Disturbance history alters the development of the HPA axis in altricial nestling birds

<p>Glucocorticoid (GC) hormones regulate the vertebrate stress response and are secreted by the hypothalamic–pituitary–adrenal (HPA) axis. Acute elevation of GCs is thought be adaptive because it promotes physiological and behavioural changes that allow animals to cope with disturbances. In contrast, chronic elevation of GCs is associated with reduced body condition, immune function, reproductive success, and survival. In adult birds, the effects of chronic stress have been well documented, including human-related disturbances. In contrast, the effects of chronic stress on nestlings have rarely been addressed. This is of interest, as many ecological or monitoring studies of wild birds require nestlings to be regularly handled. However, the consequences of repeated handling of nestlings on HPA axis function and body mass o remain poorly quantified. We examined whether daily exposure to handling stress increased corticosterone (the dominant avian glucocorticoid) secretion and reduced pre-fledging body mass relative to undisturbed control nestlings of the Noisy Miner (<em>Manorina melanocephala), </em>a native Australian passerine bird. Daily handling resulted in an elevated baseline and attenuated stress-induced corticosterone levels in disturbed 14-day nestlings, in comparison with control nestlings handled for the first time. Despite this, disturbed and control nestlings fledged at a similar body mass. Baseline and stress-induced corticosterone increased with nestling age but remained independent of nestling sex and hatching order. These findings suggest that chronic stress causes physiological alterations to the development of the HPA axis in nestling birds, in turn suggesting that researchers should minimise or account for handling stress in their experiments. These data also raise the possibility that other chronic stressors may have physiological consequences for the development of the HPA axis in nestling birds.  </p>

Data from: Efficacy of negative feedback in the HPA axis predicts recovery from acute challenges.

The glucocorticoid stress response mediates a suite of physiological and behavioral changes that allow vertebrates to cope with transient stressors. Chronically elevated glucocorticoid levels are known to result in a variety of organismal costs, but relatively little is known about the downstream effects of mounting a series of brief, acute spikes in circulating glucocorticoids. Conceptual models of stress suggest that repeated acute stressors might produce 'wear-and-tear' on the stress response system when encountered in sequence. We used a novel technique to experimentally induce acute corticosterone spikes on either three or six consecutive days in incubating tree swallows. Consistent with the 'wear-and-tear' hypothesis, we found that i) a sequence of corticosterone spikes produced cumulative effects on corticosterone regulation, ii) treatment frequency predicted the severity of consequences, and iii) individual variation in the ability to terminate the stress response through negative feedback predicted the duration of physiological disruption in the group that experienced the most frequent challenges. Our results illustrate the importance of assessing multiple aspects of the hormonal stress response and have implications for understanding both individual and population resilience to repeated transient stressors.

Data from: Experimental effects of early-life corticosterone on the HPA axis and pre-migratory behaviour in a wild songbird

1.Although laboratory studies have shown that chronic exposure to elevated glucocorticoids during development can have profound effects on the physiology and behaviour of animals, we still have a poor understanding of the proximate and ultimate consequences of early-life stress on individuals in the wild. 2. In an island population of Savannah sparrows (Passerculus sandwichensis), we examined multiple hypotheses to explain how elevated glucocorticoid exposure during the nestling period influenced both hypothalamic-pituitary-adrenal (HPA) axis function during the nestling period and the subsequent movement and survival of young after they fledged the nest. 3. We exposed nestlings to exogenous corticosterone from 2-6 d of age and then measured both baseline and stress-induced (30 min restraint) plasma corticosterone levels prior to fledging (d 7). We then recaptured young throughout the pre-migratory period and used mark-recapture analysis to estimate temporary emigration from the capture site (movement) and survival. 4. Corticosterone-treated nestlings had higher baseline corticosterone levels and lower stress reactivity than untreated individuals, and were more sensitive to inclement weather. Although there was no evidence that corticosterone treatments influenced survival, treated individuals had higher rates of temporary emigration outside of the study site than sham or controls. 5. Our results provide support for the ceiling hypothesis, which suggests that individuals with chronic elevated glucocorticoids can lead to a dampened HPA axis response. We also provide support for the CORT-activity hypothesis, which suggests that elevated glucocorticoids can increase activity levels, at least 1-2 months after leaving the nest. Our study highlights the importance of tracking individuals across multiple stages of the annual cycle to understand how early life events carry-over to influence both physiology and behaviour.
