Data from: Metabolomic perfusate analysis during kidney machine perfusion: the pig provides an appropriate model for human studies

Introduction: Hypothermic machine perfusion offers great promise in kidney transplantation and experimental studies are needed to establish the optimal conditions for this to occur. Pig kidneys are considered to be a good model for this purpose and share many properties with human organs. However it is not established whether the metabolism of pig kidneys in such hypothermic hypoxic conditions is comparable to human organs. Methods: Standard criteria human (n = 12) and porcine (n = 10) kidneys underwent HMP using the LifePort Kidney Transporter 1.0 (Organ Recovery Systems) using KPS-1 solution. Perfusate was sampled at 45 minutes and 4 hours of perfusion and metabolomic analysis performed using 1-D 1H-NMR spectroscopy. Results: There was no inter-species difference in the number of metabolites identified. Of the 30 metabolites analysed, 16 (53.3%) were present in comparable concentrations in the pig and human kidney perfusates. The rate of change of concentration for 3-Hydroxybutyrate was greater for human kidneys (p<0.001). For the other 29 metabolites (96.7%), there was no difference in the rate of change of concentration between pig and human samples. Conclusions: Whilst there are some differences between pig and human kidneys during HMP they appear to be metabolically similar and the pig seems to be a valid model for human studies.

Ethnobotanical study of medicinal plants used by Ensaro people to treat human diseases

<p class="CxSpFirst"><strong>Abstract </strong></p> <p class="CxSpMiddle"><strong>Introduction</strong>: - People all over the world have been using medicinal plants to treat various ailments and diseases throughout history. It is still common in developing countries. Because is easily accessible, affordable, and trusted by the communities. Although there are several ethnobotanical studies on medicinal plants in Ethiopia, the number of medicinal plant species and indigenous knowledge documented is still low compared to the high diversity of culture and habitats.</p> <p class="CxSpMiddle"><strong>Methods: -</strong> Ethnobotanical data were collected during the 13th of February 2019 and the 26th of October 2020 through Semi-structured interviews, field observation, a guided field walk, and focus group discussions</p> <p><strong>Result: - </strong>the study documented 142 medicinal plant species distributed in<strong> </strong>125 genera and 56 families which used to treat 52 ailments. Most of these species were collected from wild habitats. The most used plant families were Fabaceae (14) followed by lamiaceae (9) and Solanaceae (9). Most of these medicinal plants were herbs followed by shrubs.</p> <p class="CxSpMiddle"><strong>Conclusion</strong></p> <p class="CxSpMiddle">The study revealed a relatively high diversity of medicinal plants that indicated the existence of indigenous knowledge of using medicinal plants to treat ailments and diseases. The findings of this study supported up the idea that indigenous knowledge of using medicinal plants is still alive in highly degraded areas. To make sustainable use of these medicinal plants in the study area, local people must collaborate with natural resource administrators to manage them before they become seriously threatened. Both ex-situ and In-situ conservation strategies should be combined and employed urgently for optimal conservation results</p>

Data from: Incidence and risk factors of first-line antiretroviral treatment failure among human immunodeficiency virus-infected children in Amhara regional state, Ethiopia: a retrospective follow-up study

