The Regulation of Microglia Activity and the Production of IL-1α and IL-6 in the Degenerated Retina by Mesenchymal Stem Cells
<p><span>Activation of immune response and production of proinflammatory factors plays an important role in the development and progression of retinal degenerative diseases (RDD). For this reason, focusing on immunomodulation is potential option for the efficient ophthalmological therapy. Mesenchymal stem cells (MSCs) have been study in the treatment of RDD mainly due to their regenerative and neuroprotective actions. Nevertheless, MSCs also possess several immunomodulatory properties. Our study shows that NaIO<sub>3</sub>-induced degeneration increased <em>in vitro</em> and <em>in vivo</em> expression of genes for Interleukin (IL)-1α and IL-6 in the mouse retinal tissue. In addition, intraperitoneal application of NaIO<sub>3</sub> increased expression of gene for Iba-1 and infiltration of CD45<sup>+</sup>CD11b<sup>+</sup> cells into the retina. CD45<sup>+</sup> population was also responsible for the production of IL-1α while IL-6 was produced by CD45<sup>-</sup> cells. Cocultivation of degenerated retina in the presence of MSCs decreased the expression and production of both studied cytokines and expression of gene for Iba-1 in the retinal tissue. On contrary, it was observed that MSCs treated with supernatant from degenerated retina increased the expression of genes for cyclooxygenase-2, transforming growth factor-β, programmed-death ligand 1 and nerve growth factor. These results show that MSCs are able to regulate immune reaction in the degenerated retinal environment.</span></p>
Interleukin-6 (IL-6) activates the NOTCH1 signaling pathway through E-proteins in endometriotic lesions
<p><b>Context:</b> NOTCH signaling is activated in endometriotic lesions but the exact mechanisms remains unclear. Interleukin-6 (IL-6), which is increased in the peritoneal fluid of women with endometriosis, induces NOTCH1 through E-proteins including E2A and HEB in cancer development.</p> <p><b>Objective:</b> To study the role of E-proteins in inducing NOTCH1 expression under the regulation of IL-6 in endometriosis.</p> <p><b>Setting and Design:</b> The expression of E2A, HEB and NOTCH1 was first investigated in endometrium of women with endometriosis and the baboon model of endometriosis. Regulation of E-proteins and NOTCH1 expression was examined after IL-6 stimulation and siRNA mediated inhibition of E2A or/and HEB in human endometriotic epithelial cells (12Z) <i>in vitro</i>, and subsequent following <i>in vivo </i>IL-6 treatment in the mouse model of endometriosis <i>in vivo</i>.</p> <p><b>Results: </b>E2A, HEB and NOTCH1 were significantly upregulated in glandular epithelium (GE) of ectopic endometrium compared to eutopic endometrium in both women and baboon model. IL-6 treatment upregulated the expression of NOTCH1 together with E2A and HEB in 12Z cells. siRNA inhibition of E2A and HEB or HEB alone decreased NOTCH1 expression. Binding efficiency of both E2A and HEB was significantly higher at the binding sites on the human NOTCH1 promoter after IL-6 treatment. Finally, IL-6 treatment resulted in a significantly increased number of endometriotic lesions along with the increased expression of E2A, HEB and NOTCH1 in GE of the lesions compared with the vehicle treated group in an endometriosis mouse model.</p> <p><span><b>Conclusions:</b> IL-6 induced NOTCH1 expression is mediated by E-proteins in the ectopic GE cells, which may promote endometriotic lesion development. </span></p>
An intron SNP rs2069837 in IL-6 is associated with osteonecrosis of the femoral head development
<p>Two variants (rs2069837, and rs13306435) in the IL-6 gene were identified and genotyped from 566 patients with ONFH and 566 healthy controls. The associations between IL-6 polymorphisms and ONFH susceptibility were assessed using odds ratio (OR) and 95% confidence interval (95% CI) via logistic regression. The results of the overall analysis revealed that IL-6 rs2069837 is correlated with decreased risk of ONFH among the Chinese Han population (<em>p</em> < 0.05). In stratified analysis, rs2069837 also reduced the susceptibility to ONFH in older people (> 51 years), males, nonsmokers, and nondrinkers (<em>p</em> < 0.05).</p>

Maternal and perinatal obesity induce bronchial obstruction and pulmonary hypertension via IL-6-FoxO1-axis in later life
<p>Obesity is a pre-disposing condition for chronic obstructive pulmonary disease, asthma, and pulmonary arterial hypertension. Accumulating evidence suggests that metabolic influences during development can determine chronic lung diseases (CLD). We demonstrate that maternal obesity causes early metabolic disorder in the offspring. Here, interleukin-6 induced bronchial and microvascular smooth muscle cell (SMC) hyperproliferation and increased airway and pulmonary vascular resistance. The key anti-proliferative transcription factor FoxO1 was inactivated via nuclear exclusion. These findings were confirmed using primary SMC treated with interleukin-6 and pharmacological FoxO1 inhibition as well as genetic FoxO1 ablation and constitutive activation. In vivo, we reproduced the structural and functional alterations in offspring of obese dams via the SMC-specific ablation of FoxO1. The reconstitution of FoxO1 using IL-6-deficient mice and pharmacological treatment did not protect against metabolic disorder but prevented SMC hyperproliferation. In human observational studies, childhood obesity was associated with reduced forced expiratory volume in 1s/forced vital capacity ratio Z-score (used as proxy for lung function) and asthma. We conclude that the interleukin-6-FoxO1 pathway in SMC is a molecular mechanism by which perinatal obesity programs the bronchial and vascular structures and functions, thereby driving CLD development. Thus, FoxO1 reconstitution provides a potential therapeutic option for preventing this metabolic programming of CLD.</p>
