Lab disease outcomes data evaluating how antibiotic tolerant vs. non-tolerant cell-free supernatant from Pseudomonas aeruginosa affects the interaction between a fungal pathogen (Batrachochytrium dendrobatidis) and amphibian (Rana sylvaticus), 2022.

Microbes living on hosts and in the environment can play a key role in helping hosts to combat pathogens. However, antibiotic-induced alterations to microbial metabolite production could disrupt this dynamic. Here, we investigated whether antibiotic tolerance influences the anti-pathogenic properties of host-associated (living on the host; biofilms) and environmental (living in the soil of water column; planktonic) microbes in vitro and in vivo. For our model host and pathogen, we used the amphibian (Rana sylvatica)-Batrachochytrium dendrobatidis (Bd) system. For our model host-associated (biofilm) and environmental (planktonic) microbes, we used four strains of Pseudomonas aeruginosa that vary in their tolerance to antibiotics and their biofilm-forming capabilities: Planktonic, non-antibiotic tolerant (ΔsagS/VC); Planktonic, antibiotic tolerant (ΔsagS::sagS_L154A); Biofilm, non-antibiotic tolerant (ΔsagS::sagS_D105A); Biofilm, antibiotic tolerant (ΔsagS::sagS). We collected cell-free supernatants (CFS) from each strain to examine the effects of metabolites. We conducted four experiments. In our pathogen-only exposures to test direct effects of metabolites on Bd, we exposed Bd zoospores to each P. aeruginosa CFS at six concentrations. After 11 days of growth, we measured relative abundance of Bd across each treatment. In our host-only exposures to test effects of metabolites on host disease outcomes, we placed R. sylvatica tadpoles in individual units containing each P. aeruginosa CFS. After 48 hours, water was changed into clean well water (no CFS). Bd zoospores were immediately added to each experimental unit following the water change. After 5 days of Bd exposure, we measured snout-vent length (SVL), mass, developmental stage, and Bd quantification in the mouthparts using qPCR for each tadpole. Outdoor mesocosm study evaluating how mass, NaCl tolerance, and pesticide tolerance affect oxidative stress biomarkers (CAT, SOD, GR, GPx, TBARS) in larval wood frogs (Rana sylvatica) exposed to baseline and NaCl-contaminated conditions, 2019

Biomarkers of oxidative stress can aid in wildlife monitoring by allowing conservationists to detect sublethal environmental shifts. However, interpretation of stress responses can be complicated by multiple interacting factors (e.g., individual development, evolved physiological tolerance to stressors) which alter biomarker expression. Here, we investigated how individual ontogenetic traits and population-level tolerance traits influence oxidative stress responses under baseline and contaminated environmental conditions. For our model contaminant, we used NaCl (common freshwater contaminant due to factors such as coastal flooding, irrigation, airborne salt circulation, drought, runoff from road deicing salts). For our model wildlife populations, we used larval wood frogs (Rana sylvatica) from six noninteracting populations known to vary in two population-level tolerance traits: NaCl tolerance (calculated as average time to death from lethal NaCl exposure) and pesticide tolerance (determined by proxy of distance to agriculture - a consistent and highly repeatable relationship). At an outdoor research facility, R. sylvatica tadpoles were exposed to either baseline conditions (0 g/L NaCl added) or NaCl-contaminated conditions (1 g/L NaCl added for 21 days, then reduced to 0.5 g/L NaCl). Exposures were conducted in individual units with 40 replicates per population for each treatment. The experiment was terminated per individual to capture the full term of larval development (Developmental stage: Gosner stage 36), lasting between 33-48 days. For each individual, we measured mass, Snout-Vent-Length, and developmental stage before processing for biomarker expression. Although there is evidence supporting suboptimal decision-making in lupus and its association with glucocorticoids consumption, it is not clear what variables impact such decisions. The aim of this study is to explore how social, clinical, psychological, and demographic factors impact social and temporal decision-making in people with lupus. Through a within-subjects experimental-design, our participants responded to social, clinical, psychological, and demographic electronic questionnaires. Then, they participated in two behavioral economics experiments: the third-party dictator game, and the delay discounting task. Our results show that hostility, and age are essential predictors of social decisions, whereas obsessive-compulsiveness and anxiety better predict temporal decisions. These variables behave as expected, but anxiety shows unexpected results: most anxious people act patiently and prefer delayed but bigger rewards. Finally, clinical factors are critical decision predictors for social and temporal decisions. When people are in remission, they tend to impose higher punishment on those who violate the social norm, and they also tend to prefer immediate rewards. When taking glucocorticoids, they also prefer immediate rewards, and as the dosage of glucocorticoids intake increases, they tend to impose higher punishment on norm violators. Clinicians, researchers, and practitioners must consider the side effects of glucocorticoids on decision-making.</span></p>

The Biomass and Plant Functional Traits of Leymus chinensis Affected by Genotypic Diversity and Soil Nitrogen Addition through a Two-year Experiment, Tianjin, China, 2021-2023