Objective: This study aimed to assess the incidence and risk factors of treatment failure among Human Immunodeficiency Virus (HIV)/Acquired Immunodeficiency Syndrome infected (AIDS) children who were on Antiretroviral Therapy (ART) in Amhara National Regional State, Ethiopia. Methods: A retrospective follow-up study was conducted from January 2010 to March 2016. A total of 824 children under the age of 15 years who had started ART were included in the study. Data were collected from children's medical chart and ART registration logbook using a standard checklist. A Weibull regression model was used to identify the risk factors for treatment failure. Adjusted Hazard Ratios with 95% confidence intervals were used to declare statistical significance. Results: The mean (±SD) age of the children was 6.4 ± 3.6 years with a median (IQR) follow up of 30.5 (IQR: 14.6, 51.4) months. Sixty-three children (7.7%, 95%CI:5.8, 9.5) developed treatment failure. Seventeen (27.0%) of whom were immunological and 46 (73.0 %) clinical failures. The incidence rate of treatment failure was 22.1/10000 person-months. The cumulative probability of failure was 0.4 with 28562.5 person-month observations. Lack of disclosure [AHR=4. 4, 95%CI (1.8, 11.3)], opportunistic infections during initiation of ART [AHR= 2.3, 95% CI (1.3, 4.1)] and prolonged follow up [AHR = 0.06, 95% CI (0.02, 0.18)] were the main predictors of treatment failure. Conclusion: This study revealed that the incidence of treatment failure remains a significant public health concern in Ethiopia. Undisclosed HIV status to children, the presence of opportunistic infections during initiation of ART and prolonged follow up was found to be the main predictors of treatment failure. Hence, early detection of the treatment failures and further studies on viral monitor need to consider.

Caco-2 in vitro model of human gastrointestinal tract for studying the absorption of titanium dioxide and silver nanoparticles from seafood

<p><a href="https://www.sciencedirect.com/topics/chemistry/titanium-dioxide-nanoparticle">Titanium dioxide nanoparticles</a>&nbsp;(TiO<sub>2</sub>&nbsp;NPs) are widely used in industry as a white pigment (paints, paper industry and toothpastes), photocatalysts (environmental decontamination and photovoltaic cells), inorganic UV filter (sunscreens and personal care products) and as a&nbsp;<a href="https://www.sciencedirect.com/topics/chemistry/food-additive">food additive</a>&nbsp;(E171) and antimicrobial food packaging material.&nbsp;<a href="https://www.sciencedirect.com/topics/chemistry/silver-nanoparticle">Silver nanoparticles</a>&nbsp;(Ag NPs) are used in photonics, microelectronics, catalysis and medicine due to their catalytic activity, magnetic and optical polarizability, electrical and&nbsp;<a href="https://www.sciencedirect.com/topics/chemistry/thermal-conductivity">thermal conductivities</a>&nbsp;and enhanced Raman scattering. They also have antibacterial, antifungal and&nbsp;<a href="https://www.sciencedirect.com/topics/chemistry/antiviral-activity">antiviral activities</a>, as well as anti-inflammatory potential. The huge increase in the use of nano-based products, mainly metallic&nbsp;<a href="https://www.sciencedirect.com/topics/chemistry/nanoparticle">NPs</a>, implies the presence of&nbsp;<a href="https://www.sciencedirect.com/topics/chemistry/nanomaterial">nanomaterials</a>&nbsp;in the environment, and hence, the unintentional human ingestion through water or foods (gastrointestinal tract is the main pathway of NPs intake in humans).</p> <p>The presence of TiO<sub>2</sub>&nbsp;NPs and Ag NPs in seafood samples was firstly established using an ultrasound assisted&nbsp;<a href="https://www.sciencedirect.com/topics/chemistry/enzymatic-hydrolysis">enzymatic hydrolysis</a>&nbsp;procedure and sp-ICP-MS analysis. Several clams, cockles, mussels, razor clams, oysters and variegated scallops, which contain TiO<sub>2</sub>&nbsp;NPs and Ag NPs, were subjected to an&nbsp;<em>in vitro</em>&nbsp;digestion process simulating human gastrointestinal digestion in the stomach and in the small and large intestine to determine the bioaccessibility of these NPs. Caco-2&nbsp;cells were selected as model of human intestinal epithelium for transport studies because of the development of membrane transporters that are responsible for the uptake of chemicals. Parameters as transepithelial electrical resistance (TEER) and permeability of Lucifer Yellow were studied for establishing cell monolayer integrity. TiO<sub>2</sub>&nbsp;NPs and Ag NPs transport as well as total Ti and Ag concentrations passing through the gastrointestinal epithelial barrier model (0&ndash;2&nbsp;h) were assessed by sp-ICP-MS and ICP-MS in several molluscs.</p>
