MRI raw data for: A novel phantom with dia- and paramagnetic substructure for quantitative susceptibility mapping and relaxometry
<p>MRI raw data from three different magnetic field strength (1.5 T, 3 T, 7T; 7T data are in separate datasets) for the publication 'A novel phantom with dia- and paramagnetic substructure for quantitative susceptibility mapping and relaxometry', in which a phantom was presented that allows for an experimental evaluation of QSM reconstruction algorithms. The phantom contains susceptibility producing particles with dia- and paramagnetic properties embedded in an MRI visible medium (gelatin and agarose gel) and is suitable to assess the performance of algorithms that attempt to separate isotropic dia- and paramagnetic susceptibility at the sub-voxel level. The dataset additionally contains raw data for a phantom that only contains diamagnetic and paramagnetic particles, respectively, for magnetic field strengths of 1.5 T and 3 T (additional 7 T data are provided in separate datasets).</p>

Paramagnetic tailored experiments for the NMR investigation of reduced and oxidized [2Fe-2S]-mitoNEET
<p>We report here the complete data sets and the pulse programs for the NMR spectra of the human iron sulfur binding protein mitoNEET, in its oxidized and reduced states. These data have been used in the publication “The long-standing relationship between Paramagnetic NMR and Iron-Sulfur proteins: the mitoNEET example. An old method for new stories or the other way around?”. This folder includes: paramagnetic 1D proton NOE experiments, 15N-IR-HSQC-AP experiments and CON 13C direct detection experiments in their diamagnetic and paramagnetic version. All these data have been used to identify NMR signals of residues surrounding the metal cofactor. Along with the raw data we report also the pulse programs of the paramagnetic tailored experiments. The NMR spectra of both oxidation states of mitoNEET are significantly different from those reported for previously investigated [Fe2S2]2+/+ proteins and provide a detailed description of the unique electronic properties of mitoNEET, that is essential for the understanding of its the biological function.</p>
Distinct association of HRAS and KRAS with Mn(II) ion illustrated by paramagnetic NMR
<p><a name="OLE_LINK4"></a><span>RAS proteins are among crucial oncogenic proteins and are involved in several essential intracellular processes. The RAS protein has an intrinsic metal binding site for Mg2+, which is important for the conformational stability of the active site. Recently, it was reported that a second metal ion binding site, located further from the active site in HRAS, binds Ca2+ with millimolar affinity.</span><span> As one of the most abundant metal ions in cells, Mn2+ is a potential candidate for the second metal ion binding site in RAS proteins. Here, we examined the interaction of Mn2+ with HRAS and KRAS using high resolution NMR spectroscopy. The NMR data showed that both the second metal ion binding site and the switch I and II regions bind Mn2+ in the RAS proteins. Furthermore, our paramagnetic NMR results disclosed the conformational differences in helix </span><span>a</span><span>3 and the following loop between HRAS and KRAS, accompanied by the association with metal ion binding. These results provide new insights into the interaction of RAS proteins and Mn2+ in the respective biological processes in cells.</span></p>
