLP1/2 depletion impacts a ribosome density for a small subset of cellular mRNAs. Importantly, the polarity of ribosomes on mRNAs encoding multiple TMDs was disproportionately affected by RPLP1/2 knockdown, implying a role for RPLP1/2 in multipass transmembrane protein biogenesis. These analyses of viral and host RNAs converge to implicate RPLP1/2 as functionally important for ribosomes to elongate through ORFs encoding multiple TMDs. We suggest that the effect of RPLP1/2 at TMD associated pauses is mediated by improving the efficiency of co-translational folding and subsequent protein stability.

Estimating densities of larval Salmonflies (Pteronarcys californica) through multiple pass removal of post-emergent exuvia in Colorado rivers

Traditional methods of collecting, sorting, and identifying benthic macroinvertebrate samples are useful for stream biomonitoring and ecological studies, however, these methods are time consuming, expensive, and require taxonomic expertise. Estimating larval densities through collection of post-emergent exuvia can be a practical and time efficient alternative. We evaluated the use of multiple pass depletion techniques of the post-emergent exuvia of Pteronarcys californica to estimate larval densities at ten sites in three Colorado rivers. Exuvia density was highly correlated with both final-instar larval density (R2 = 0.90) and total larval density (R2 = 0.88) and the multiple pass removal technique performed well. Exuvia surveys found P. californica at three low density sites where benthic sampling failed to detect it. At moderate and high density sites the exuvia surveys always produced lower density estimates than benthic surveys. Multiple pass depletion estimates of exuvia proved to be an accurate and efficient technique at estimating larval densities and provided an effective alternative for traditional benthic sampling when objectives are detecting and monitoring P. californica, especially at low density sites.
