Air temperature data for C1 chart recorder, 1952 - ongoing.

Temperature data were collected on a daily time-scale from the C1 climate station (3018 m) since 1952. Over time various circumstances have led to days with missing values. Some missing values were estimated from redundant sensors and nearby climate stations using various methods. Greenland 1987 was a basis for the methodology. However when it was not possible to use this methodology, new methods were developed.

Hubbard Brook Experimental Forest: 15-Minute Air Temperature Record, 2014 - present

Air temperature is measured at 15-minute intervals in six clearings throughout the Hubbard Brook experimental watersheds and at the Headquarters Station. Beginning in 2014, digital sensors housed in aspirated solar radiation shields collected air temperature measurements. These data are gathered at the Hubbard Brook Experimental Forest in Woodstock, NH, which is operated and maintained by the USDA Forest Service, Northern Research Station.

Hubbard Brook Experimental Forest: Daily Temperature Record, 1955 - ongoing

Air temperature at the Hubbard Brook Experimental Forest (HBEF) is measured at five locations in rain gage clearings throughout the experimental watersheds and at the Headquarters building. The oldest air temperature record dates back to October 20,1955 at Station 1. From 1955 - 2014, temperature measurements were made continuously using hygrothermographs housed in standard shelters. Beginning in 2013, digital sensors housed in solar radiation shields collected daily minimum and maximum temperature measurements. These data are gathered at the Hubbard Brook Experimental Forest in Woodstock, NH, which is operated and maintained by the USDA Forest Service, Northern Research Station.

Infilled daily air temperature data for C1 chart recorder, 1952 - ongoing.

Daily minimum and maximum surface air temperature data have been collected at the C1 climate station (3022 m asl) almost continuously from 1952 to the present. C1 is on locally level terrain on the southeastern flank of Niwot Ridge, 9.7 km east of the Continental Divide. Surrounding vegetation is generally closed-canopy subalpine conifer forest. Temperature minima and maxima were measured with a chart-recording hygrothermograph housed in a Stevenson screen located in an open, sparsely treed area adjacent to the forest proper. Hygrothermograph records were calibrated at the time of chart changes against liquid-in-glass thermometers. Processing included quality checks and infilling of missing daily data (see Methods).

Infilled daily air temperature data for D1 chart recorder, 1952 - ongoing.

Daily minimum and maximum surface air temperature data have been collected at the D1 climate station (3739 m asl) almost continuously from 1952 to the present. D1 is on a narrow, exposed ridge on the westernmost part of Niwot Ridge, 2.6 km east of and ca. 200 m lower in elevation than the Continental Divide. Surrounding vegetation is low-stature alpine tundra. Temperature minima and maxima were measured with a chart-recording hygrothermograph housed in a Stevenson screen. Hygrothermograph records were calibrated at the time of chart changes against liquid-in-glass thermometers. Processing included quality checks and infilling of missing daily data (see Methods).

Air temperature data for Saddle chart recorder, 1981 - 2017.

Air temperature data were collected on a daily time-scale from the Saddle climate station (3525 m) since 1981. Over time, various circumstances have led to days with missing values. Some of these values were estimated from redundant sensors and nearby climate stations. Greenland 1987 was a basis for the methodology; however when it was not possible to use this methodology, new methods were developed.

Twenty four hour continuous tympanic temperature recordings in healthy volunteers and patients presented with Undifferentiated fever

Twenty-four-hour continuous tympanic temperature recordings were obtained using high-accuracy tympanic probes placed at the auditory canal. Measurements were recorded at one-minute intervals over a 24-hour period, yielding 1,440 data points per subject and enabling high-resolution temporal profiling of body temperature.

In Phase I, a total of 100 healthy adult volunteers were recruited to establish baseline circadian thermoregulatory patterns. These recordings were stratified by gender to examine physiological variability among individuals without fever.

In Phase II, 184 adult patients presenting with undifferentiated fever of seven or more days’ duration were enrolled. Based on clinical examination and laboratory confirmation, the temperature profiles were analysed across a range of conditions, including tuberculosis, non-tubercular bacterial infections, dengue fever, malaria, leptospirosis, pyogenic sepsis, thyroiditis, malignancies, and non-infectious inflammatory diseases. This dual-phase dataset facilitated a comparative analysis between normative and pathological thermoregulatory patterns, supporting the development of classification models for diagnostic differentiation.

Hippocampal CA1 pyramidal cell membrane voltage recorded in response to noise stimuli at two temperatures.

Electrophysiological recording of the membrane voltage (whole-cell patch-clamp) of three hippocampal CA1 pyramidal cells. Cells are stimulated with a current step chosen to ensure a firing rate around 5-10Hz (amplitude of the current step is given in the filenames) and a noise stimulus with zero mean (Ornstein-Uhlenbeck process with 4ms timescale). Each CSV file contains three columns, the timepoints (saved at 10000Hz), the noise stimulus, and the voltage trace recorded in response to the given noise stimulus. Voltages are recorded at low temperatures (around 32 degrees Celsius) and at high temperatures (around 37 degrees Celsius for cell 1, around 40 degrees Celsius for cells 2 and 3), exact temperatures are given in the filenames. The file metadata.csv contains additional information.

Ambient temperature CiPA dynamic hERG protocol dataset recorded on the SyncroPatch 384SE

Abstract

The Comprehensive in vitro Proarrhythmic Assay (CiPA) has promoted use of in silico models of drug effects on cardiac repolarization to improve proarrhythmic risk prediction. These models contain a pharmacodynamic component describing drug binding to hERG channels that required in vitro data for kinetics of block, in addition to potency, to constrain them. To date, development and validation has been undertaken using data from manual patch-clamp. The application of this approach at scale requires the development of a high-throughput, automated patch-clamp (APC) implementation. Here, we present a comprehensive analysis of the implementation of the Milnes, or CiPA dynamic protocol, on an APC platform, including quality control and data analysis. Kinetics and potency of block were assessed for bepridil, cisapride, terfenadine and verapamil with data retention/QC pass rate of 21.8% overall, or as high as 50.4% when only appropriate sweep lengths were considered for drugs with faster kinetics. The variability in IC50 and kinetics between manual and APC was comparable to that seen between sites/platforms in previous APC studies of potency. Whilst the experimental success is less than observed in screens of potency alone, it is still significantly greater than manual patch. With the modifications to protocol design, including sweep length, number of repetitions, and leak correction recommended in this study, this protocol can be applied on APC to acquire data comparable to manual patch clamp.

Description

High-throughput patch clamp electrophysiology dataset acquired on Nanion SyncroPatch 384PE platform describing the potency and kinetics of drug block of hERG channels stably expressed in CHO cells. The voltage protocols used were 10 or 40 s 0mV steps with a 15s, -80mV inter-pulse intervals, repeated 5 or 10 times in the absence (control) and presence of drug. Drug was added and cells clamped at -80 mV for 4 minutes prior to recording to ensure drug equilibration prior to recording the onset (kinetics) of drug block. This dataset accompanies the paper titled 'High throughput measurement of hERG drug block kinetics using the CiPA dynamic protocol' by Monique J.