In order to investigate the effects of soil nitrogen addition on the genotypic diversity of Leymus chinensis, 12 genotypes of Leymus chinensis were used as plant material and a two-factor experimental design was carried out in this study. Factor one was genotypic diversity of L. chinensis, including three levels: mono-genotype (G1), three genotypes (G3), and six genotypes (G6). Factor two was the soil nitrogen addition level, which included four levels: no nitrogen addition (N0), 2.5 g N/(m²·a) nitrogen application (N2.5), 5 g N/(m²·a) nitrogen application (N5), and 10 g N/(m²·a) nitrogen application (N10). Each treatment had 12 combinations as replicates, and 12 genotypes of L. chinensis were used. The frequency of each genotype was standardized across all treatment levels of genotypic diversity × soil nitrogen addition. The experiment commenced in September 2021 and soil nitrogen was applied every 2 months. Plants were cultivated in the experimental field at Nankai University, but were moved to a greenhouse for overwintering from November to February each year. During the experiment, there were no stresses or disturbances such as shading, drought, or insect feeding; weeds were regularly removed.

Using the Tea Bag Index to unravel how interactions between an antibiotic (Trimethoprim) and endocrine disruptor (17a-estradiol) affect aquatic microbial activity: Supporting Dataset 1

<p>The constant release of complex mixture of pharmaceuticals, including antimicrobials and endocrine disruptors, into the aquatic environment. These have the potential to affect aquatic microbial metabolism and alter biogeochemical cycling of carbon and nutrients. We used&nbsp;the Tea Bag Index (TBI) for decomposition within a series of contaminant exposure experiments to test how interactions between an antibiotic (trimethoprim) and endocrine disruptor (17a-estradiol) affects microbial activity in an aquatic system. The TBI is a citizen science tool used to test microbial activity by measuring the differential degradation of green and rooibos tea as proxies for labile and recalcitrant organic matter decomposition. Here we present the raw data on pharmaceutical exposures and the mass loss of the Rooibos and Green tea bags within the experiment. From Tea Bag mass loss we then calculated the Stabilisation Factor (S) and Initial Decomposition Rate of the labile organic matter fraction.</p>

Sex affects transcriptional associations with schizophrenia across the dorsolateral prefrontal cortex, hippocampus, and caudate nucleus

<p>This is supplementary data and source data for the manuscript,&nbsp;<em>"Sex affects transcriptional associations with schizophrenia across the dorsolateral prefrontal cortex, hippocampus, and caudate nucleus"</em>.</p> <p><strong>Abstract</strong>: Schizophrenia is a complex neuropsychiatric disorder with sexually dimorphic features, including differential symptomatology, drug responsiveness, and male incidence rate. Prior large-scale transcriptome analyses for sex differences in schizophrenia have focused on the prefrontal cortex. Analyzing BrainSeq Consortium data (caudate nucleus: n=399, dorsolateral prefrontal cortex: n=377, and hippocampus: n=394), we identified 831 unique genes that exhibit sex differences across brain regions, enriched for immune-related pathways. We observed X-chromosome dosage reduction in the hippocampus of male individuals with schizophrenia. Our sex interaction model revealed 148 junctions dysregulated in a sex-specific manner in schizophrenia. Sex-specific schizophrenia analysis identified dozens of differentially expressed genes, notably enriched in immune-related pathways. Finally, our sex-interacting expression quantitative trait loci analysis revealed 704 unique genes, nine associated with schizophrenia risk. These findings emphasize the importance of sex-informed analysis of sexually dimorphic traits, inform personalized therapeutic strategies in schizophrenia, and highlight the need for increased female samples for schizophrenia analyses.</p>

<p><span>This dataset is related to the operation of the second KIOS CoE sandboxing use case (SUC2) which inclused 3 scenarios (S1-S3) which examins the behavious a WAP scheme of power grids in case of a short circuit fault and in case of two types of cyber attacks. The description of the architecture of the University of Cyprus/ KIOS CoE sandboxing environmnet used for extracting these datasets along with the full list of scenarios and their detailed implementation are described in the supporting documents.</span></p> <p><span>Brief description of each of the 3 scenarios of this SUC2 are provided below.</span></p> <p><span>The datasets for the first scenario (S1) of SUC2</span><span> examines the operation of a wide area protection scheme in a transmission line which receives data sent from PMUs at the two ends of the lines, when a short-circuit fault occurred in the range of the transmission line between buses 7 and 8 of the system. More details about the scenario SUC2/S1 related to this scenario's dataset can be found in Section&nbsp;</span><span>1.3.1</span><span> of the SUC2 supporting document. </span><span><span>The dataset includes electrical measurements of the current flow in line 7-8 (of the IEEE 9-bus system), in both magnitude and sinusoidal form</span><span>.</span><span> The dataset is provided in the form of time-series measurements available as MATLAB (.mat) and CSV files, which were recorded with a 30-second and 40-second time resolution, respectively.
