Data from: Sustained neurotrophin-3 delivery from hyaluronic acid hydrogels for neural tissue regeneration

<p>The goal of this work was to design a polymer-based platform capable of localized, long-term delivery of biologically active neurotropic factors using an affinity-based approach. Here, we synthesized hyaluronic acid-methylfuran (HA-mF) hydrogels that provide sustained, affinity-based release of neurotrophin-3 (NT-3), a growth factor that promotes axon growth for 28 days. A Diels-Alder crosslinking reaction between HA-mF and polyethylene glycol (PEG)-dimaleimide occurs within 15 minutes under physiological conditions, resulting in injectable hydrogels that can be polymerized in the presence of cells and growth factors. We also tuned the hydrogel's storage modulus to match that of native rat spinal cord tissue, providing a platform not only for localized drug delivery but also a suitable vehicle for cellular transplantation. The NT-3 released from the HAmF hydrogels remains bioactive for at least 14 days, promoting axonal growth from primary sensory neurons as well as stem cell-derived V2a interneurons and motoneurons <em>in vitro</em>. The hydrogels also supported cell growth allowing for 3-dimensional axonal extensions within the scaffold matrix. Here we confirm the protective role of HA-mF on matrix-bound NT-3 activity and show that these hydrogels are an excellent platform for drug delivery for neural applications.</p>

Schiff Base Crosslinked Hyaluronic Acid Hydrogels with Tunable and Cell Instructive Time-Dependent Mechanical Properties

<p>This folder contains raw data for the paper titled "<strong>Schiff Base Crosslinked Hyaluronic Acid Hydrogels</strong><strong> with Tunable and Cell Instructive Time-Dependent Mechanical Properties</strong>" authored by Taha Behroozi Kohlan, Yanru Wen, Carina Milena Mini, and Anna Finne-Wistrand, published in <em>Carbohydrate Polymers</em> journal. The raw data presented here contains NMR, FTIR, SEC, swelling and stability, and rheology data used to create figures.</p> <p><strong>Abstract:</strong></p> <p>The dynamic interplay between cells and their native extracellular matrix (ECM) influences cellular behavior, imposing a challenge in biomaterial design. Dynamic covalent hydrogels are viscoelastic and show self-healing ability, making them a potential scaffold for recapitulating native ECM properties. We aimed to implement kinetically and thermodynamically distinct crosslinkers to prepare self-healing dynamic hydrogels to explore the arising properties and their effects on cellular behavior. To do so, aldehyde-substituted hyaluronic acid (HA) was synthesized to generate imine, hydrazone, and oxime crosslinked dynamic covalent hydrogels. Differences in equilibrium constants of these bonds yielded distinct properties including stiffness, stress relaxation, and self-healing ability. The effects of degree of substitution (DS), polymer concentration, crosslinker to aldehyde ratio, and crosslinker functionality on hydrogel properties were evaluated. The self-healing ability of hydrogels was investigated on samples of the same and different crosslinkers and DS to obtain hydrogels with gradient properties. Subsequently, human dermal fibroblasts were cultured in 2D and 3D to assess the cellular response considering the dynamic properties of the hydrogels. Moreover, assessing cell spreading and morphology on hydrogels having similar modulus but different stress relaxation rates showed the effects of matrix viscoelasticity with higher cell spreading in slower relaxing hydrogels.</p>

Dataset_Ultrasound guided hip injections with high density hyaluronic acid: outcome at one year follow up.

<p><span>The ultrasound-guided viscosupplementation of the hip joint with hyaluronic acid (HA) is considered a standard procedure among the conservative treatments for hip arthritis. The aim of this study was to evaluate the clinical benefit and the incidence of adverse events of the technique in an observational study at one year follow up. Methods:<span>We</span><strong><span> </span></strong><span>evaluated a consecutive series of 85 patients with a diagnosis of symptomatic arthritis who underwent intra-articular ultrasound-guided hyaluronic acid injections. The scales used for evaluation were </span></span><span>modified Harris Hip Score (mHHS), WOMAC</span><span>, and </span><span>Hip Outcome Score (HOS) with subscale Sport (HOSs), for pain the Visual Analogic Scale (VAS). The patients were classified according to Tonnis&rsquo; radiological classification of arthritis (range 0&ndash;3): 20 patients (grade 0), 32 (grade 1), 18 (grade 2), 15 (grade 3). Results:At last follow up, all the scales</span><span> increased: mHHS from 59.35 to 82.1, HOS from 69.45 to 78.53, HOss from 47.4 to 58.11, VAS from 6.09 to 3.97, WOMAC from 33.2 to 31.5 (<em>p </em>&lt;<em> </em>0.05 for all the parameters); the results were elaborated with GraphPad Prism v5.0 (Prism Software La Jolla, CA, USA) using Wilcoxon&rsquo;s test. A total of </span><span>13 patients out of 85 needed arthroplasty, all classified as Tonnis grade 3.</span><span> No serious adverse events were noted due to the procedure.Conclusion:</span><span> <span>Based</span><strong> </strong>on our findings, indication for the use of hyaluronic acid is limited to patients with mild to moderate arthritis. Patients in advanced arthritis refusing replacement surgery and asking for this treatment should be informed about the poor results of the technique even in the short term.</span></p>
