Effects of high dietary zinc supplementation timing on the biological responses of gestating sows and their piglets

<p class="MsoNormal">High dietary zinc (Zn) fed to gestating sows may have utility as a fetal imprinting strategy to decrease pre-weaning mortality of piglets. However, the biological action that Zn may work through is unknown. High Zn may modulate the microbiome of the sow and the microbial seeding of the offspring's gut microbiome. Additonally, high dietary Zn and piglet birth weight may alter gene expression in piglet whole blood. Sows (n = 267) were fed 1 of 3 dietary treatments: 1) Control: a corn-soybean meal-based diet containing 125 ppm total supplemental zinc, 2) Breed-to-Farrow: as Control + 141 ppm supplemental Zn as ZnSO<sub>4 </sub>fed from 5 days post-breeding to farrowing; and 3) Day 110-to-Farrow: as Control + 2,715 ppm supplemental Zn as ZnSO<sub>4 </sub>starting on day 110 of gestation until farrowing. A subset of third parity sows (n = 30) were selected to assess the microbiome of colostrum, milk, and rectal and vaginal surfaces of sows. At farrowing, 4 pigs per litter (n = 120) were selected based on birthweight (BiW), as 2 average BiW pigs and 2 pigs with BiW below the litter average were selected for assessing the piglet gut microbiome on the day of birth (day 0) and day 5 of age. 16S rRNA sequencing were implemented for milk and colostrum samples while all other sample types were sequenced using shotgun metagenomics to determine taxonomic and functional profiles. On a different subset of pigs, whole blood was collected from 9 LBW pigs per treatment and 8 ABW Control pigs for RNA-sequencing to evaluate differentially expressed genes (DEGs) and pathways. Only 2 to 3 genes were differentially expressed between Control LBW and LBW pigs born to sows fed high Zn. However, 262 DEGs were identified when comparing LBW and ABW pigs, mostly reflecting pathways associated with translation, ribosome biogenesis, and amino acid and protein synthesis. Measures of alpha diversity (richness and Shannon's H Index) and beta diversity (Bray-Curtis, PERMANOVA) were conducted along with indicator species analyses. Species with an indicator value of &gt; 0.50 were confirmed with a generalized linear mixed model as each <em>P</em>-value was corrected for false discovery rate (FDR) to generate a Q-value. For piglet samples, the MaAsLin2 R package was used to determine multivariate associations between dietary treatment and piglet BiW. High dietary concentrations of Zn fed to gestating sows did not affect the colostrum, milk, or vaginal microbial diversity or populations of sows. Pathogenic bacteria such as <em>Shigella flexneri </em>and <em>Salmonella enterica</em> were less abundant in fecal samples from Breed-to-Farrow sows compared to Control sows. For piglets born to Breed-to-Farrow sows, their gut microbiome favored fiber fermenting, short chain fatty acid generating microbial species compared to Control pigs. Day 110-to-Farrow piglets demonstrated a lower abundance of SCFA producing bacteria compared to Control piglets. Gene families and pathways playing roles in central metabolic functions (starch, pyruvate, sucrose, amino acid metabolism) were more abundant in Breed-to-Farrow piglets compared to pigs born to Control sows. In conclusion, high Zn fed to gestating sows may influence SCFA-producing species and may reduce the abundance of potential pathogenic bacteria in the sow and piglet. Piglet birth weight may have greater effects on gene expression of neonatal pigs. </p>

Microbiota of piglets with three different feed additives via maternal or neonatal administration route

<p>Feed additives aiming to improve gastrointestinal health are frequently supplied to piglets after weaning, but might be more effective when administered before weaning. In this period, feed additives can either be administered directly to neonates, or indirectly via sows feed. It is yet unknown what the effect of the administration route is on gut functionality and health. Therefore, we compared the effect of different dietary interventions on gut functionality after maternal administration (lactation feed) to the neonatal administration route (oral gavage). These feed interventions included medium chain fatty acids (MCFA), beta-glucans (BG), and galacto-oligosaccharides (GOS). We measured intestinal gene expression and microbiota composition after birth (d1) and after weaning (d31). Our results show that the type of intervention and the administration route influence gut functionality (microbiome and gene expression profiles). MCFA administration led to a more differentially orchestrated response when comparing the neonatal and maternal administration route then the other two additives, indicating the route of administration of the feed interventions is determinative for the outcome.. This implies that for each nutritional intervention in early life of a pig the optimal route of administration needs to be determined.</p>

