Metadata and data definitions are contained in a.radiate_bloodwork_readme.txt.</p> <p>Dataset contains hematology and biochemistry values of 120 previously confiscated, clinically healthy subadult radiated tortoises living under human care within their native habitat at the Tortoise Conservation Center (TCC), Madagascar. To evaluate the effects of different commonly used techniques on these parameters, we compared results between two venipuncture sites (subcarapacial sinus and brachial vein) and three different WBC quantification methods (Natt and Herrick, Leukopet<sup>TM</sup>, and slide estimate). Data from tortoises removed from the final analyses due to sample quality issues are also included as frequency of sample quality issues varied by venipuncture site.&nbsp;See associated publication for detailed methods.&nbsp;&nbsp;</p> <p>&nbsp;</p>

Heat stress responses of the kelp Laminaria digitata (Phaeophyceae) across Northeast Atlantic populations: growth, biochemistry, chlorophyll fluorescence, pigments

<p>To understand the thermal plasticity of a coastal foundation species across its latitudinal distribution, we assess physiological responses to high temperature stress in the kelp <i>Laminaria digitata</i> in combination with population genetic characteristics and relate heat resilience to genetic features and phylogeography. We hypothesize that populations from Arctic and cold-temperate locations are less heat resilient than populations from warm distributional edges. Using meristems of natural <i>L. digitata</i> populations from six locations ranging between Kongsfjorden, Spitsbergen (79°N), and Quiberon, France (47°N), we performed a common-garden heat stress experiment applying 15°C to 23°C over eight days. We assessed growth, photosynthetic quantum yield, carbon and nitrogen storage, and xanthophyll pigment contents as response traits. Population connectivity and genetic diversity were analysed with microsatellite markers. Results from the heat stress experiment suggest that the upper temperature limit of <i>L. digitata</i> is nearly identical across its distribution range, but subtle differences in growth and stress responses were revealed for three populations from the species' ecological range margins. Two populations at the species' warm distribution limit showed higher temperature tolerance compared to other populations in growth at 19°C and recovery from 21°C (Quiberon, France), and photosynthetic quantum yield and xanthophyll pigment responses at 23°C (Helgoland, Germany). In <i>L. digitata</i> from the northernmost population (Spitsbergen, Norway), quantum yield indicated the highest heat sensitivity. Microsatellite genotyping revealed all sampled populations to be genetically distinct, with a strong hierarchical structure between southern and northern clades. Genetic diversity was lowest in the isolated population of the North Sea island of Helgoland and highest in Roscoff in the English Channel. All together, these results support the hypothesis of moderate local differentiation across <i>L. digitata</i>'s European distribution, whereas effects are likely too weak to ameliorate the species' capacity to withstand ocean warming and marine heatwaves at the southern range edge.</p>

Glider and satellite high resolution monitoring of a mesoscale eddy in the algerian basin: Effects on the mixed layer depth and biochemistry

<p>Despite an extensive bibliography for the circulation of the Mediterranean Sea and its sub-basins, the debate on mesoscale dynamics and their impacts on bio-chemical processes is still open because of their intrinsic time scales and of the difficulties in their sampling. In order to clarify some of these processes, the “Algerian BAsin Circulation Unmanned Survey-ABACUS” project was proposed and realized through access to the JERICO Trans National Access (TNA) infrastructure between September and December 2014. In this framework, a deep glider cruise was carried out in the area between the Balearic Islands and the Algerian coast to establish a repeat line for monitoring of the basin circulation. During the mission a mesoscale eddy, identified on satellite altimetry maps, was sampled at high-spatial horizontal resolution (4 km) along its main axes and from the surface to 1000 m depth. Data were collected by a Slocum glider equipped with a pumped CTD and biochemical sensors that collected about 100 complete casts inside the eddy. In order to describe the structure of the eddy, in situ data were merged with next generation remotely sensed data: daily synoptic sea surface temperature (SST) and chlorophyll concentration (Chl-a) images from the MODIS satellites, as well as sea surface height and geostrophic velocities from AVISO. From its origin along the Algerian coast in the eastern part of the basin, the eddy propagated northwest at a mean speed of about 4 km/day, with a mean diameter of 112–130 km, mean amplitude of 15.7 cm; the eddy was clearly distinguished from the surrounding waters thanks to its higher SST and Chl-a values. Temperature and salinity values over the water column confirm the origin of the eddy from the Algerian Current (AC) showing the presence of recent Atlantic water in the surface layer and Levantine Intermediate Water (LIW) in the deeper layer. The eddy footprint is clearly evident in the multiparametric vertical sections conducted along its main axis.</p> <p>Deepening of temperature, salinity and density isolines at the center of the eddy is associated with variations in Chl-a, oxygen concentration and turbidity patterns. In particular, at 50 m depth along the eddy borders, Chl-a values are higher (1.1–5.2 μg/l) in comparison with the eddy center (0.5–0.7 μg/l) with maximum values found in the southeastern sector of the eddy.</p> <p>Calculation of geostrophic velocities along transects and vertical quasi-geostrophic velocities (QG-w) over a regular 5 km grid from the glider data helped to describe the mechanisms and functioning of the eddy. QG-w presents an asymmetric pattern, with relatively strong downwelling in the western part of the eddy and upwelling in the southeastern part. This asymmetry in the vertical velocity pattern, which brings LIW into the euphotic layer as well as advection from the northeastern sector of the eddy, may explain the observed increases in Chl-a values</p>

