Dataset related to article "Tumor-Infiltrating Lymphocytes and Macrophages in Intrahepatic Cholangiocellular Carcinoma. Impact on Prognosis after Complete Surgery."

<p>BACKGROUND:</p> <p>Immune infiltrate impacts prognosis of several tumors. To assess the prognostic impact of tumor-infiltrating lymphocytes and macrophages in patients undergoing resection for intrahepatic cholangiocellular carcinoma (ICC).</p> <p>METHODS:</p> <p>All consecutive patients undergoing surgery for ICC between 2008 and 2016 were considered. Inclusion criteria were complete resection and follow-up &gt;&thinsp;12&nbsp;months. Tissue sections were immunostained for CD3+, CD4+, CD8+, Foxp3+, and CD68+. The number of positive cells was quantified using a computer-aided image analysis system. Different cut-off values were tested as predictors of overall survival (OS).</p> <p>RESULTS:</p> <p>Fifty-three patients were analyzed. ICC were T1 in 28 patients, multifocal in 11, and N+ in 13. After a median follow-up of 42&nbsp;months, 5-year OS was 52.1%. The following immune infiltrate values were associated with better OS: CD3+ &gt;&thinsp;0.10% (5-year OS 63.3% vs. 13.6% if &le;&thinsp;0.10%, p&nbsp;=&thinsp;0.001); CD8+ &gt;&thinsp;0.10% (56.2% vs. 28.6% if &le;&thinsp;0.10%, p&nbsp;=&thinsp;0.051); Foxp3+ absent (59.4% vs. 16.0% if present, p&nbsp;=&thinsp;0.049). CD4+ and CD68+ infiltrates were not associated with OS. Three-year OS rates in patients with 0, 1, and &ge;&thinsp;2 negative prognostic factors were 73.6%, 47.3%, and 14.3%, respectively (p&nbsp;&lt;&thinsp;0.001). CD3+ infiltrate stratified prognosis in T1 tumors (3-year OS 71.7% if CD3+ &gt;&thinsp;0.10% vs. 14.3% if &le;&thinsp;0.10%, p&nbsp;&lt;&thinsp;0.001).</p> <p>CONCLUSIONS:</p> <p>Tumor-infiltrating lymphocytes are associated with prognosis of ICC patients after complete surgery. CD3+ and CD8+ infiltrate is associated with higher survival and lower recurrence risk, while Foxp3+ infiltrate is associated with worse prognosis. CD3+ infiltrate allows refining prediction of prognosis in early tumors.</p>

<p>This record contains data related to article &quot;Copy number alterations in stage I epithelial ovarian cancer highlight three genomic patterns associated to prognosis&quot;</p> <p><strong>Background:&nbsp;</strong>Stage I epithelial ovarian cancer (EOC) encompasses five histologically different subtypes of tumors confined to the ovaries with a generally favorable prognosis. Despite the intrinsic heterogeneity, all stage I EOCs are treated with complete resection and adjuvant therapy in most of the cases. Owing to the lack of robust prognostic markers, this often leads to overtreatment. Therefore, a better molecular characterization of stage I EOCs could improve the assessment of the risk of relapse and the refinement of optimal treatment options.</p> <p><strong>Materials and methods:&nbsp;</strong>205 stage I EOCs tumor biopsies with a median follow-up of eight years were gathered from two independent Italian tumor tissue collections, and the genome distribution of somatic copy number alterations (SCNAs) was investigated by shallow whole genome sequencing (sWGS) approach.</p> <p><strong>Results:&nbsp;</strong>Despite the variability in SCNAs distribution both across and within the histotypes, we were able to define three common genomic instability patterns, namely stable, unstable, and highly unstable. These patterns were based on the percentage of the genome affected by SCNAs and on their length. The genomic instability pattern was strongly predictive of patients&#39; prognosis also with multivariate models including currently used clinico-pathological variables.</p> <p><strong>Conclusions:&nbsp;</strong>The results obtained in this study support the idea that novel molecular markers, in this case genomic instability patterns, can anticipate the behavior of stage I EOC regardless of tumor subtype and provide valuable prognostic information. Thus, it might be propitious to extend the study of these genomic instability patterns to improve rational management of this disease.</p>

Per- and polyfluoroalkyl substances (PFASs) exposure in melanoma patients: a retrospective study on prognosis and histological features

<p>Per- and polyfluoroalkyl substances (PFASs) are endocrine disrupting chemicals and previous studies showed that they could be associated with altered vitamin D levels. Since 1965, a massive environmental contamination by PFASs has occurred in North-eastern Italy. This study compared histopathology and prognosis between melanoma patients exposed (n=194) and unexposed (n=488) to PFAS. All patients were diagnosed and/or treated for melanoma at the Veneto Oncological Institute and the University Hospital of Padua (Italy) in 1998-2014. Patients were stratified according to the area of residency and the classification provided by the regional administration. Presence of mitoses was found in 70.5% of exposed patients and 58.7% of unexposed patients (p=0.005). Median follow-up was 90 months (IQR 59-136). 5-year overall survival was 83.7% in exposed patients and 88.0% in unexposed patients (p=0.20); 5-year disease-specific survival was 88.0% in exposed patients and 90.9% in unexposed patients (p=0.50); 5-year disease-free survival was 83.8% in exposed patients and 87.3% in unexposed patients (p=0.20). Our findings suggested that exposure to PFAS was associated with higher level of mitosis in melanoma patients, which may be related to the vitamin D deficiency induced by PFAS. Further studies are required to investigate this relationship and all effects of PFAS on prognosis.</p>
