Visual-Evoked Potential (VEP) Event-Related Files from the General Anesthesia and Brain Activity (GABA) Study and Infant Sibling Project (ISP)
<p>HAPPE+ER software was optimized for developmental data using a subset of EEG files from 4-month and 10-month old infants in the General Anesthesia and Brain Activity (GABA) Study. While medically necessary, 1-2 million infants each year undergo general anesthesia – a process that sedates brain activity and impacts early sensory experiences during a time typically characterized by rapid neurocognitive development. The GABA study examines sensory and socioemotional neurodevelopment longitudinally from infancy through childhood in individuals who have and who have never undergone general anesthesia during different windows in the first year of life. The GABA study was carried out in accordance with the recommendations of the Institutional Review Board at Boston Children’s Hospital. All caregivers provided assent for their child’s participation in the GABA study and for the release of the deidentified data. </p> <p>To facilitate the use and understanding of HAPPE+ER software, we have provided a subset of the validation files from the GABA study to serve as a tutorial dataset for how to run event-related potential (ERP) data through this automated processing pipeline. Five files (a.raw - e.raw) are from four 4-month and one 10-month old infants during a pattern reversal visual-evoked potential (VEP) paradigm. Pattern reversal occurred every 500 milliseconds. The pattern stimulus onset is indicated in each file by the code: vep+. Data was collected using a 128-channel EGI HydroCel Geodesic Sensor Net and EGI Net Amps 400, sampled at 1000Hz with an online reference to channel CZ. </p> <p>We have also included a subset of files from the Infant Sibling Project (ISP), an investigation examining infants at high versus low familial risk for autism spectrum disorder over the first 3 years of life. Baseline EEG data was collected while a young child sat in a parent’s lap watching a research assistant blow bubbles or show toys for several minutes. The Infant Sibling Project was carried out in accordance with the recommendations of the Institutional Review Board at Boston University and Boston Children’s Hospital (#X06-08-0374), with written informed consent from all caregivers prior to their child’s participation in the study. All files here have been deidentified, including alteration of exact acquisition dates. Acquisition times have not been altered. For additional information about data collection paradigms, and sample studies published on the larger ISP data set, please see the following references:</p> <ol> <li>Levin, A. R., Varcin, K. To create these files, we selected a subset of 39 spatially-distributed channels in the baseline EEG files and created sixteen 30-second files using continuous segments of relatively artifact-free (clean) baseline data from the full-length files. Next, from 30-second sections of the same individuals’ EEG that were artifact-laden, we ran ICA and extracted artifact independent components (identified by an expert and labeled artifact by both ICLabel and MARA automated algorithms). We inserted the artifact ICs into that individual’s clean 30-second data segment to create an additional 16 artifact-added files. We then selected a channel from a simulated VEP dataset (included here as simulated_full.set) with a stereotyped and prominent simulated VEP waveform, in this case Oz, and added its timeseries (included here as simulated_singleChan.set) to each channel of the clean and artifact-added files to create two VEP datasets with a known ERP morphology (sim-artifact_a-p and sim-clean_a-p). For additional information about the creation of this simulated data and VEP data with a known ERP morphology, please refer to Monachino et al., in revision; DOI: https://doi.org/10.1101/2021.07.02.450946.</p> <p>Additional files included below are the HAPPE+ER data and pipeline quality metric output spreadsheets for the five GABA study data files for an example run, the output spreadsheet containing the ERP timeseries from the generateERPs script, the .mat file containing the parameter settings for HAPPE+ER for that run, an Excel file with the bad channels for each file, and a tutorial document illustrating the results of this example run. </p>