Data from: Elevational divergence in pigmentation plasticity is associated with selection and pigment biochemistry

<p>Phenotypic plasticity is predicted to evolve in environmentally variable habitats, or those experiencing a high frequency of strong selection. However, the evolution of plasticity may be constrained by costs or physiological constraints. In flowers, UV-absorbing pigmentation ameliorates UV damage to pollen, and is linked with elevated UV exposure. Whether plasticity contributes to this pattern remains unclear. Petals of <i>Argentina anserina</i> have larger UV-absorbing petal areas at high elevations where they experience higher and more variable UV exposure compared to low elevations. We measured UV-induced pigmentation plasticity in high- and low-elevation populations (hereafter, '<i>high</i>, '<i>low'</i>), and selection on pigmentation via male fitness. We dissected UV pigment biochemistry using metabolomics to explore biochemical mechanisms underlying plasticity. <i>H</i><i>igh</i> displayed positive UV-induced pigmentation plasticity but <i>low</i> lacked plasticity. Selection favored elevated pigmentation under UV in <i>high</i>, supporting adaptive plasticity. In <i>high</i>, UV-absorption was conferred by flavonoids produced in one flavonoid pathway branch. However, in <i>low</i>, UV-absorption was associated with many compounds spanning many branches. Elevated plasticity was thus associated with reduced pigment diversity. Results are consistent with adaptive floral pigmentation plasticity in more extreme and variable environments. We discuss how biochemical underpinnings of pigmentation may permit or constrain the evolution of pigmentation plasticity.</p>

Varying digestible isoleucine level to determine effects on performance, egg quality, serum biochemistry, and ileal protein digestibility in diets of young laying hens

<p class="CxSpFirst">To ascertain an appropriate level of isoleucine for LSL-LITE layers (23- to 30-week-old), diets containing total isoleucine concentrations (levels) of 0.66 (Control), 0.69, 0.72, 0.75, 0.78, 0.81, and 0.84% were fed as 7 treatments (2730 kcal/kg metabolizable energy) x 7 replicates x 10 birds per replicate.  Significance for performance, egg quality, serum bioc Level, week, and level*week (L*W) were significant for production, egg mass, and feed intake.  Level and week were significant for FCR. Week was significant for weight gain.</p> <p>Level was significant for egg weight, specific gravity, and shell thickness; week was also significant for these external egg parameters as well as shape index and proportional shell thickness.  L*W was significant for all except shape index.  For internal egg measurements, level was significant for proportional yolk, proportional albumen, yolk index, and yolk:albumen. Week was significant for internal egg parameters while L*W significantly affected Haugh unit, proportional albumen weight, yolk index, albumen index, and yolk color.  Level was significant for <span class="Heading1Char"><span>globulin and glucose in serum. </span></span></p> <p><span class="Heading1Char"><span>Is</span></span>oluecine at 0.72%, 0.81% and 0.84% produced the lowest FCR, an important standard in the poultry industry.  Considering the low FCR of 1.45 and cost for inclusion as a dietary ingredients, 0.72% isoleucine was chosen for further study.</p>
