Effects of Soil Warming and Substrate Complexity on Microbial Carbon Use Efficiency at Harvard Forest 2017
Soil microbial carbon use efficiency (CUE) is a combination of growth and respiration, which may respond differently to climate change depending on physical protection of soil carbon (C) and its availability to microbes. In a mid-latitude hardwood forest in central Massachusetts, 27 years of soil warming (+5 ◦C) has resulted in C loss and altered soil organic matter (SOM) quality, yet the underlying mechanisms remain unclear. Here, we hypothesized that long-term warming reduces physical aggregate protection of SOM, microbial CUE, and its temperature sensitivity. Soil was separated into macroaggregate (250–2000 μm) and microaggregate (less than 250 μm) fractions, and CUE was measured with 18O-enriched water in samples incubated at 15 and 25 ◦C for 24 h. We found that long-term warming reduced soil C and nitrogen concentrations and extracellular enzyme activity in macroaggregates, but did not affect physical protection of SOM. Long-term warming showed little effect on CUE or microbial biomass turnover time because it reduced both growth and respiration. However, CUE was less temperature sensitive in macroaggregates from the warmed compared to the control plots. Our findings suggest that microbial thermal responses to long-term warming occur mostly in soil compartments where SOM is less physically protected and thus more vulnerable to microbial degradation.
Dataset of "Cobalt and nickel doped WSe2 as efficient electrocatalysts for water splitting and as cathodes in hydrogen evolution reaction PEM water electrolysis"
<p>Efficient electrocatalysts are crucial for water splitting and fuel cells. Using cheap alternatives that can improve reaction kinetics is essntial for advancing fuel cell technology. Although, tungsten diselinide (WSe2) is promising for electrocatalysis is not fully explored, especially in oxygen evolution and in applications such as polymer electrolyte membrane water electrolyzer.<br>In this work, we used a simple approach to dope WSe2 with cobalt and/or nickel atoms. The doped material was subsequently tested for hydrogen evolution reaction and oxygen evolution reaction. Accordingly, the two electrocatalysts are highly active and stable, affording low overpotentials comparable to those of noble metals. The effective introduction of heteroatoms causes the retention of coordination vacancies, furnishing active catalytic sites that enhanced electrocatalytic performance both in activity and charge transfer. Moreover, both doped materials show excellent performance and stability as cathode electrocatalysts in the polymer electrolyte membrane water electrolyzer with great promise for real-world applications.</p>
Properties and formaldehyde removal efficiency of biocarbon-MnO2 particles
<p>The dataset includes information about biocarbon particles doped with different concentrations of MnO2 photocatalyst (BC-MnO2)</p><p>Six different samples: MnO2, biocarbon, BC-MnO2-1, BC-MnO2-2, BC-MnO2-3, BC-MnO2-4</p><p>Characterization of the samples:</p><p>* SEM images collected using scanning electron microscope (Carl Zeiss SUPRA 35 VP)</p><p>* XRD data collected using Bruker D2 Phaser diffractometer</p><p>* Porosity data collected using physisorption analyzer (Autosorb iQ-XR-AG-AG). The dataset contains data about isotherms and pores size distributions from tests under nitrogen gas (meso and macro porosity) and CO2 gas (microporosity).</p><p>Formaldehyde removal potential of the samples: The raw data were generated from an electrochemical formaldehyde sensor (Stox-HCHO) at ambient conditions (temperature of 23 °C, relative humidity between 40% and 46%, and conventional visible light). The sensor was placed in a test chamber equipped with the sensor and 8 µl of formaldehyde (HCHO) solution was injected. Then the test chamber was hermetically closed and changes in formaldehyde levels were measured. The same sensor provided information about temperature and relative humidity in the test chamber. Data was aquired using TVOC-HCHO logger software. The formaldehyde removal efficiency (%) of the samples after 8h of experiment was determined from the raw data.</p>

Briefly, 4 channels (data sets Channel_0,1,2,3, organized in a group called 'ExtracellularData'; extracellular recordings along the Purkinje cell layer of a young rat cerebellar cortex slice) of a linear 'Michigan' (now Neuronexus) probe and a loose cell-attached recording (data set Reference, in group 'CellAttached') from one of the Purkinje cells that is also extracellularly recorded: a 'ground truth' for spike sorting algorithms. Each group has three attributes: SamplingRate, HighPass and LowPass. The last two are the filter settings used prior to A/D conversion. These attributes have identical values for the 5 traces (2 groups): the data were sampled at 15 kHz, high-passed at 300 Hz and low-passed at 5 kHz.</p>
Enhanced Biosafety of the Sleeping Beauty Transposon System by Using mRNA as Source of Transposase to Efficiently and Stably Transfect Retinal Pigment Epithelial Cells
<p>Raw data of the publication "Enhanced Biosafety of the Sleeping Beauty Transposon System by Using mRNA as Source of Transposase to Efficiently and Stably Transfect Retinal Pigment Epithelial Cells".</p> <p>Abstract: Neovascular age-related macular degeneration (nvAMD) is characterized by choroidal<br> neovascularization (CNV), which leads to retinal pigment epithelial (RPE) cell and photoreceptor<br> degeneration and blindness if untreated. Since blood vessel growth is mediated by endothelial cell<br> growth factors, including vascular endothelial growth factor (VEGF), treatment consists of repeated,<br> often monthly, intravitreal injections of anti-angiogenic biopharmaceuticals. Frequent injections are<br> costly and present logistic difficulties; therefore, our laboratories are developing a cell-based gene<br> therapy based on autologous RPE cells transfected ex vivo with the pigment epithelium derived factor<br> (PEDF), which is the most potent natural antagonist of VEGF. Gene delivery and long-term expression<br> of the transgene are enabled by the use of the non-viral Sleeping Beauty (SB100X) transposon system<br> that is introduced into the cells by electroporation. The transposase may have a cytotoxic effect and a<br> low risk of remobilization of the transposon if supplied in the form of DNA. Here, we investigated<br> the use of the SB100X transposase delivered as mRNA and showed that ARPE-19 cells as well as<br> primary human RPE cells were successfully transfected with the Venus or the PEDF gene, followed<br> by stable transgene expression. In human RPE cells, secretion of recombinant PEDF could be detected<br> in cell culture up to one year. Non-viral ex vivo transfection using SB100X-mRNA in combination<br> with electroporation increases the biosafety of our gene therapeutic approach to treat nvAMD while<br> ensuring high transfection efficiency and long-term transgene expression in RPE cells.</p>