The files were, however, renamed for convenience. Video stream parameters: MPEG-4 (DIVX) codec, color space yuv420p, 1280x512 pixels, 30 fps. Along with each video file, there is an associated text file (“.txt”) containing the metadata of video recording and the timestamps of hardware state changes. In these files, unmodified after creation by the EthoVision software, the status of hardware TTL inputs was logged whenever a change of state of these inputs was detected. Typically, “input 2” was sampling the gating signal from the Master-8 pulse generator, with the "high" signal level indicating the application of train of light pulses. This hardware state, signaling the presence of the light train, is noted in the individual metadata files; </li> <li>widefield fluorescent images of single coronal sections containing the MeApd (Figures 2, S2) were converted from the proprietary format of Olympus slide scanning microscope into composite TIFF format, readable by FIJI/ImageJ (<a href="https://fiji.sc/">https://fiji.sc/</a> or <a href="https://imagej.net/Fiji/Downloads">https://imagej.net/Fiji/Downloads</a>). The information on pixel resolution and inter-section distance is embedded in the individual image files as TIFF metadata. Attribution of fluorescent probes to the color channels is given in the corresponding metadata files.</li> <li>confocal fluorescent image stacks acquired from single coronal sections containing MeApd (Figure S3) are provided in composite TIFF format after stitching the tiles (originally stored as “lsm” format; Carl Zeiss) using a stitching plugin (Preibisch et al., Bioinformatics 2009) in FIJI. The information on pixel resolution is embedded inside the individual image files as TIFF metadata. Attribution of fluorescent probes to the color channels is given in the corresponding metadata file. For each stack, a region of interest (ROI) highlighting the borders of the MeApd is provided as a separate file in a “.roi“ format (FIJI).</li> <li>patch clamp recordings (Figures 4, S4) are provided as “.dat” files, unmodified from the original version created by the acquisition software PatchMaster (HEKA Elektronik, Germany). Fitness costs associated with a GABA receptor mutation conferring dieldrin resistance in Aedes albopictus
<p><span>Understanding the dynamics of insecticide resistance genes in mosquito populations is pivotal for a sustainable use of insecticides. Dieldrin resistance in <em>Aedes</em> <em>albopictus</em> is conferred by the alanine to serine substitution (A302S or <em>RdlR</em> allele) in the γ-aminobutyric acid (GABA) receptor encoded by the <em>Rdl</em> gene. On Reunion Island, dieldrin resistance was initially reported in natural <em>Ae</em>. <em>albopictus</em> populations sampled in 2008 despite the ban of dieldrin since 1994. To monitor insecticide resistance in <em>Ae</em>. <em>albopictus</em> on the island and to identify its drivers, we measured (i) the frequency of resistance alleles in 19 distinct natural populations collected between 2016 and 2017, (ii) fitness costs associated with dieldrin resistance in laboratory-controlled experiments, and (iii) the resistance conferred by </span><span><em>RdlR</em></span><span> to fipronil, an insecticide widely used on the island and reported to cross-react with </span><span><em>RdlR</em></span><span>. The results show a persistence of </span><span><em>RdlR</em></span><span> in <em>Ae</em>. <em>albopictus</em> natural populations at low frequencies. Among the measured life history traits, mortality in pre-imaginal stages, adults' survival as well as the proportion of egg-laying females were significantly affected in resistant mosquitoes. Finally, bioassays revealed resistance of </span><span><em>RdlR</em></span><span> mosquitoes to fipronil, suggesting that the use of fipronil <em>in</em> <em>natura</em> could select for the </span><span><em>RdlR</em></span><span> allele. This study shows that dieldrin resistance is persistent in natural mosquito populations likely as a result of combined effects between fitness costs associated with </span><span><em>RdlR</em></span><span> and selection exerted by cross-reacting environmental insecticides such as fipronil.</span></p>

Source data for: Presynaptic NMDARs cooperate with local spikes toward GABA release from the reciprocal olfactory bulb granule cell spine
<p><span>In the rodent olfactory bulb the smooth dendrites of the principal glutamatergic mitral cells (MCs) form reciprocal dendrodendritic synapses with large spines on GABAergic granule cells (GC), where unitary release of glutamate can trigger postsynaptic local activation of voltage-gated Na+-channels (Navs), i.e. a spine spike. Can such single MC input evoke reciprocal release? We find that unitary-like activation via two-photon uncaging of glutamate causes GC spines to release GABA both synchronously and asynchronously onto MC dendrites. This release indeed requires activation of Navs and high-voltage-activated Ca2+-channels (HVACCs), but also of NMDA receptors (NMDAR). Simulations show temporally overlapping HVACC- and NMDAR-mediated Ca2+-currents during the spine spike, and ultrastructural data prove NMDAR presence within the GABAergic presynapse. The cooperative action of presynaptic NMDARs allows to implement synapse-specific, activity-dependent lateral inhibition and thus could provide an efficient solution to combinatorial percept synthesis in a sensory system with many receptor channels. </span></p>
