Data from: Digital twin mathematical models suggest individualized hemorrhagic shock resuscitation strategies
<p><strong>Background:</strong> Optimizing resuscitation to reduce inflammation and organ dysfunction following human trauma-associated hemorrhagic shock is a major clinical hurdle. This is limited by the short duration of pre-clinical studies and the sparsity of early data in the clinical setting.</p> <p><strong>Methods:</strong> We sought to bridge this gap by linking preclinical data in the porcine model with clinical data from patients from the Prospective, Observational, Multicenter, Major Trauma Transfusion (PROMMTT) study via a three-compartment ordinary differential equation model of inflammation and coagulation.</p> <p><strong>Results:</strong> The model accurately predicts physiologic, inflammatory, and laboratory measures in both the porcine model and patients, as well as the outcome and time of death in the PROMMTT cohort. Model simulation suggests that resuscitation with plasma and red blood cells outperformed resuscitation with crystalloid or plasma alone, and that earlier plasma resuscitation reduced injury severity and increased survival time.</p> <p><strong>Conclusions:</strong> This workflow may serve as a translational bridge from pre-clinical to clinical studies in trauma-associated hemorrhagic shock and other complex disease settings.</p>

Data from: the characteristics and treatment for severe postpartum hemorrhage in different midwifery hospitals in one district of Beijing in China: an institution-based, retrospective cohort study
<p><span><strong>Objective:</strong> </span><span>To identify the characteristics and treatment approaches for Severe Postpartum Hemorrhage (SPPH) patients in various midwifery institutions in one district in Beijing, especially those without identifiable antenatal PPH risk factors, to improve regional SPPH rescue capacity.</span></p> <p><strong><span>Design:</span></strong><span> Retrospective cohort study</span></p> <p><span><strong>Setting:</strong> </span><span>This study was conducted at n</span><span>ine tertiary-level hospitals and ten secondary-level hospitals</span><span> in Haidian district of Beijing from January 2019 to December 2022. </span></p> <p><strong><span>Participants:</span></strong> <span>The major inclusion criterion was SPPH cases with blood loss </span>≥<span>1500 ml or needing a packed blood product transfusion </span>≥<span>1000 ml within 24 h after birth</span><span>.</span><span> A total of 324 mothers suffering from SPPH were reported to the Regional Obstetric Quality Control Office from 19 midwifery hospitals. </span></p> <p><span><strong>Outcome measures:</strong> </span><span>The pregnancy characteristics collected included: age at delivery, gestational weeks at delivery, height, parity, delivery mode, antenatal PPH risks, etiology of PPH, bleeding amount, PPH complications, transfusion amount, and PPH management. SPPH characteristics were compared between two levels of midwifery hospitals and their association with antenatal PPH high-risk factors was determined.</span></p> <p><span><strong>Results:</strong> </span><span>SPPH was observed in 324 mothers out of 106697 mothers in the four years. There were 74.4% and 23.9% cases of SPPH without detectable antenatal PPH high-risk factors in secondary and tertiary midwifery hospitals, respectively. Primary uterine atony was the leading cause of SPPH in secondary midwifery hospitals, whereas placental-associated disorders were the leading causes in tertiary institutions, accounting for over 50% of cases. In all SPPH cases, the rates of red blood cell transfusion over 10U, </span><span>unscheduled returns to the operating room,</span><span> and adverse PPH complications were higher in patients without antenatal PPH risk factors. Secondary hospitals had significantly higher rates of trauma compared with tertiary institutions.</span></p> <p><span><strong>Conclusion:</strong> </span><span>Examining SPPH cases at various institutional levels offers a more comprehensive view of regional SPPH management and enhances targeted training in this area.</span></p>

