Measurement of Absolute Retinal Blood Flow Using a Laser Doppler Velocimeter Combined with Adaptive Optics

Data set of measurements related to the following:

Purpose: Development and validation of an absolute laser Doppler velocimeter (LDV) based on an adaptive optical fundus camera which provides simultaneously high definition images of the fundus vessels and absolute maximal red blood cells (RBCs) velocity in order to calculate the absolute retinal blood flow.

Methods: This new absolute laser Doppler velocimeter is combined with the adaptive optics fundus camera (rtx1, Imagine Eyes©,Orsay, France) outside its optical wavefront correction path. A 4 seconds recording includes 40 images, each synchronized with two Doppler shift power spectra. Image analysis provides the vessel diameter close to the probing beam and the velocity of the RBCs in the vessels are extracted from the Doppler spectral analysis. Combination of those values gives an average of the absolute retinal blood flow. An in vitro experiment consisting of latex microspheres flowing in water through a glass-capillary to simulate a blood vessel and in vivo measurements on six healthy human retinal venous junctions were done to assess the device.

Results: In the in vitro experiment, the calculated flow varied between 1.75 μl/min and 25.9 μl/min and was highly correlated (r2 = 0.995) with the imposed flow by a syringe pump. In the in vivo experiment, the error between the flow in the parent vessel and the sum of the flow in the daughter vessels was between −25% and 17% (mean±sd −2 ± 17%). Retinal blood flow in the main temporal retinal veins of healthy subjects varied between 1.3 μL/min and 28.7 μL/min

Conclusion: This adaptive optics LDV prototype (aoLDV) allows the measurement of absolute retinal blood flow derived from the retinal vessel diameter and the maximum RBCs velocity in that vessel.

First evaluation of an absolute quantum gravimeter (AQG#B01) for future field experiments

Quantum gravimeters are a promising new development allowing for continuous absolute gravity monitoring while
remaining user-friendly and transportable. In this study, we present experiments carried out to assess the capacity of the
AQG#B01 in view of future deployment as a field gravimeter for hydro-geophysical applications. The AQG#B01 is the field
version follow-up of the AQG#A01 portable absolute quantum gravimeter developed by the French quantum sensor company
Muquans. We assess the instrument’s performance in terms of stability (absence of instrumental drift), sensitivity in relation to
other gravimeters. Furthermore, the measurements allowed for the successful monitoring of a hydrological mass change. We
discuss the observations concerning the accuracy of the AQG#B01 in comparison with a state-of-the-art absolute gravimeter
(Micro-g-LaCoste, FG5#228). Repeatability is tested by instrument displacement between close-by measurement positions.
We report the repeatability to be better than 50 nm.s−2 . No significant instrumental drift was observed over several weeks
of measurement. This study furthermore investigates whether changes of instrument tilt and external temperature and com-
bination of both, which are likely to occur during field campaigns, influence the measurement of gravitational attraction. We
◦repeatedly tested external temperatures between 20 and 30 C and did not find any significant effect. We report two weeks
of stable operation under semi-terrain conditions in a garage without temperature-control. As an example of a geophysical
signal, a 100 nm.s−2 gravity change is detected with the AQG#B01 after a rainfall event at the Larzac geodetic observatory
(Southern France). The data agreed with the gravity changes measured with a superconducting relative gravimeter (GWR,
iGrav#002) and the expected gravity change simulated as an infinite Bouguer slab approximation. We close with operational
recommendations for potential users and discuss specific possible future field applications. While not claiming completeness,
we nevertheless present the first characterisation of a quantum gravimeter carried out by future users. Crucial criteria for the
assessment of its suitability in field applications have been investigated and are complemented with a discussion of further
necessary experiments.

Data from: Cell wall microstructure, pore size distribution and absolute density of hemp shiv

This paper, for the first time, fully characterises the intrinsic physical parameters of hemp shiv including cell wall microstructure, pore size distribution and absolute density. Scanning Electron Microscopy revealed microstructural features similar to hardwoods. Confocal Microscopy revealed three major layers in the cell wall: middle lamella, primary cell wall and secondary cell wall. Computed tomography improved the visualisation of pore shape and pore connectivity in three dimensions. Mercury intrusion porosimetry showed the average accessible porosity was 76.67 ± 2.03 % and pore size classes could be distinguished into micropores (3-10 nm) and macropores (0.1-1 µm and 20-80 µm). The absolute density was evaluated by helium pycnometry, mercury intrusion porosimetry (MIP) and Archimedes' methods. The results show that these methods can lead to misinterpretation of absolute density. The MIP method showed a realistic absolute density (1.45 g/cm3) consistent with the density of the known constituents: including lignin, cellulose and hemi-cellulose. However, helium pycnometry and Archimedes' methods gave falsely low values due to 10% of the volume being inaccessible pores, which require sample pretreatment in order to be filled by liquid or gas. This indicates that the determination of the cell wall density is strongly dependent on sample geometry and preparation.

Data from: Sequence entropy of folding and the absolute rate of amino acid substitutions

Adequate representations of protein evolution should consider how the acceptance of mutations depends on the sequence context in which they arise. However, epistatic interactions among sites in a protein result in hererogeneities in the substitution rate, both temporal and spatial, that are beyond the capabilities of current models. Here we use parallels between amino acid substitutions and chemical reaction kinetics to develop an improved theory of protein evolution. We constructed a mechanistic framework for modelling amino acid substitution rates that uses the formalisms of statistical mechanics, with principles of population genetics underlying the analysis. Theoretical analyses and computer simulations of proteins under purifying selection for thermodynamic stability show that substitution rates and the stabilization of resident amino acids (the 'evolutionary Stokes shift') can be predicted from biophysics and the effect of sequence entropy alone. Furthermore, we demonstrate that substitutions predominantly occur when epistatic interactions result in near neutrality; substitution rates are determined by how often epistasis results in such nearly neutral conditions. This theory provides a general framework for modelling protein sequence change under purifying selection, potentially explains patterns of convergence and mutation rates in real proteins that are incompatible with previous models, and provides a better null model for the detection of adaptive changes.
