3D Printed Bigel: A Novel Delivery System for Cannabidiol-Rich Hemp Extract

<h2>Abstract</h2> <div>The therapeutic potential of&nbsp;<span>Cannabis sativa</span> L. extract has gained significant attention due to its diverse medical applications. Sublingual administration remains a common delivery method of cannabinoids; however, challenges often arise due to the inconvenient form of the extract and its taste. To address these issues, a novel bigel formulation was developed, combining water and oil phases to enhance stability and bioavailability. This formulation incorporates a cannabidiol-rich hemp extract, hyaluronic acid for its moisturizing properties, and a taste-masking agent to improve patient compliance and comfort. Using a standardized hemp extract rich in cannabinoids and a well-characterized terpene profile, the printability of the bigels was evaluated through 3D printing technology. A printout with known cannabidiol (CBD) and cannabidiolic acid (CBDA) content of 11.613 mg &plusmn; 0.192 of CBD and 4.732 mg &plusmn; 0.280 of CBDA in the printout was obtained. In addition, the release profile of CBD and CBDA was evaluated to determine the delivery efficiency of the active ingredient&mdash;dissolved active ingredient levels ranged from 74.84% &plusmn; 0.50 to 80.87% &plusmn; 3.20 for CBD and from 80.84 &plusmn; 1.33 to 98.31 &plusmn; 1.70 for CBDA depending on the formulation. Rheological studies were conducted to evaluate the viscosity of the bigels under varying temperature conditions, ensuring their stability and usability. Findings suggest that this 3D-printed bigel formulation could significantly enhance the delivery of cannabis extracts, offering a more convenient and effective therapeutic option for patients. This research underscores the importance of innovation in cannabinoid therapies and paves the way for further advancements in personalized medicine.</div>

Toward this end, 3 was functionalized with 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid (DOTA) and 1,4,7-triazacyclononanetriacetic acid (NOTA). On the basis of convincing affinity data, both tracers, [68Ga]NOTA analogue ([68Ga]-5) and [68Ga]DOTA analogue ([68Ga]-4), were evaluated for PET imaging in &ldquo;in vivo&rdquo; models of CHO-hCXCR4 and Daudi lymphoma cells. PET imaging and biodistribution studies revealed higher CXCR4-specific tumor uptake and high tumor/background ratios for the [68Ga]NOTA analogue ([68Ga]-5) than for the [68Ga]DOTA analogue ([68Ga]-4) in both in vivo models. Moreover, [68Ga]-4 and [68Ga]-5 displayed rapid clearance and very low levels of accumulation in all nontarget tissues but the kidney. Although the high tumor/background ratios observed in the mouse xenograft model could partially derive from the hCXCR4 selectivity of [68Ga]-5, our results encourage its translation into a clinical context as a novel peptide-based tracer for imaging of CXCR4-overexpressing tumors.</p> <p>&nbsp;</p> <p>&nbsp;</p>

Genetic background and thermal regime influence adaptation to novel environment in the seed beetle, Callosobruchus maculatus

<p>Climate change is associated with the increase in both mean and variability of thermal conditions. Therefore, the use of more realistic fluctuating thermal regimes is the most appropriate laboratory method for predicting population responses to thermal heterogeneity. However, the long- and short-term implications of evolving under such conditions are not well understood. Here, we examined differences in key life history traits among populations of seed beetles (<em>Callosobruchus</em> <em>maculatus</em>) that evolved under either constant control conditions or in an environment with fluctuating daily temperatures. Specifically, individuals from two distinct genetic backgrounds were kept for 19 generations at one of two temperatures, a constant temperature (T=29°C) or a fluctuating daily cycle (T<sub>mean</sub>=33°C, T<sub>max</sub>=40°C, and T<sub>min</sub>=26°C), and were assayed either in their evolved environment or in the other environment. We found that beetles that evolved in fluctuating environments but were then switched to constant 29°C conditions had far greater lifetime reproductive success compared to beetles that were kept in their evolved environments. This increase in reproductive success suggests that beetles raised in fluctuating environments may have evolved greater thermal breadth than control condition beetles. In addition, the degree of sexual dimorphism in body size and development varied as a function of genetic background, evolved thermal environment, and current temperature conditions. These results highlight not only the value of incorporating diel fluctuations into climate research but also suggest that populations that experience variability in temperature may be better able to respond to both short- and long-term changes in environmental conditions.</p>

