Demographic data from long-term symbiont removal experiments with grasses and Epichloë fungal endophytes

This project was designed to understand the demographic effects of vertically transmitted fungal endophytes (Epichloë spp.) on their grass hosts. The experiment includes seven host-symbiont taxonomic pairs: Agrostis perennans - E. amarillans, Elymus villosus - E. elymi, Elymus virginicus - E. elymi or EviTG-1, Festuca subverticillata - E. starrii, Poa alsodes - E. alsodes, Poa sylvestris - E. PsyTG-1, Schedonorus arundinaceus - E. coenophiala. Experimental plots were established at the Indiana University Lilly-Dickey Woods Research and Teaching Preserve in south-central Indiana, USA in 2007. For each species, 5-10 plots were planted with naturally symbiotic (S+) hosts, and 5-10 plots were plated with hosts that were disinfected of fungal endophytes by heat treatment (S-). Over 15 years (2007-2022) we collected demographic data on the survival, growth, reproduction, and recruitment of all plants in all plots. Beginning in 2018 we also collected data on the locations of all plants in every plot.

Silicon and Epichloë-endophyte defences in a model temperate grass diminish feeding efficiency and immunity of an insect folivore

Plants deploy diverse anti-herbivore defences which reduce feeding and performance of herbivores. Temperate grasses use silicon (Si) accumulation and Epichloë-endophytes for physical and chemical (i.e. endophytic-alkaloids) defence against insect herbivores. Recent studies suggest that Epichloë-endophytes increase Si accumulation in their host grass. It is unknown, however, how this affects Si-deposition on the leaf surface, their impacts on insect herbivore feeding efficiency and their immunity to potential infection/parasitism.

To address this knowledge gap, we grew tall fescue (Festuca arundinacea) hydroponically with and without Si, in the absence or presence of the novel AR584 Epichloë-strain. We exposed plants to Helicoverpa armigera (Lepidoptera: Noctuidae) in both in-situ (intact leaves) and ex-situ (excised leaves) feeding trials and determined the effects of Si and endophyte defences on herbivore feeding efficiency, growth rates and immunity against potential infection/parasitism.

Endophytic plants supplied with Si showed 110% and 143% increases in leaf silica density and leaf Si concentrations, respectively, when exposed to herbivory, compared to non-endophytic plants that were herbivore-free. Despite the endophyte-mediated increases in Si concentrations, H. armigera was only affected by Si supply; growth rates decreased by 87% and most feeding efficiency indices decreased by at least 30%. Si supply also increased mandibular wear by 16%, which was negatively correlated with H. armigera growth rates. Cellular and humoral immunity of H. armigera were negatively affected by both Si and endophytes. Endophytic-loline alkaloid concentrations were unaffected by Si supply or herbivory, whereas herbivory increased peramine concentrations by 290%.

To our knowledge, this is the first report of Si defences and Epichloë-endophyte-derived alkaloids compromising insect immunity via reduced melanisation response. Using tall fescue and H. armigera, our study suggests that deploying both physical (i.e. Si accumulation) and chemical (i.e. endophytic-alkaloids) defences acting against multiple insect herbivore traits, including feeding efficiency, growth and immunity, may be a successful defence strategy in temperate grasses. This multi-faceted defence may be particularly difficult for insect herbivores to overcome.

The Fungal Endophyte Epichloë festucae var. lolii Plays a Limited Role in Mediating Crown Rust Severity in Perennial Ryegrass

Perennial ryegrass (Lolium perenne L.) is an important turf and forage species that often becomes infected with Puccinia coronata f. sp. lolii, the causal pathogen of crown rust. Disease control through clavicipitaceous endophytes has been proposed as a potential biocontrol. Two field experiments were designed to determine the influence of native Epichloë endophyte infection on natural rust development across a diverse panel of perennial ryegrass germplasm. Experiment 1 used an isogenic population design in which infected (E+) or endophyte-free (E−) clonal plants were nested within 14 perennial ryegrass entries. Experiment 2 consisted of isofrequent E+ and E− progeny from isogenic parents. Results showed the endophyte had no consistent main effect on crown rust severity across or within entries; however, several isogenic host pairs did show either favorable or antagonistic effects. Despite these sporadic effects, no differences were found between isofrequent family pairs, indicating the presence of a host  endophyte interaction. Genotypic and phenotypic data revealed that endophyte isolates were similar within entry, indicating that host genotype could be responsible for the highly specific endophyte effect on crown rust. The preponderance of host resistance in mediating rust severity was supported by large broad (>.75) and narrow-sense (.76) heritability estimates. These findings support the conclusion that endophyte infection does not play a substantial role in mediating crown rust severity on a population scale in perennial ryegrass.
