Physarum polycephalum Repeated Maze Honors Thesis, Tulane University, SE Louisiana 2024-2025

We conducted an experimental maze study from January to March, 2025, on Physarum polycephalum at Tulane University in New Orleans, Louisiana. We collected data on changes in locomotive behavior as P. polycephalum repeatedly solved the same maze. From daily photographs of growth, we recorded path choice, contamination presence and location, number of times P. polycephalum grew directly over maze walls, how many of the four dead-ends P. polycephlaum grew down, and efficiency through surface area covered. Data collection is complete. We found a significant increase in efficiency, and a significant decrease in both dead-end paths and wall jumping, as maze repetitions increased. This points to evidence of information storage and retrieval, and therefore cognitive processes such as memory, within the single-cellular protist Physarum polycephalum.

Data from: Texas field crickets (Gryllus texensis) use visual cues to place learn but perform poorly when intra- and extra-maze cues conflict

<p>Central place foraging field crickets are an ideal system for studying the adaptive value of learning and memory, but more research is needed on ecology-relevant cognition in these invertebrates. Here, we test the visuospatial place learning of Texas field crickets (<em>Gryllus texensis</em>) in a radial arm maze. Our study expands previous work on <em>G. texensis</em> cognition for accuracy measures and extends our previous findings on females to both sexes. Additionally, our study examines whether crickets use intra- or extra-maze cues to locate a food reward using a maze rotation putting the cues in conflict. We found that male and female crickets improved performance over trials when measured by accuracy variables but not latency variables; thigmotaxis negatively impacted performance in both sexes. In a reward-absent trial, both male and female crickets demonstrated place memory. When intra- and extra-maze cues conflicted during a rotation trial, crickets' performance was not better than chance. Our rotation results suggest that crickets may experience reciprocal overshadowing of conflicting cues – a result most often seen in other taxa with conflicting multi-modal cues. We conclude that crickets do not rely solely on: (1) a single-cue association; (2) route-following; or (3) their own scent cues to navigate the maze. Instead, male and female Texas field crickets seem to learn the location of the reward using a combination of proximal and distal cues. The possibility to test large numbers of wild-caught or laboratory-reared individuals opens the door to future investigations on the evolutionary ecology of visuospatial learning in these invertebrates.</p>

Liquid metal solves maze

<p>These are supplementary videos for the paper ``Liquid Metal Solves Mazes&#39;. The abstract of the paper is below.&nbsp;</p> <p>&nbsp;</p> <p>A room temperature liquid metal is a metal whose melting point is around room temperature. We use liquid metal gallium due to its non-toxicity. A physical maze is a a connected set of Euclidean domains separated by impassable walls. We demonstrate that a maze filled with sodium hydroxide is solved by a gallium droplet when direct current is applied between start and destination loci. During the maze solving the droplet stays compact due to its large surface tension, navigate along lines of the highest electrical current due its high electrical conductivity, and goes around corners of the maze&#39;s corridors due to its high flexibility. The &nbsp;droplet maze solver has a long life-time due to negligible vapour of the liquid gallium.</p>

Behavior data from T-maze assay

<p>Dietary magnesium (Mg2+) supplementation can enhance memory in young and aged rats. Memory-enhancing capacity was largely ascribed to increases in hippocampal synaptic density and elevated expression of the NR2B subunit of the NMDA-type glutamate receptor. Here we show that Mg2+ feeding also enhances long-term memory in <i>Drosophila</i>. Normal and Mg2+ enhanced fly memory appears independent of NMDA receptors in the mushroom body and instead requires expression of a conserved<i> </i>CNNM-type Mg2+-efflux transporter encoded by the <i>unextended</i> (<i>uex</i>) gene. UEX contains a putative cyclic nucleotide-binding homology domain and its mutation separates a vital role for <i>uex</i> from a function in memory. Moreover, UEX localization in mushroom body Kenyon Cells is altered in memory defective flies harboring mutations in cAMP-related genes. Functional imaging suggests that UEX-dependent efflux is required for slow rhythmic maintenance of Kenyon Cell Mg2+. We propose that regulated neuronal Mg2+ efflux is critical for normal and Mg2+ enhanced memory.</p>

Data from: Maze learning and memory in a decapod crustacean

Spatial learning is an ecologically important trait well studied in vertebrates and a few invertebrates yet poorly understood in crustaceans. We investigated the ability of European shore crabs, Carcinus maenas, to learn a complex maze over four consecutive weeks using food as a motivator. Crabs showed steady improvement during this conditioning period in both the time taken to find the food and in the number of wrong turns taken whilst doing so. Crabs also clearly remembered the maze as when placed back in the maze two weeks later and without any food, they all returned to the end of the maze in under eight minutes. Crabs that had not been conditioned to the maze (naïve animals) took far longer to reach the end and many (42 %) did not venture to the end of the maze at all during the one-hour study period. This study provides an initial description of spatial learning in a benthic decapod; a better appreciation of this adaptive trait in these animals will develop our understanding of resource exploitation by benthic crustaceans and their ecological roles.

<p class="MsoNormal"><strong><span>Background</span></strong></p> <p class="MsoNormal"><span>Aging population is rapidly expanding worldwide, and age-related cognitive impairments proves detrimental for achieving a better productive and quality of life.