Early life growth and telomere length in wild boar piglets 2018

<p><span>Life history theory predicts a trade-off between growth rates and lifespan, which is reflected by telomere length, a biomarker of somatic state. We investigated the correlation between telomere</span><span> length and early life growth of wild boar piglets, <i>Sus scrofa, </i>kept under semi-natural conditions with</span><span> high food availability to examine our hypothesis that increased pre-and post-natal growth will lead to</span><span> telomere length attrition but that a high supply of nutrient may provide the possibility to compensate telomere loss via telomere repair mechanisms. As predicted, our data showed a clear negative correlation between birth body mass and initial telomere length: heavier neonates had shorter telomeres at birth, and we did not find an influence of the mother on initial telomere length. Body mass at birth correlated with body mass later in life and post-natal growth rate did not affect telomere length. We observed an increase in telomere length during postnatal development, suggesting that high food availability allowed piglets to invest into both, growth and telomere restoration. The increase in telomere length over the duration of the study was not accompanied by telomerase activity, thus telomere elongation was either caused by alternative mechanisms or by short pulses of telomerase activity, that we missed. Taken together, this study suggests a trade-off between investment into growth and telomere maintenance even before birth and the possibility to compensate telomere attrition during growth under high amounts of available energy.</span></p>

Effect of dietary Chlorella vulgaris and carbohydrate-active enzymes on the general health, immune status, antioxidant capacity, and liver lipids and metabolites of weaned piglets

<p>It was evaluated&nbsp;the impact of dietary inclusion of&nbsp;<em>Chlorella vulgaris&nbsp;</em>(CH) and carbohydrases on the general health, immune and oxidative status, and hepatic lipids and metabolites of piglets. Forty-four male weaned piglets were allocated into four diets: control (<em>n</em>=11), CH (control diet with 5% CH,&nbsp;<em>n</em>=10), CH+R (control diet with 5% CH plus 0.005% Rovabio<sup>&reg;</sup>Excel AP,&nbsp;<em>n</em>=10), and CH+M (control diet with 5% CH plus 0.01% of a pre-selected four-CAZyme mixture,&nbsp;<em>n</em>=11) for 15 days. Total cholesterol, LDL-cholesterol and VLDL-cholesterol were increased by CH.&nbsp;In piglets fed CH<em>-</em>based diets, the interplay observed between IgG increase and IgM decrease contributed for piglets&rsquo; survival during weaning.&nbsp;<em>n</em>-6 PUFA were lower in piglets fed CH with or without feed enzymes and the inverse occurred for&nbsp;<em>n</em>-3 PUFA, thus benefiting&nbsp;<em>n</em>-6/<em>n</em>-3 ratio in the liver. The discriminant analysis applied to hepatic variables revealed a good separation between control and CH-based diets but failed to discriminate feed enzymes addition. Minor variations were promoted by Rovabio or the pre-selected enzymatic mixture, thus suggesting some improvement on microalga nutrients digestibility. Taken together, our findings indicate a health promoting effect of CH as feed ingredient in piglets&rsquo; nutrition during the weaning period, without negatively impacting on animals&rsquo; performance.</p> <p>&nbsp;</p>

Porcine reproductive and respiratory syndrome virus infects the reproductive system of male piglets and impairs development of the blood–testis barrier

<p>Porcine reproductive and respiratory syndrome virus (PRRSV) causes a highly contagious disease that threatens the global swine industry. Recent studies have focused on the damage that PRRSV causes to the reproductive system of male pigs, although <span>pathological research is lacking</span>. Therefore, <span>we </span><span>examined the pathogenic mechanisms in male piglets infected with PRRSV</span>.&nbsp;Gross and histopathological changes indicated that PRRSV affected the entire reproductive system, as confirmed via immunohistochemical analysis. PRRSV infected Sertoli cells and spermatogonia.&nbsp;To test the <span>new </span>hypothesis that PRRSV infection in piglets impairs blood&ndash;testis barrier (BTB) development, we investigated the <span>pathology</span>&nbsp;of PRRSV damage in the BTB.&nbsp;PRRSV infection significantly decreased the quantity and proliferative capacity of Sertoli cells constituting the BTB. Zonula occludens-1 and &beta;-catenin were downregulated in cell&ndash;cell junctions. Transcriptome analysis revealed that several crucial genes and signaling pathways involved in the growth and development of Leydig cells, Sertoli cells, and tight junctions in <span>the </span>testes were downregulated. <span>Apoptosis, necroptosis</span>, inflammatory, and oxidative stress-related pathways were activated, whereas hormone secretion-related pathways were inhibited. Many Sertoli cells and spermatogonia underwent apoptosis during early&nbsp;differentiation. Infected piglets exhibited disrupted androgen secretion, leading to significantly reduced testosterone and anti-M&uuml;llerian hormone levels. A cytokine storm occurred, notably upregulating cytokines such as tumor necrosis factor-&alpha; and interleukin-6. Markers of oxidative-stress damage (i.e., H<sub>2</sub>O<sub>2</sub>, malondialdehyde, and glutathione) <span>were upregulated</span>, whereas antioxidant-enzyme activities (i.e., superoxide dismutase, total antioxidant capacity, and catalase) <span>were downregulated</span>. Our results demonstrated that PRRSV infect<span>ed</span>&nbsp;multiple organs in the male reproductive system, <span>which imparied</span> growth in the BTB.&nbsp;</p>