Novel disease state model finds most juvenile green turtles develop and recover from fibropapillomatosis

<p>Fibropapillomatosis (FP) is a sea turtle disease characterized by benign tumor development on skin, eyes, and/or internal organs. It primarily affects juvenile green turtles (Chelonia mydas) in coastal foraging sites. The Indian River Lagoon (IRL), Florida, USA, is a coastal green turtle foraging site where the observed FP annual rate averaged 49% between 1983 and 2018. FP is not a major cause of sea turtle mortality and most individuals fully recover; however, the overall dynamics of this disease are poorly understood because prior disease history is unknown for individuals without FP at capture time, and future disease outcome is unknown for individuals with FP at capture time. To better evaluate FP dynamics for green turtles in the IRL, we developed a hierarchical model for predicting disease state change. We used data from 4,149 captures of 3,700 individual green turtles captured in the IRL. The hierarchical disease state model contained two levels: level one modeled whether an individual would develop FP, and level two modeled disease state progression, including states for pre-FP affliction, active FP affliction, and full recovery from FP. From the hierarchical model, we estimated 99.8% (95% credibility intervals 99.1-100%) of juvenile green turtles in the IRL developed FP, indicating that nearly every individual in the IRL is affected by this disease. The model also suggested that turtles quickly developed FP upon recruitment to the IRL and then recovered at different rates, with most completely recovering before emigrating from the IRL as they mature. This is the first analysis of long-term sea turtle data suggesting nearly every turtle in an aggregation both develops and recovers from FP.</p>

Novel polarimetric technique to constrain the magnetic field structure and strength of Gamma-ray burst jets

<p>Gamma-ray bursts (GRBs) are extremely energetic events of cosmological origin. Observed GRBs have high luminosity and rapid variability that requires ultra-relativistic motion in the production mechanism which drive the synchrotron radiation associated with the relativistic jets and their shocked interactions with the local ambient medium. They are broadly divided into two types based on the gamma-ray duration; long GRBs (&gt;2 seconds), and short GRBs (&lt;2 seconds). Long GRBs are thought to be originated from explosions of very massive stars and short GRBs are thought to be produced by the merger of compact binaries. Several key open questions about our understanding of GRB physics remain: What is the driving mechanism of GRB jets? What is the origin and role of magnetic fields in driving the explosion? Since these events happen at cosmological distances, they can not be resolved using traditional astronomical techniques. However, polarimetric observations of GRBs have allowed us to start the exploration of the structure and magnetic field configurations of their relativistic jets. Generally, polarization is measured via the ratio of fluxes by taking consecutive exposures, however for rapidly varying objects such as GRBs, it is not an effective way to observe polarization. Liverpool Telescope (LT) has utilized rapidly rotating polaroids to overcome this problem and created a series of polarimeters that have successfully detected early-time optical polarimetry of various GRBs. I will present photometric and polarimetric results of various GRBs observed by RINGO3. 10 GRBs were bright enough to perform analysis and we were able to perform polarimetric analysis for 7 GRBs. I will discuss how polarimetric detection for a long GRB 191016A along with photometric data constraint the energy injection mechanism for the central engine. In addition, I will present how polarization depends on various properties of GRBs such as photometric decay index, isotropic energy of GRBs, redshift etc.</p>

Word2Vec Models built from a Collection of French 20th-Century Novels

<p>The models were trained&nbsp;using the Gensim library for&nbsp;Python, developed by Radim Rehurek, in 2017. All models are based on the same collection of 20th century French novels that&nbsp;covers the period from 1900 to 2010, with a large range of authors and genres respresented. The collection&nbsp;contains approximately 1,200 novels and about 60 million tokens.</p> <p>The models were created using&nbsp;the SGNS (Skip-Gram with Negative Sampling) architecture, the&nbsp;context window was always of size of 6 + 6 around the target word, and the texts were lemmatised and POS-tagged beforehand. POS-Tags remain attached to each token (as in &quot;souris_nom&quot;).&nbsp;Other&nbsp;parameters vary by model: some have 200, some have 300 dimensional vectors; the minimum frequency of the words in the model varies with values of 50, 100 and 200, something which influences the size of the vocabulary and the size of the model.&nbsp;</p>