Data from: Isolation of diverse simian arteriviruses causing hemorrhagic disease
<p>Genetically diverse simian arteriviruses (simarteriviruses) naturally infect geographically and phylogenetically diverse monkeys, and cross-species transmission and emergence are of considerable concern. Characterization of most simarteriviruses beyond sequence analysis has not been possible because the viruses fail to propagate in the laboratory. We attempted to isolate 4 simarteriviruses, Kibale red colobus virus 1, Pebjah virus, simian hemorrhagic fever virus, and Southwest baboon virus 1, by inoculating an immortalized grivet cell line (known to replicate simian hemorrhagic fever virus), primary macaque cells, macrophages derived from macaque induced pluripotent stem cells, and mice engrafted with macaque CD34+-enriched hematopoietic stem cells. The combined effort resulted in successful virus isolation; however, no single approach was successful for all 4 simarteriviruses. We describe several approaches that might be used to isolate additional simarteriviruses for phenotypic characterization. Our results will expedite laboratory studies of simarteriviruses to elucidate virus-host interactions, assess zoonotic risk, and develop medical countermeasures.</p>
Skin eruptions in acute hemorrhagic edema of young children: systematic review of the literature
<p><strong>Background:</strong> Acute hemorrhagic edema is a skin limited small-vessel leukocytoclastic vasculitis, which affects infants 4 weeks to 2 years of age and remits within 3 weeks. The diagnosis is made clinically in not-ill appearing children with acute onset of raised annular or nummular eruptions and edema. In this vasculitis, type, distribution and evolution of the rash have never been systemically investigated. To address this issue, we employed the data contained in the Acute Hemorrhagic Edema Bibliographic Database, which incorporates all reports on acute hemorrhagic edema. <strong>Summary:</strong> Key features of rash were documented in 383 children. Annular eruptions in strict sense, usually targetoid, were reported in 375 (98%) cases (many children also presented polycyclic or arciform eruptions). Nummular eruptions were also very common (N=358; 93%). Purpuric eruptions and ecchymoses were reported in the vast majority of cases. Macules and wheals were described in a minority of cases. Edema, detected in all cases, was mostly painful, indurated and non-pitting. Following regions were affected, in decreasing order, by annular or nummular eruptions: legs, feet, face, arms, ears, trunk, and genitals. With the exception of feet, which were very often affected, the same distribution was reported for edema. The initial eruption was often a wheal or a macule that evolved into a nummular or an annular eruption. Nummular eruptions successively evolved into annular ones<strong>. Key message</strong>: this study carefully characterizes type, distribution and evolution of skin eruption in acute hemorrhagic edema. The data help physicians to rapidly and noninvasively make the clinical diagnosis of this vasculitis.</p>
Normalised absorbance ratios of IgG for myxoma and rabbit hemorrhagic disease viruses: Longitudinal dataset wild European rabbits
<p>This study addresses the long-term dynamics of humoral immunity to two viral pathogens of the European rabbit, an endangered keystone species of major ecological relevance. Such studies are particularly challenging in free-ranging species, and a combination of longitudinal capture-mark-recapture and semi-quantitative serology was used to address this question. Over 600 normalised absorbance ratios of indirect ELISA, obtained from 505 individual rabbits in 7 populations over five years were analysed using linear mixed models. The results support a lifelong acquired humoral immunity to myxoma virus and rabbit hemorrhagic disease virus upon natural infection and suggest the presence of maternal immunity to the latter in wild juvenile rabbits. These results contribute to understanding the epidemiology of two viral diseases threatening this keystone species and assist in developing conservation programs.</p>

Hemostasis defects underlying the hemorrhagic syndrome caused by Mammarenaviruses in a cynomolgus macaque model
<p>Abstract: Viral hemorrhagic fevers (HF) are a group of acute febrile diseases with high mortality rates. While hemostatic dysfunction appears to be a major determinant of the severity of the disease, it is still unclear what pathogenic mechanisms lead to it. In clinical studies, arenaviruses such as Lassa, Machupo and Guanarito viruses caused HF that vary in symptoms and biological alterations.</p> <p>In this study we aimed to characterize the hemostatic dysfunction induced by arenaviral HF to determine its implication in the severity of the disease and to elucidate the origin of this syndrome.</p> <p>We found that lethal infection with Machupo, Guanarito and Lassa viruses is associated with cutaneo-mucosal, cerebral, digestive and pulmonary hemorrhages. The affected animals developed a severe alteration of the coagulation system, which was concomitant with acute hepatitis, minor deficit of hepatic factor synthesis, presence of a plasmatic inhibitor of coagulation and dysfunction of the fibrinolytic system. Despite signs of increased vascular permeability, endothelial cell infection was not a determinant factor of the hemorrhagic syndrome. There were also alterations of the primary hemostasis during lethal infection, with moderate to severe thrombocytopenia and platelet dysfunction. Finally, we show that lethal infection is accompanied by a reduced hematopoietic potential of the bone marrow. This study provides an unprecedented characterization of the hemostasis defects induced by several highly pathogenic Arenaviruses.</p> <p>Transcriptomics data presented in Figure 3 and in Supp Figure 2</p>