Metabolic effects of a high level of dietary zinc in piglet diets to induce immune competence

<p>There is increasing consumer pressure to reduce the use of therapeutic antibiotics in animal production. Solutions to this challenge require a multi-targeted approach, including use of particular functional ingredients, such as pre- and probiotics that are not only used to boost the growth and productivity but also to influence the development of immune competence and support responsiveness of the immune system at later age. In this context there is a large interest in the development and evaluation of nutritional interventions during early life stages in farm animal species. However, apart from effects on animal growth and productivity, there is lack of knowledge in livestock on ways to modify development of immune competence by nutritional manipulation. Thus an associated project is being conducted with the objective to evaluate effects of a &ldquo;model intervention&rdquo; in piglets to modify development of immune competence. The associated project investigates the effects of provision of diet with a high concentration of zinc (2790 mg/kg) over different time periods in post-weaned piglets as a &ldquo;model intervention&rdquo; for manipulation of immune competence, compared to a reference treatment, in which the animals are fed a diet with a basal level of zinc.</p> <p>The objective of the associated project is to investigate the effects of high concentration of zinc on functional and metabolic changes, including processes related to digestive functions of the gut, its integrity and barrier function, and on immunological processes as effected by the nutritional manipulation. In the associated project we already studied the effects of high concentration of zinc on growth performance, on composition and diversity of the microbiota in the gut, on genome-wide gene expression in intestinal tissue, and on several immune markers in blood. In this proposal we apply for funds to provide us with expertise and access to facilities in order to get a global view of the metabolic changes as induced by the high concentration of zinc by analyzing a range of metabolites in blood. &nbsp;&nbsp;</p>

Data from: An in vivo three-dimensional Magnetic Resonance Imaging-based averaged brain collection of the neonatal piglet (Sus scrofa)

Due to the fact that morphology and perinatal growth of the piglet brain is similar to humans, use of the piglet as a translational animal model for neurodevelopmental studies is increasing. Magnetic resonance imaging (MRI) can be a powerful tool to study neurodevelopment in piglets, but many of the MRI resources have been produced for adult humans. Here, we present an average in vivo MRI-based atlas specific for the 4-week-old piglet. In addition, we have developed probabilistic tissue classification maps. These tools can be used with brain mapping software packages (e.g. SPM and FSL) to aid in voxel-based morphometry and image analysis techniques. The atlas enables efficient study of neurodevelopment in a highly tractable translational animal with brain growth and development similar to humans.

Data from: Consequences for piglet performance of group housing lactating sows at one, two, or three weeks post-farrowing

Housing lactating sows with piglets in a multi-suckling pen from around 14 days post-farrowing is common practice in Swedish organic piglet production. However, nursing-suckling interaction is less frequent in multi-suckling pens than in individual farrowing pens, thus affecting piglet performance, e.g., piglet growth. Moreover, piglet mortality is higher in systems using multi-suckling pens. Three management routines whereby lactating sows with piglets were moved from individual farrowing pens to multi-suckling pens at one, two, or three weeks post-farrowing were compared in terms of nursing-suckling interaction and piglet performance. Correlations between nursing-suckling interaction, piglet performance, and piglet mortality were also examined. In total, 43 Yorkshire sows with piglets were included in the study. Nursing-suckling interaction and all piglet performance parameters except piglet mortality did not differ between management routines. Piglet mortality in the individual farrowing pens did not differ between management routines, but piglet mortality in the multi-suckling pen was lower (P&lt;0.05) when piglets were group housed at three weeks compared with one week post-farrowing. Overall piglet mortality was positively correlated with mortality in the multi-suckling pen for piglets group housed at one week (r = 0.61: P&lt;0.05) and at two weeks post-farrowing (r = 0.62: P&lt;0.05) but not for piglets group housed at three weeks post-farrowing. In conclusion, overall piglet mortality could be reduced if sows and piglets are group housed at three weeks post-farrowing and piglet survival the first week post-farrowing is improved.
