Too hot for the devil? Did climate change cause the mid-Holocene extinction of the Tasmanian devil (Sarcophilus harrisii) from mainland Australia?

<p>The possible role of climate change in late Quaternary animal extinctions is hotly debated, yet few studies have investigated its direct effects on animal physiology to assess whether past climate changes might have had significant impacts on now-extinct species. Here we test whether climate change could have imposed physiological stress on the Tasmanian devil (Sarcophilus harrisii) during the mid-Holocene, when the species went extinct on mainland Australia. Physiological values for the devil were quantified using mechanistic niche models of energy and water requirements for thermoregulation, and soil-moisture-based indices of plant stress from drought to indirectly represent food and water availability. The spatial pervasiveness, extremity, and frequency of physiological stresses were compared between a period of known climatic and presumed demographic stability (8000-6010 BP) and the extinction period (5000-3010 BP). We found no evidence of widespread negative effects of climate on physiological parameters for the devil on the mainland during its extinction window. This leaves cultural and demographic changes in the human population or competition from the dingo (Canis dingo) as the main contending hypotheses to explain mainland loss of the devil in the mid-Holocene.</p>

Data from: Disease swamps molecular signatures of genetic-environmental associations to abiotic factors in Tasmanian devil (Sarcophilus harrisii) populations

Landscape genomics studies focus on identifying candidate genes under selection via spatial variation in abiotic environmental variables, but rarely by biotic factors such as disease. The Tasmanian devil (Sarcophilus harrisii) is found only on the environmentally heterogeneous island of Tasmania and is threatened with extinction by a nearly 100% fatal, transmissible cancer, devil facial tumor disease (DFTD). Devils persist in regions of long-term infection despite epidemiological model predictions of species' extinction, suggesting possible adaptation to DFTD. Here, we test the extent to which spatial variation and genetic diversity are associated with the abiotic environment and/or DFTD. We employ genetic-environment association analyses using a RAD-capture panel including 6,886 SNPs from 3,286 individuals sampled pre- and post-disease arrival. Pre-disease, we find significant correlations of allele frequencies with environmental variables, including 365 unique loci linked to 71 genes, suggesting local adaptation to abiotic environment. The majority of candidate loci detected pre-DFTD were not detected post disease arrival. Several post-DFTD candidate loci were associated with disease prevalence and were in linkage disequilibrium with genes involved in tumor suppression and immune response. Loss of apparent signal of abiotic local adaptation post-disease suggests swamping by the strong selection resulting from the rapid onset of DFTD.

Data from: Pedigree analysis reveals a generational decline in reproductive success of captive Tasmanian devil (Sarcophilus harrisii): implications for captive management of threatened species

Captive breeding programs are an increasingly popular tool to augment the conservation of threatened wild populations. Many programs keep detailed pedigrees, which are used to prescribe breeding targets to meet demographic and genetic goals. Annual breeding targets are based on previous productivity, but do not account for changes in reproductive success that may occur over generations in captivity and which may impair the ability of a program to meet its goals. We utilise a large studbook from the Tasmanian devil (Sarcophilus harrisii) captive breeding program to investigate biological, genetic and environmental factors that affect variation in reproductive success among individuals and over generations of captive breeding. Reproductive success declined with increasing generations in captivity: wild-born females had a 56.5% chance of producing a litter compared to a 2.8% chance for generation 5 captive-born females (N = 182) and when they did, wild-born females produced more offspring (3.1 joeys, 95% CI: 2.76 - 3.38, compared to 2.7 joeys, 95% CI: 2.55 - 2.90, in captive-born females [N = 105]). Reproductive success also declined as dam age at first breeding increased. Our results reveal a conflict with the widely-cited conservation strategy to limit opportunity for selection by extending generation length through delaying reproduction, as captive breeding programs that delay female breeding with this goal in mind risk reduced productivity. Our data demonstrate the benefit of pedigree analysis to identify biological processes that reveal crucial trade-offs with conservation best-practice.

Transmissible cancer influences immune gene expression in an endangered marsupial, the Tasmanian devil (Sarcophilus harrisii)

<p>Understanding wildlife diseases' effects on populations requires insight into local environmental conditions, host defence mechanisms, host life-history trade-offs, pathogen population dynamics, and their interactions. The survival of Tasmanian devils (<em>Sarcophilus harrisii</em>) is challenged by a novel, fitness limiting pathogen, Tasmanian devil facial tumour disease (DFTD), a clonally transmissible, contagious cancer. In order to understand the devils' capacity to respond to DFTD, it is crucial to gain information on factors influencing the devils' immune system. By using RT-qPCR, we investigated how DFTD infection in association with intrinsic (sex and age) and environmental (season) factors influences the expression of 10 immune genes in Tasmanian devil blood. Our study showed that the expression of immune genes (both innate and adaptive) differed across seasons, a pattern that was altered when infected with DFTD. The expression of immunogbulins IgE and IgM:IgG showed downregulation in colder months in DFTD infected animals. We also observed strong positive association between the expression of an innate immune gene, CD16, and DFTD infection. Our results demonstrate that sampling across seasons, age groups and environmental conditions are beneficial when deciphering the complex eco-evolutionary interactions of not only conventional host-parasite systems, but also of host and diseases with high mortality rates, such as transmissible cancers.</p>
