DataSet of "No renal dysfunction or salt and water retention in acute mountain sickness at 4,559 m among young resting males after passive ascent"
<p><strong>Abstract</strong></p> <p><strong>Purpose</strong>: This study examined the role and function of the kidney at high altitude in relation to fluid balance and the development of acute mountain sickness (AMS), avoiding confounders that have contributed to conflicting results in previous studies.</p> <p><strong>Methods</strong>: We examined 18 healthy male volunteers (18 - 40 years) not acclimatized to high altitude while on a controlled diet and resting recumbently for 24 h at Lausanne (altitude: 560 m) followed by a period of 44 hours after reaching the Regina Margherita hut (4,559 m) by helicopter.</p> <p><strong>Results</strong>: AMS scores peaked after 20 h at 4,559 m. AMS was defined as functional Lake Louise score <span class="math-tex">\({\ge}\)</span> 2.There were no significant differences between 10 subjects with and 8 subjects without AMS for urinary flow, fluid balance and weight change. Sodium excretion rate was lower in those with AMS after 24 h at altitude. Microalbuminuria increased at altitude but not differently between the groups. Creatinine clearance was not affected by altitude or AMS, while sinistrin and PAH clearances decreased slightly, more markedly in those without AMS. Plasma concentrations of epinephrine, norepinephrine, atrial natriuretic factor and vasopressin increased while renin activity, angiotensin and aldosterone decreased at altitude. Hormones levels did not differ between those with and without AMS.</p> <p><strong>Conclusions</strong>: 1) Renal function is not affected by hypoxia at 4,559 m in resting subjects except for minor microalbuminuria, 2) high altitude diuresis does not occur and 3) AMS is not associated with salt and water retention or renal dysfunction.</p>
Statistical analysis and dataset for: Acute exposure to caffeine improves foraging in an invasive ant
<p>Linked to the journal article published in iScience (https://doi.org/10.1016/j.isci.2024.109935).</p> <p><em><strong>Abstract</strong></em></p> <p>Argentine ants, <em>Linepithema humile</em>, are a particularly concerning invasive species. Control efforts often fall short likely due to a lack of sustained bait consumption. Using neuroactives, such as caffeine, to improve ant learning and navigation could increase recruitment and consumption of toxic baits. Here, we exposed <em>L. humile</em> to a range of caffeine concentrations and a complex ecologically relevant task: an open landscape foraging experiment. Without caffeine, we found no effect of consecutive foraging visits on the time the ants take to reach a reward, suggesting a failure to learn the reward’s location. However, under low to intermediate caffeine concentrations ants were 38% faster with each consecutive visit, implying that caffeine boosts learning. Interestingly, such improvements were lost at high doses. In contrast, caffeine had no impact on the ants’ homing behavior. Adding moderate levels of caffeine to baits could improve ant’s ability to learn its location, improving bait efficacy.</p> <p> </p> <ul> <li><strong>sample_videos.zip</strong>: A subset of the videos used for data extraction. KSR inhibitor APS-2-79 sensitivity test in JURKAT and ALL-SIL T-cell acute lymphoblastic leukemia cell lines measured by Cell Counting Kit 8
<p>APS-2-79 compound was purchased from MedChemExpress (Monmoutyh Junction, NJ, USA). The 20 mg/ml stock solution was prepared in DMSO. To calculate the IC50, JURKAT and ALL-SIL cells were cultured for 72h with a range of APS-2-79 concentrations (5-15 µM) added as equal volumes. Cells treated with 0.5% DMSO (vehicle) were used as negative control. Cells treated with 10% DMSO were used as positive control. The viability of cells was measured using Cell Counting Kit 8 (Sigma Aldrich) and GloMax Microplate Reader system (Promega) with 450 nm wavelength and 600 nm as reference wavelength. The relevant reads are made from following wells: 2A-2D (15 µM APS-2-79), 3A-3D (12.5 µM APS-2-79), 4A-4D (10 µM APS-2-79), 5A-5D (7.5 µM APS-2-79), 6A-6D (5 µM APS-2-79), 7A-7D (vehicle), 8A-8D (positive control).</p>

Acute Respiratory Distress Syndrome-Database of Genes (ARDS-DB)
<p>To better understand the gene level associations that are most relevant to Acute Respiratory Distress Syndrome (ARDS), a comprehensive resource is needed. There’s currently no freely available database dedicated to ARDS that provides comprehensive gene lists from experimentally verifiable studies, gene function, gene location, and additional metadata for tracking related link out resources. The need for such a database is only accentuated by the steep rise in ARDS cases due to the 2020 Coronavirus pandemic, in which infected patients admitted to the ICU develop ARDS at a rate of 67% to 85%, calling for an increase into ARDS research. </p> <p>Our goal was to develop such a resource for use by the scientific community to enhance our studies of ARDS and associated genes. Our first step was to perform data mining and curation of scientific literature through a robust review process. Subsequent steps enabled us to refine our data by capturing specific metadata and incorporating these into our database. The version 1 of the database will provide users with access to the database flat file with current genes, gene location, chromosomal information, and more in a freely accessible and downloadable format. Future project goals are to develop a standalone web portal that will integrate the gene level information with network analysis, and other visualizations for users. </p> <p> </p>
Dataset for the paper "Prolonged prothrombin time as an early prognostic indicator of severe acute respiratory distress syndrome in patients with COVID-19 related pneumonia"
