ERK activity in migrating MDA-MB-231 cells (clover-ERK-KTR + sir-DNA)
<p>MDA-MB-231 or U2OS cells stably expressing clover-ERK-KTR were seeded on fibronectin-coated (1 µg /ml) Ibidi 8-well slides (Ibidi) 1 day before imaging. Four hours before imaging, the medium was supplemented with 250 nM sir-DNA (Cytoskeleton) and 25 mM HEPES (Sigma). Cells were then imaged live (37 °C, 5% CO<sub>2</sub>) using a Nikon Eclipse Ti2-E microscope (Nikon) equipped with an sCMOS Orca Flash4.0 camera (Hamamatsu) and controlled by the NIS-Elements software (Nikon, v 5.11.01). MDA-MB-231 cells were imaged using a 20× Nikon CFI Plan Apo Lambda objective (NA 0.75), either 1 frame per minute for 2 hours or 1 frame every 5 minutes for 17 hours. In these experiments, a camera binning of 2 × 2 was used. </p> <p>This dataset consists of 20 videos.</p>

<p>Three scientific databases (PubMed, Scopus, and Web of Science (WoS)) were consulted (July 14, 2022) to assess the frequency of eight nomenclatural forms of malondialdehyde (MDA). Due to the peculiarities of the search interface of the selected databases, PubMed was searched in the Title and Abstract fields (search query example in PubMed: "malone dialdehyde"[Title/Abstract]), Scopus was searched in the Title, Abstract, and Keywords fields (search query example in Scopus: TITLE-ABS-KEY (“malone dialdehyde”)), and WoS Core Collection was searched in the Title, Abstract, Author keywords, and Keywords Plus fields (search query example in WoS Core Collection: TS=(“malone dialdehyde”)). All types of publications for the years 2002-2021 are taken into account.</p>

<p>Considerable amount of ligands will be consumed in the dual luciferase assay (DLA) which will be used for the screening of ALK2 inhibitors and off-target inhibition of ALK5 routinely. Ligands are costly to procure. In order to minimise wastage, it is important to determine the least amount of ligands needed to achieve optimal stimulation of ALK2 and ALK5. In these experiments, the activation of ALK2 and ALK5 by different concentrations of ligands in MDA-MB-231 reporter cells was analysed in Western Blot and DLA.</p>
