Images of mouse VTA and SNc sections labeled for neurotransmitter phenotype markers via RNAscope
<p>48 coronal sections from 3 male and 3 female C57Bl/6J mice were labeled for mRNAs encoding the canonical vesicular transporters for dopamine (VMAT2), GABA (VGAT), and glutamate (VGLUT2). These sections were then imaged via confocal microscopy and saved as CZI files, editable with Zeiss Zen software. These image files include 'event marker' graphics, indicating where neurons positive for one or more of the above transporters are. </p> <p>The protocols used for sample preparation, labeling, imaging, and counting are linked in the Related Works section. </p> <p>Cell count data are organized by subregion (SUBREGION counts for R.csv), anterior-posterior (e.g. VGATVTA.csv), or anterior-posterior and medial-lateral distribution (e.g. 'snc triple.csv' or 'snc vmat2 AP and ML.csv').</p> <p>The R code provided uses these csv files to visualize expression patterns across VTA and SNc. </p>
Linking the microarchitecture of neurotransmitter systems to large-scale MEG resting state networks
<p>Information processing and communication in neuronal circuits is enabled by dynamic networks of inter-areal coupling of neuronal oscillations in which hubs play a central role for regulation of communication. Oscillations are shaped by interactions between pyramidal cells and interneurons and are locally influenced by neuromodulatory systems. Here, we set out to investigate how sparial variability in neurotransmitter receptor and transporter density influences frequency-specific large-scale networks of phase-synchrony (PS) and amplitude-correlation (AC) in human magnetoencephalography data. We found that node centrality - indexing which individual brain regions function as hubs - covaried positively with GABA, NMDA, dopaminergic, and most serotonergic receptor and transporter densities in lower frequency bands (delta to low-alpha for PS, and delta for AC) and in the gamma band, but negatively in between. These results establish how local microarchitecture influences large-scale connectivity networks of neuronal oscillations in the human brain in frequency- and spatially-specific patterns.</p>

Genetically encoded fluorescent glutamate indicators (iGluSnFRs) enable neurotransmitter release and diffusion to be visualized in intact tissue. Synaptic iGluSnFR signal time courses vary widely depending on experimental conditions, often lasting 10-100 times longer than the extracellular lifetime of synaptically released glutamate estimated with uptake measurements. iGluSnFR signals typically also decay much more slowly than the unbinding kinetics of the indicator. To resolve these discrepancies, here we have modeled synaptic glutamate diffusion, uptake and iGluSnFR activation to identify factors influencing iGluSnFR signal waveforms. Simulations suggested that iGluSnFR competes with transporters to bind synaptically released glutamate, delaying glutamate uptake. Accordingly, synaptic transporter currents recorded from iGluSnFR-expressing astrocytes in mouse cortex were slower than those in control astrocytes. Simulations also suggested that iGluSnFR reduces free glutamate levels in extrasynaptic spaces, likely limiting extrasynaptic receptor activation. iGluSnFR and lower-affinity variants nonetheless provide linear indications of vesicle release, underscoring their value for optical quantal analysis.
Data from: Neurotransmitter content heterogeneity within an interneuron class shapes inhibitory transmission at a central synapse
<p>Neurotransmitter content is deemed the most basic defining criterion for neuronal classes, contrasting with the intercellular heterogeneity of many other molecular and functional features. Here we show, in the adult mouse brain, that neurotransmitter content variegation within a neuronal class is a component of its functional heterogeneity. Most Golgi cells (GoCs), the well-defined class of cerebellar interneurons inhibiting granule cells (GrCs), contain cytosolic glycine, accumulated by the neuronal transporter GlyT2, and GABA in various proportions. To assess the functional consequence of this neurotransmitter variation, we paired GrCs recordings with optogenetic stimulations of single GoCs, which preserve the intracellular transmitter mixture. We show that the strength and decay kinetics of GrCs IPSCs, which are entirely mediated by GABA<sub>A</sub> receptors are negatively correlated to the presynaptic expression of GlyT2 by GoCs. We isolate a slow spillover component of GrCs inhibition that is also affected by the expression of GlyT2, leading to a 56 % decrease in relative charge. Acute manipulations of cytosolic GABA and glycine supply recapitulate the modulation of IPSC charge, supporting the hypothesis that presynaptic loading of glycine negatively impact the GABAergic transmission in mixed interneurons through a competition for vesicular filling. Our results suggest that heterogeneity of neurotransmitter supply within the GoC class may provide a presynaptic mechanism to tune the gain of the stereotypic granular layer microcircuit, thereby expanding the realm of possible dynamic behavior.</p>