<p>The dataset contains the data on ICU-transferred (N=100) and Stable (N=131) patients with COVID-19 (N=156) and Non-COVID-19 viral pneumonia (N=75). Among COVID-19 patients of this study, 82 patients developed Refractory Respiratory Failure (RRF) or Severe Acute Respiratory Distress Syndrome (SARDS) and were transferred to Intensive Care Unit (ICU), 74 patients had a Stable course of disease and were not transferred to ICU. SHOCK Trial - Early Revascularization in Acute Myocardial Infarction Complicated by Cardiogenic Shock
<p>The leading cause of death in patients hospitalized for acute myocardial infarction is cardiogenic shock. We conducted a randomized trial to evaluate early revascularization in patients with cardiogenic shock. In patients with cardiogenic shock, emergency revascularization did not significantly reduce overall mortality at 30 days. However, after six months there was a significant survival benefit. Our conclusion was that early revascularization should be strongly considered for patients with acute myocardial infarction complicated by cardiogenic shock. We are making original data that was collected for this trial available here for further analysis.</p>
Dysglycemias in patients admitted to ICUs with severe acute respiratory syndrome due to COVID-19 versus other causes – A cohort study - Dataset
<p>Dataset of a cohort whose summary is described below.</p> <p>Abstract</p> <p>Importance: Dysglycemias have been associated with worse prognosis in critically ill patients with or without diabetes, but data on their association with severe COVID-19 and outcomes are lacking. Objectives: To analyze the relationship of dysglycemias with COVID-19 in hospitalized patients with severe acute respiratory syndrome (SARS) and assess the influence of dysglycemias on mortality. Design, Setting and Participants: Cohort of consecutive patients with SARS and suspected COVID-19 hospitalized in intensive care units (ICUs) across eight hospitals in Curitiba-Brazil. Main Outcomes and Measures: The primary outcome was the influence of COVID-19 on the variation of the following parameters of dysglycemia: highest glucose level at admission, mean and highest glucose levels during ICU stay, mean glucose variation, and percentage of days with hyperglycemia. The secondary outcome was the influence of COVID-19 and each of the five parameters of dysglycemia on hospital mortality within 30 days from ICU admission. Results: We compared 703 patients with COVID-19 and 138 without COVID-19 admitted to the ICUs due to SARS. Compared with patients without COVID-19, those with COVID-19 had significantly higher glucose peaks at admission (198.1mg/dL vs. 167.8mg/dL, respectively) and during ICU stay (285.9mg/Dl vs. 230.9md/dL), higher mean daily glucose values (167.9mg/dL vs. 149.8mg/dL), higher percentage of days with hyperglycemia during ICU stay (vs. 45.0 vs. 31.5), and greater mean daily glucose variations (85.3mg/dL vs. 63.5mg/dL). However, these associations were lost after adjustment for APACHE II scores, SOFA scores, CRP level, corticosteroid use and nosocomial infection. Dysglycemia and COVID-19 were each independent risk factors for mortality. Conclusions and Relevance: Patients with SARS due to COVID-19 had higher mortality and more frequent dysglycemia than patients with SARS due to other causes. This association seemed to be related to disease severity and inflammation and was independent of corticosteroid use, suggesting no specific relationship with the SARS-CoV-2 infection.</p>

Consensus models to predict oral rat acute toxicity and validation on a dataset coming from the industrial context
<p>We report predictive models of acute oral systemic toxicity representing a follow-up of our previous work in the framework of the NICEATM project. It includes the update of original models through the addition of new data and an external validation of the models using a dataset relevant for the chemical industry context. A regression model for LD50 and classification model for toxicity classes according to the Global Harmonized System categories were prepared. ISIDA descriptors were used to encode molecular structures. Machine learning algorithms included Support Vector Machine (SVM), Random Forest (RF) and Naïve Bayesian. Selected individual models were combined in consensus.</p> <p>The different datasets were compared using the Generative Topographic Mapping approach. It appeared that the NICEATM datasets were lacking some relevant chemotypes for chemical industry. The new models trained on enlarged data sets have applicability domain (AD) sufficiently large to accommodate industrial compounds. The fraction of compounds inside the models’ AD increased from 58 % (NICEATM model) to 94 % (new model). Yet, the increase of training sets only slightly improved of the models’ prediction performance: RMSE values decreased from 0.56 to 0.47 and balanced accuracies increased from 0.69 to 0.71 for NICEATM and new models, respectively.</p>
Characterization of Metabolism Associated with Outcomes in Severe Acute Pancreatitis: Insights from Serum Metabolomic Analysis
<p>1H NMR spectra data of SAP patients (Survivors/ Non-survivors). The spectra were binned as 0.02 ppm spectral buckets. The chemical shift regions corresponding to the water region and TSP were excluded to avoid spectral interference. This dataset was used for the metabolomics related study to highlight the dysregulation of metabolites in the study group.</p> <p> </p>
Identification of Genes Regulating Dexamethasone Resistance and Prognostic Model Development in Acute Lymphoblastic Leukemia
<p>This study investigates the mechanisms of dexamethasone resistance in acute lymphoblastic leukemia (ALL) and presents a prognostic model to predict patient outcomes and immunotherapy responses. By analyzing gene expression data, we identified autophagy-related genes associated with dexamethasone resistance, particularly focusing on STK38L’s role in modulating autophagy via ULK1. Our results reveal that high STK38L expression enhances dexamethasone resistance by promoting autophagy markers LC3II/LC3I and beclin-1. This study provides valuable insights into the molecular basis of dexamethasone resistance and highlights STK38L as a potential biomarker and therapeutic target for improving ALL treatment strategies.</p>
