Detection of HER2+ Breast Cancer Cells using Bioinspired DNA-Based Signal Amplification
<p>Circulating tumor cells (CTC) are promising biomarkers for metastatic cancer detection and monitoring progression. However, CTC detection remains challenging due to their low frequency and heterogeneity. Herein, we report a bioinspired approach to detect individual cancer cells, based on a signal amplification cascade using a programmable DNA hybridization chain reaction (HCR) circuits. We applied this approach to detect HER2+ cancer cells using the anti-HER2 antibody (trastuzumab) coupled to initiator DNA eliciting a HCR cascade that leads to a fluorescent signal at the cell surface. At 4°C, this HCR detection scheme resulted in highly efficient, specific and sensitive signal amplification of the DNA hairpins specifically on the membrane of the HER2+ cells in a background of HER2- cells and peripheral blood leukocytes, which remained almost non-fluorescent. The results indicate that this system offers a new strategy that may be further developed toward an in vitro diagnostic platform for the sensitive and efficient detection of CTC.</p>
Adaptive Nonlinear Control For Perching of a Bioinspired Ornithopter
<p>This dataset contains the flight data recorded by the onboard flight computer for perching experiments. During the experiments nonlinear guidance and control laws are used for trajectory tracking. The filght computer is designed around the NanoPi Neo Air. A custom made PCB attached to the NanoPi, and acts as a carrier for an STM32 microcontroller. The microcontroller regulates the servos via a PWM signal. At the same time, the flight computer connects with an Optitrack motion capture system. The Optitrack emits data at 120 Hz to a local network using a VRPN protocol. This data is recorded by the flight computer, used for state estimation, and the control is executed at 100 Hz.</p>

Data from: Hydrodynamic analysis of bioinspired vortical cross-step filtration by computational modelling
<p><span><span>Research on the suspension-feeding apparatus of fishes has led recently to the identification of novel filtration mechanisms involving vortices. Structures inside fish mouths form a series of 'backward-facing steps' by protruding medially into the mouth cavity. In paddlefish and basking shark mouths, porous gill rakers lie inside 'slots' between the protruding branchial arches. Vortical flows inside the slots of physical models have been shown to be important for the filtration process, but the complex flow patterns have not been visualized fully. Here we resolve the three-dimensional hydrodynamics by computational fluid dynamics simulation of a simplified mouth cavity including realistic flow dynamics at the porous layer. We developed and validated a modelling protocol in ANSYS Fluent software that combines a porous media model and permeability direction vector mapping. We found that vortex shape and confinement to the medial side of the gill rakers result from flow resistance by the porous gill raker surfaces. Anteriorly directed vortical flow shears the porous layer in the centre of slots. Flow patterns also indicate that slot entrances should remain unblocked, except for the posterior-most slot. This new modelling approach will enable future design exploration of fish-inspired filters.</span></span></p>

Data for: Bioinspired Lubricity from Surface Gel Layers
<p>Surface gel layers on commercially-available contact lenses have been shown to reduce frictional shear stresses and mitigate damage during sliding contact with fragile epithelial cell layers <em>in vitro</em>. Spencer and coworkers recently demonstrated that surface gel layers could arise by polymerizing hydrogels within molds composed of low surface energy materials and near oxygen-rich interfaces from peroxidation gradients. In this study, polyacrylamide hydrogel shell probes (7.5 wt.% polyacrylamide, 0.3 wt.% methylenebisacrylamide) were polymerized in three hemispherical molds listed in order of decreasing surface energy and increasing oxygen permeability: borosilicate glass, polyetheretherketone (PEEK), and polytetrafluoroethylene (PTFE). Hydrogel probes polymerized in PEEK and PTFE molds exhibited 100´ lower elastic moduli at the surface (<em>E*PEEK </em>= 80 ± 31 Pa and <em>E*PTFE</em> = 106 ± 26 Pa, respectively) than those polymerized in glass molds (<em>E*glass</em> = 31,560 ± 1,570 Pa), in agreement with previous investigations by Spencer and coworkers. Biotribological experiments revealed that hydrogel probes with surface gel layers reduced frictional shear stresses against cells (<em><em>τ</em>PEEK</em> = 35 ± 15 Pa and <em>τPTFE</em> = 22 ± 16 Pa) more than those without (<em>τ</em><em>glass</em> = 68 ± 15 Pa) and offered greater protection against cell damage when sliding against human telomerase-immortalized corneal epithelial (hTCEpi) cell monolayers. Our work demonstrates that the "mold effect" resulting in oxygen-inhibition polymerization creates hydrogels with surface gel layers that reduce shear stresses in sliding contact with cell monolayers, similar to the protection offered by gradient mucin gel networks across epithelial cell layers.</p>

Data from: Macro-to-nanoscale investigation of wall-plate joints in the acorn barnacle Semibalanus balanoides: correlative imaging, biological form and function, and bioinspiration
Correlative imaging combines information from multiple modalities (physical–chemical–mechanical properties) at various length scales (centimetre to nanometre) to understand the complex biological materials across dimensions (2D–3D). Here, we have used numerous coupled systems: X-ray microscopy (XRM), scanning electron microscopy (SEM), electron backscatter diffraction (EBSD), optical light microscopy (LM) and focused ion beam (FIB-SEM) microscopy to ascertain the microstructural and crystallographic properties of the wall-plate joints in the barnacle Semibalanus balanoides. The exoskeleton is composed of six interlocking wall plates, and the interlocks between neighbouring plates (alae) allow barnacles to expand and grow while remaining sealed and structurally strong. Our results indicate that the ala contain functionally graded orientations and microstructures in their crystallography, which has implications for naturally functioning microstructures, potential natural strengthening and preferred oriented biomineralization. Elongated grains at the outer edge of the ala are oriented perpendicularly to the contact surface, and the c-axis rotates with the radius of the ala. Additionally, we identify for the first time three-dimensional nanoscale ala pore networks revealing that the pores are only visible at the tip of the ala and that pore thickening occurs on the inside (soft bodied) edge of the plates. The pore networks appear to have the same orientation as the oriented crystallography, and we deduce that the pore networks are probably organic channels and pockets, which are involved with the biomineralization process. Understanding these multiscale features contributes towards an understanding of the structural architecture in barnacles, but also their consideration for bioinspiration of human-made materials. The work demonstrates that correlative methods spanning different length scales, dimensions and modes enable the extension of the structure–property relationships in materials to form and function of organisms.