[Data from:] Genetic Analysis Reveals Three Novel QTLs Underpinning a Butterfly Egg-Induced Hypersensitive Response-Like Cell Death in Brassica Rapa

<p><strong>Background</strong></p> <p>Cabbage white butterflies (<em>Pieris</em>&nbsp;spp.) can be severe pests of&nbsp;<em>Brassica</em>&nbsp;crops such as Chinese cabbage, Pak choi (<em>Brassica rapa</em>) or cabbages (<em>B. oleracea</em>). Eggs of&nbsp;<em>Pieris</em>&nbsp;spp. can induce a hypersensitive response-like (HR-like) cell death which reduces egg survival in the wild black mustard (<em>B. nigra</em>). Unravelling the genetic basis of this egg-killing trait in&nbsp;<em>Brassica</em>&nbsp;crops could improve crop resistance to herbivory, reducing major crop losses and pesticides use. Here we investigated the genetic architecture of a HR-like cell death induced by&nbsp;<em>P. brassicae</em>&nbsp;eggs in&nbsp;<em>B. rapa.</em></p> <p><strong>Results</strong></p> <p>A germplasm screening of&nbsp;<em>B. rapa</em>&nbsp;56 accessions, representing the genetic and geographical diversity of a&nbsp;<em>B. rapa</em>&nbsp;core collection, showed phenotypic variation for cell death. An image-based phenotyping protocol was developed to accurately measure size of HR-like cell death and was then used to identify two accessions that consistently showed weak (R-o-18) or strong cell death response (L58). Screening of 160 RILs derived from these two accessions resulted in three novel QTLs for&nbsp;P<em>ieris</em>&nbsp;b<em>rassicae-</em>induced&nbsp;cell death on chromosomes A02 (<em>Pbc1</em>), A03 (<em>Pbc2</em>), and A06 (<em>Pbc3</em>). The three QTLs&nbsp;<em>Pbc1-3</em>&nbsp;contain cell surface receptors, intracellular receptors and other genes involved in plant immunity processes, such as ROS accumulation and cell death formation. Synteny analysis with&nbsp;<em>A. thaliana</em>&nbsp;suggested that&nbsp;<em>Pbc1</em>&nbsp;and&nbsp;<em>Pbc2</em>&nbsp;are novel QTLs associated with this trait, while&nbsp;<em>Pbc3</em>&nbsp;contains also LecRK-I.1, a gene of&nbsp;<em>A. thaliana</em>&nbsp;previously associated with cell death induced by a&nbsp;<em>P. brassicae</em>&nbsp;egg extract.</p> <p><strong>Conclusions</strong></p> <p>This study provides the first genomic regions associated with the&nbsp;<em>Pieris</em>&nbsp;egg-induced HR-like cell death in a&nbsp;<em>Brassica</em>&nbsp;crop species. It is a step closer towards unravelling the genetic basis of an egg-killing crop resistance trait, paving the way for breeders to further fine-map and validate candidate genes.</p>

Smartphone imaging repository: a novel method for creating a CT image bank

<p>In a submitted manuscript,&nbsp;test an image capture method using smartphone camera video-derived images of brain computed tomography (CT) scans of traumatic intracranial hemorrhage. The deidentified videos are emailed or uploaded from the emergency department for central adjudication.</p> <p>We measured the time in seconds it took to capture and send the files. The primary outcomes were hematoma volume measured by ABC/2, Marshall Scale, midline shift measurement, image quality by contrast-to-noise ratio (CNR) and time to capture. A radiologist and an imaging scientist applied ABC/2 method, calculated the Marshall scale and midline shift on the data acquired on different smartphones and the PACS in a randomized order. We calculate the intraclass correlation coefficient (ICC). We measured image quality by calculating contrast-to-noise ratio (CNR). We report summary statistics on time to capture in the smartphone group without a comparator.</p>

Data from: Pop‐off data storage tags reveal niche partitioning between native and non‐native predators in a novel ecosystem