Herein, these light-matter interactions are investigated by UV-visible and surface-enhanced Raman scattering (SERS) spectroscopies, supported by finite-difference time-domain (FDTD) calculations. Our results reveal the importance of combining plasmonic nanomaterials and J-aggregates with near-zero-refractive index. As plexcitonic nanostructures nanorattles are employed, based on J-aggregates of the cyanine dye 5,5,6,6-tetrachloro-1,1-diethyl-3,3-bis(4-sulfobutyl)benzimidazolocarbocyanine (TDBC) and plasmonic silver-coated gold nanorods, confined within mesoporous silica shells, which facilitate the adsorption of the J-aggregates onto the metallic nanorod surface, while providing high colloidal stability. Electromagnetic simulations show that the electromagnetic field is strongly confined inside the J-aggregate layer, at wavelengths near the upper plexcitonic mode, but it is damped toward the J-aggregate/water interface at the lower plexcitonic mode. This behavior is ascribed to the sharp variation of dielectric properties of the J-aggregate shell close to the plasmon resonance, which leads to a high opposite refractive index contrast between water and the TDBC shell, at the upper and the lower plexcitonic modes. This behavior is responsible for the high SERS efficiency of the plexcitonic nanorattles under both 633 nm and 532 nm laser illumination. SERS analysis showed a detection sensitivity down to the single-nanoparticle level and, therefore, an exceptionally high average SERS intensity per particle. These findings may open new opportunities for ultrasensitive biosensing and bioimaging, as superbright and highly stable optical labels based on the strong coupling effect.</p>

Dataset of "Mn-doped WSe2 as an efficient electrocatalyst for hydrogen production and as anode material for lithium-ion batteries"
<p>The ongoing energy crisis has made it imperative to develop low-cost, easily fabricated, yet efficient materials. It is highly desirable for these nanomaterials to function effectively in multiple applications. Among transition metal dichalcogenides, tungsten diselenide (WSe2) shows great promise but remains understudied. In this work, we doped WSe2 with Mn using a simple hydrothermal method. The resulting material exhibited excellent electrocatalytic activity for the hydrogen evolution reaction, achieving a low overpotential of –0.28 V vs RHE at -10 mA/cm2, enhanced conductivity, and high stability and durability. Moreover, as an anode material in in lithium-ion batteries, the Mn-doped WSe2 outperformed pristine WSe2, reaching discharge and charge capacities of 1223 and 922 mAh g−1, respectively. Additionally, the Mn-doped material maintained a significantly higher discharge capacity of 201 mAh g−1 compared to intact WSe2, which had 68 mAh g−1 after 150 cycles. This work offers novel insights into designing efficient bifunctional nanomaterials using transition metal dichalcogenides.</p>

We performed 10 genome-wide association studies of resting state network measures of intrinsic brain activity in up to 36,150 participants of European ancestry in the UK Biobank. We found that the heritability of global network efficiency was largely explained by blood oxygen level-dependent (BOLD) resting state fluctuation amplitudes (RSFA), which are thought to reflect the vascular component of the BOLD signal. RSFA itself had a significant genetic component and we identified 24 genomic loci associated with RSFA, 157 genes whose predicted expression correlated with it, and 3 proteins in the dorsolateral prefrontal cortex and 4 in plasma. We observed correlations with cardiovascular traits, and single-cell RNA specificity analyses revealed enrichment of vascular related cells. Our analyses also revealed a potential role of lipid transport, store-operated calcium channel activity, and inositol 1,4,5-trisphosphate binding in resting-state BOLD fluctuations. We conclude that that the heritability of global network efficiency is largely explained by the vascular component of the BOLD response as ascertained by RSFA, which itself has a significant genetic component.</p> <p> </p> <p>Further information on the files uploaded here can be found in the README. Interagency Ecological Program and US Fish and Wildlife Service: Beach Seine Efficiency Study, collected by the Delta Juvenile Fish Monitoring Program, 2013-2018
Within the San Francisco Estuary, beach seining has been used since the 1970s by the U.S. Fish and Wildlife Service's Delta Juvenile Fish Monitoring Program (DJFMP) to monitor the relative abundance and distribution of juvenile Chinook salmon and other fishes of management concern occurring in near shore habitats. Over 2,000 seine samples are currently being collected throughout the San Francisco Estuary annually. Data from these surveys are used to make various water operation decisions and help inform managers on the status of fish populations within the San Francisco Estuary. By not accounting for the variability of species- and size-specific efficiency of beach seines across environmental gradients, the metrics developed by the DJFMP are likely underestimated and biased to an unknown extent. Consequently, the ability of the DJFMP to document the true occupancy of fishes for water operations and population vicissitude may be inhibited. Therefore, the first objective of this study is to estimate the species- and size-specific capture efficiency of DJFMP beach seines for fishes occurring in near shore habitats within the San Francisco Estuary. The second objective is to model the observed efficiency estimates using habitat variables to allow managers to account for incomplete detection when estimating fish abundance and occupancy from DJFMP beach seine data.