1. Niche partitioning might be predicted to be particularly dynamic in 'novel ecosystems' characterized by human-altered environmental conditions and biological invasions. Restoration efforts for native species in such systems can be informed by detailed characterization of niche partitioning. 2. In Lake Ontario, fishery management agencies have been engaged in a long-term struggle to restore native top predators including lake trout (Salvelinus namaycush). Meanwhile, management agencies continue to stock non-native species like Chinook salmon (Oncorhynchus tshawytscha) into the lake to support a recreational fishery and to help control the abundance of a non-native forage fish, the alewife (Alosa pseudoharengus). 3. We used pop-off data storage tags to study fine scale (9.1M lines of data from 22 animals) behaviour and habitat use by lake trout (native) and Chinook salmon (non-native) in Lake Ontario in terms of depth and temperature, recorded at ≤70 s intervals for periods of up to 12 months. 4. Chinook salmon occupied warmer and shallower waters during summer than did lake trout, and their niche breadth was wider. They achieved greater niche breadth in part because they were much more active vertically, cumulatively traveling 103±1 m hour-1 during summer (model-estimated median), whereas most lake trout were relatively inactive vertically (7±1 m hour-1). In each of our analyses, there was more inter-individual variation among lake trout than among Chinook salmon, driven by some lake trout that spent considerable time making forays into warmer, shallower waters. 5. Synthesis and applications. Our results illustrate the different foraging tactics used by two species in the Great Lakes and reflect their distinct life histories. Vertical and thermal niche partitioning between Chinook salmon and lake trout helps to explain how these species can co-exist in a multi-species fishery even while having substantial overlap in diet. The diversity of behaviours exhibited here by native lake trout have likely helped them persist during dramatic changes to the forage base in recent decades; that flexibility could help underlie their long-term prospects for restoration during future changes to the ecosystem.

Social information use about novel aposematic prey depends on the intensity of the observed cue

<p>Animals gather social information by observing the behavior of others, but how the intensity of observed cues influences decision-making is rarely investigated. This is crucial for understanding how social information influences ecological and evolutionary dynamics. For example, observing a predator's distaste of unpalatable prey can reduce predation by naïve birds, and help explain the evolution and maintenance of aposematic warning signals. However, previous studies have only used demonstrators that responded vigorously, showing intense beak-wiping after tasting prey. Therefore, here we conducted an experiment with blue tits (<em>Cyanistes caeruleus</em>) informed by variation in predator responses. First, we found that the response to unpalatable food varies greatly, with only few individuals performing intensive beak-wiping. We then tested how the intensity of beak-wiping influences observers' foraging choices using video-playback of a conspecific tasting a novel conspicuous prey item. Observers were provided social information from: (1) no distaste response, (2) a weak distaste response, or (3) a strong distaste response, and were then allowed to forage on evolutionarily novel (artificial) prey. Consistent with previous studies, we found that birds consumed fewer aposematic prey after seeing a strong distaste response, however a weak response did not influence foraging choices. Our results suggest that while beak-wiping is a salient cue, its information content may vary with cue intensity. Furthermore, the number of potential demonstrators in the predator population might be lower than previously thought, although determining how this influences social transmission of avoidance in the wild will require uncovering the effects of intermediate cue salience.</p>

Novel code for: Synchrony in adult survival is remarkably strong among common temperate songbirds across France

<p>Synchronous variation in demographic parameters across species destabilizes populations, metapopulations and metacommunities and increases extinction risks. Revealing the processes that synchronize population dynamics across species allows us to identify trans-specific demographic processes that are subject to environmental forcing of overarching importance. Using a Bayesian, hierarchical multi-site, multi-species mark-recapture model, we investigated temporal interspecific synchrony in annual adult local survival across 16 common songbird species across France for the period 2001–2016. Adult annual survival was largely synchronous among species (73% [47–94] of the variation among years was common to all species), despite species differing in ecological niche and life-histories. This result was robust to differences in migratory strategy among species, uneven species sample sizes, and time de-trending. Shared synchrony across migratory strategy suggests that environmental forcing during the 4-month temperate breeding season has large-scale, cross-specific, impacts among songbirds. At a scale ~1000 km, a likely proximate mechanism of synchronization is forcing by weather-driven variation in resources, which, in particular, determines the cost of reproduction. However, the strong interspecific synchrony was not easily explained by a set of a priori defined candidate weather variables, with spring weather variables explaining only 1.4% [0.01–5.5] of synchrony, while the contribution of large-scale winter weather indices may be stronger, but uncertain (12% [0.3–37]). Future research may up-scale these results to community dynamics, to understand compensatory intra- and inter-specific demographic processes that preserve meta-communities from synchronization.</p>
